In depth
The transcription factor that commands the body's own antioxidant and detoxification arsenal.
What drives it
Oxidative stress increases it.
Mitochondrial ROS activate Nrf2 (review, PMC7072240): rising ROS modify Keap1 cysteine residues, which releases Nrf2 from degradation so it translocates and switches on the antioxidant/are program. With the existing the Nrf2 ⊣ Oxidative Stress (ROS) link this closes the hormetic feedback loop — mild oxidative stress from caloric restriction or exercise (mitohormesis) raises the very defense that then lowers ROS. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: ROS oxidize Keap1 sensor cysteines (Cys-151, -273, -288 in mouse), releasing Nrf2 from the Keap1/cullin-3 E3-ubiquitin ligase complex and preventing its degradation, so Nrf2 accumulates and translocates to the nucleus to drive are genes. Not the only route: a non-canonical p62-dependent pathway activates Nrf2 independently of Keap1 oxidation, and growth factors can activate Nrf2 via PI3K/GSK3-beta. Verbatim: “Reactive mouse Keap1 cysteines (Cys-151, -273, and -288) are redox sensors, and upon oxidation by ROS, results in the dissociation of Nrf2 from Keap1/Cul3 which allows Nrf2 stabilization and translocation into the nucleus”. doi 10.1016/j.cellsig.2012.01.008 · PMID 22286106 · PMC7072240
NRF2 restraint reduces it.
Itoh 1999, Genes Dev 13:76 — Keap1 represses Nrf2 via the Neh2 domain; oxidants inactivate Keap1, releasing Nrf2 to the ARE. The limit: the primary (Itoh 1999, Genes Dev 13:76) was not opened; this rests on secondary confirmation only, and remains at ladder T2 rather than T3.
Adaptive stress response increases it.
NRF2 induction by plant xenobiotics is xenohormesis - a mild chemical stress producing an antioxidant response. Carried by the hormesis, inflammaging and thermogenesis reviews. doi 10.1002/med.21743

The entire NRF2 enzymatic output depends on NADPH supply. Carried by the intermediary-metabolism and clinical reviews. doi 10.1016/j.molmet.2023.101755 · PMC9967593
Mitochondrial oxidant leak increases it.
Mitochondrial ROS act as a signal that activates Nrf2-dependent antioxidant transcription — the hormetic arm. doi 10.1016/j.cell.2022.11.001 · PMID 23746838 · PMC7139312
Ethanol-metabolizing enzyme reduces it.
Verbatim (PMC6316574): CYP2E1 'produces via CYP 2E1 many reactive oxygen species (ROS) such as ethoxy radical, hydroxyethyl radical, acetyl radical, singlet radical, superoxide radical, hydrogen peroxide, hydroxyl radical, alkoxyl radical, and peroxyl radical.' And PMC12086950: 'Increased CYP2E1 activity results in oxidative stress due to generation of reactive oxygen species (ROS)... In addition, alcohol metabolism leads to glutathione depletion, mitochondrial damage, lipid peroxidation.' The link runs inversely: the oxidant load outruns the antioxidant response - glutathione is depleted, not raised. cyp2e1 is Liver Regenerator's sole-carrier lane; this edge describes node biology and claims no lane. What it drives
It increases Master antioxidant.
Nrf2/GCLc study 2013, Hepatol Mon — Nrf2 binds the antioxidant-response element in the glutamate-cysteine ligase (GCLc, rate-limiting for GSH) promoter, inducing glutathione synthesis. PMID 23967023. Sources: Redox Biol 2018 doi 10.1016/j.redox.2018.06.006; PMC3166574; PMC6259123; PMC3677633. NRF2 acts by binding the antioxidant response element as a heterodimer with small MAF proteins — it raises capacity to make glutathione, it does not itself supply cysteine, so substrate availability remains limiting. The caveat that must travel with every NRF2 claim: NRF2 activation is not uniformly good. Tumors carry activating NRF2 mutations, and NRF2 cancer target genes are functionally linked to metabolic, proliferative and chemoresistance advantages; glutathione and thioredoxin are described as essential for cancer initiation and progression (Redox Biol 2018, doi 10.1016/j.redox.2018.06.006). In SDHB-deficient tumor cells, NRF2-guided glutathione synthesis supports survival and proliferation, and NRF2 blockade caused severe cytotoxicity — the NRF2 inhibitor brusatol is being developed as a therapy. doi 10.1016/j.redox.2018.06.006 · PMID 23967023 · PMC3166574
It reduces Follicle oxidative stress.
NRF2 is the master redox regulator expressed in the hair follicle and counters follicular oxidative stress. In detail: NRF2 is a CNC-bZIP transcription factor regulating the basal and inducible expression of a wide array of antioxidant genes; on dissociation from KEAP1 it accumulates in the nucleus and transactivates the antioxidant response element of NQO1, HMOX1, glutamate-cysteine ligase and the glutathione-S-transferases (PMC6259123; PMC3677633). Loss-of-function is decisive: mice with targeted disruption of the nrf2 gene ARE prone to develop lesions in response to environmental toxicants, carcinogens, drugs and inflammatory insults. ChIP-seq in mouse embryonic fibroblasts and human lymphoblastoid cells found several hundred NRF2 targets in processes other than oxidative stress — protein transport, phosphorylation, cell cycle, growth, adipogenesis, iron and heme metabolism. NRF2 is not a dedicated antioxidant switch. It reduces Oxidative stress.
Itoh 1997, BBRC 236:313 — Nrf2 drives ARE-linked phase-II antioxidant genes that neutralize ROS. DOI 10.1006/bbrc.1997.6943 Verbatim: “As an important antioxidant factor, NRF2 activation reduces ROS levels and prevents oxidative stress”. doi 10.1006/bbrc.1997.6943 · PMC11748272
It increases Pigment stem cells.
NRF2 provides a protective effect in melanocytes against both UV and oxidative stress, through transcriptional upregulation of antioxidant response element genes including SOD-1 and the glutathione system (PMC7603349). A second sensor sits alongside it: ATM expression rises in primary human scalp follicle melanocytes on H2O2 exposure, that rise is prevented by antioxidants, and chemically inhibiting ATM promotes loss of melanocyte viability under oxidative stress — loss-of-function confirming ATM is protective, not merely correlated. ATM expression correlates positively with pigmentary status in graying follicles. PMC7603349
It increases Hair.
H2O2 induced premature catagen and hair growth inhibition, whereas Nrf2 activation significantly ameliorated H2O2-dependent hair growth inhibition. Damage and rescue in the same experimental system is what lifts this above an antioxidant marketing claim. The mechanism is specified: activation of Nrf2 signaling enhances cellular resistance to oxidative damage through induction of antioxidant and cytoprotective enzymes. And there is a second route worth noting because it lands on a node already drawn here: growing evidence demonstrates functional crosstalk between Nrf2-mediated antioxidant defenses and Wnt/BETA-CATENIN signaling — Nrf2 activation can stabilize beta-catenin and enhance expression of its downstream targets. the Wnt → Hair Growth link is drawn, so part of this edge may run through it. Cultured human dermal papilla cells throughout; no in vivo Nrf2 manipulation with hair endpoints was retrieved. PMC10181221
It increases Tissue renewal signaling.
Growing evidence demonstrates functional crosstalk between Nrf2-mediated antioxidant defenses and Wnt/beta-catenin signaling — Nrf2 activation can stabilize beta-catenin and enhance the expression of its downstream targets. This says they are not two separate benefits — part of what antioxidant defense does for a follicle it does by stabilizing the growth switch. The phrasing in the source is growing evidence and can stabilize, not established mechanism. PMC12898851
