Oxidative stress
The balance here is dose, not direction — and the numbers make the point: of the 69 things this drives, fifty-seven are harmful and only two are not. Those two are the whole story. A brief oxidative pulse is the signal that switches on Nrf2 and AMPK — it is how exercise and fasting produce adaptation at all, and it is why high-dose antioxidants taken around training blunt the result you trained for. Everything else is the cost of the Smouldering Version: DNA damage, nf-κB, apoptosis, endothelial dysfunction, hearing, vision, skin. You want the spike and not the baseline. Nobody needs a higher resting level.
In depth
Reactive oxygen — at low levels a signaling messenger that sharpens metabolism and defense; in excess, the oxidative fire that corrodes cells.
What drives it
Training load increases it.
Exercise generates reactive oxygen/nitrogen species in working muscle, and these RONS are not merely damage — they are the signal that drives the adaptive response, including Nrf2 activation (PMC6617290; human skeletal-muscle Nrf2/Keap1 during exercise to exhaustion, PMC7358388). The proof is that blocking it blocks the benefit: exercise-induced ROS regulate AMPK, MAPK, NRF2 and PGC-1-alpha, driving increased glucose uptake, mitochondrial biogenesis and hypertrophy — and most of these adaptations ARE blunted by antioxidants, which the authors state reveals the crucial role ROS play during and after exercise. Sources are named: NADPH OXIDASES, xanthine oxidase and mitochondria. And a paradox that kills the simple reading: ROS generation also rises in skeletal muscle after prolonged disuse (immobilization). ROS is not a marker of activity — both loading and unloading raise it, and what differs is the context in which it is read. PMC6617290
Heat instead of storage reduces it.

Tissue remodeling reduces it.
10.3389/fbioe.2022.908033 — The heart-failure and atherosclerosis reviews both discuss fibrosis and oxidative stress in the same breath: oxidative stress and chronic, low-grade inflammation ARE important contributors, and metabolic abnormalities activate pro-fibrotic pathways resulting in cardiac stiffening. Sign and direction ARE both less settled than the source count suggests, and this is exactly the known false-positive mode: twelve sources touching both nodes is not twelve sources claiming a relation. Drawn inhibitory on the reading that established fibrotic tissue sustains oxidative stress rather than resolving it, but the dominant documented direction in these reviews runs the Oxidative Stress (ROS) → Type-I Procollagen link. doi 10.3389/fbioe.2022.908033
Oxidant-producing enzyme increases it.
NOX is distinct from other enzymatic sources because its primary function is to produce ROS; NOX enzymes are considered the main source of ROS production in endothelial cells, and the major sources of superoxide generated in the body are NADPH oxidases. doi 10.2337/db12-0009 · PMC7880919

What it drives
It reduces Protects light-sensing cells.
In rd1 mice, markers of oxidative damage (protein carbonyl adducts, acrolein staining) rose in the retina as photoreceptors degenerated, and an antioxidant combination roughly doubled surviving cone density and partly preserved cone electroretinogram function, showing that oxidative stress drives photoreceptor death. doi 10.1073/pnas.0604056103 · PMID 16849425
It increases Antioxidant defense.
Mitochondrial ROS activate Nrf2 (review, PMC7072240): rising ROS modify Keap1 cysteine residues, which releases Nrf2 from degradation so it translocates and switches on the antioxidant/are program. With the existing the Nrf2 ⊣ Oxidative Stress (ROS) link this closes the hormetic feedback loop — mild oxidative stress from caloric restriction or exercise (mitohormesis) raises the very defense that then lowers ROS. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: ROS oxidize Keap1 sensor cysteines (Cys-151, -273, -288 in mouse), releasing Nrf2 from the Keap1/cullin-3 E3-ubiquitin ligase complex and preventing its degradation, so Nrf2 accumulates and translocates to the nucleus to drive are genes. Not the only route: a non-canonical p62-dependent pathway activates Nrf2 independently of Keap1 oxidation, and growth factors can activate Nrf2 via PI3K/GSK3-beta. Verbatim: “Reactive mouse Keap1 cysteines (Cys-151, -273, and -288) are redox sensors, and upon oxidation by ROS, results in the dissociation of Nrf2 from Keap1/Cul3 which allows Nrf2 stabilization and translocation into the nucleus”. doi 10.1016/j.cellsig.2012.01.008 · PMID 22286106 · PMC7072240
It reduces DNA methylation.
Oxidative-stress/epigenetics study 2022, Int J Mol Sci — oxidative stress reduces global 5-methylcytosine and DNMT3a (with increased 5hmC/TET2), lowering DNA methylation. doi:10.3390/ijms24010153. (Biology Insights 2025 review of 8-oxoG epigenetics; Trends Cell Biol S0962-8924(23)00045-4). Oxidative stress produces global hypomethylation at repetitive elements and promoter regions, and oxidative lesions recruit TET enzymes that perform active demethylation. But the effect is dual and context-dependent, and the source says so: some loci instead undergo hypermethylation, silencing tumor-suppressor genes. The reviewers state this dual effect complicates interpretation because the same oxidative stress can either enhance OR suppress gene activity depending on context. A single global arrow cannot express that. An UNMAPPED reverse arrow exists: the DNA demethylation reaction itself produces ROS as by-products, generating 8-oxoG -- so methylation turnover feeds back into oxidative load. doi 10.3390/ijms24010153.
It increases Hypoxia response.
Chandel 2000 — mitochondrial (complex III) ROS are required and sufficient to stabilize HIF-1α; exogenous H2O2 stabilizes HIF-1α even under normoxia. PMID 10833514. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: oxidants activate HIF-1-alpha among the redox-responsive transcription factors, and HIF-1-alpha is further regulated by Ref-1 redox mechanisms. PMID 10833514
It increases Energy sensing.
AMPK/mROS study 2017 — mitochondrial reactive oxygen species are a physiological activator of AMPK, triggering a PGC-1α-dependent antioxidant response. PMID 28978464. Functionally confirmed: H2O2 and the glycolysis blocker 2-DG induced AMPK phosphorylation and autophagy in endothelial cells, and this was blocked by the antioxidants Tempol and N-acetylcysteine or by overexpressing superoxide dismutase 1 or catalase (PMC3046135). But the direct model failed its test: it was proposed that ROS act on redox-sensitive cysteines (Cys-299/Cys-304) on the AMPK alpha subunit. Murphy's group assessed whether physiological ROS directly alter AMPK activity and titled the result 'mitochondria-derived ROS activate AMPK indirectly' (J Biol Chem 2018, doi 10.1074/jbc.RA118.002579) — the effect runs through altered adenine-nucleotide ratios, not direct cysteine oxidation. And ROS here can be protective: conditional endothelial Nox2 overexpression raising EC-ROS 1.8-fold improved coronary vasodilation via a CaMKK-beta-AMPK-eNOS axis and induced autophagy that protected cells from oxidant death (PMC3765580) — the authors note recent antioxidant trials in cardiovascular disease failed, which challenges the blanket anti-ROS position. doi 10.1074/jbc.RA118.002579 · PMID 28978464 · PMC3046135
It increases Growth & survival signaling.
PTEN-redox review 2024, Antioxidants — ROS (H2O2) oxidize PTEN at its active-site cysteine, inactivating it and thereby de-repressing PI3K/AKT signaling. PMC10886030. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified, and the mechanism is inactivation of a brake, not direct activation: H2O2 oxidizes and inactivates human PTEN via a disulfide bond between catalytic Cys-124 and Cys-71. Endogenously generated ROS following insulin, EGF or PDGF causes PTEN oxidation leading to activation of the PI3K pathway. It is reversible — peroxiredoxin II reverses PTEN oxidation. Verbatim: “It was also demonstrated that endogenously generated ROS following treatment with peptide growth factors such as insulin, EGF, or PDGF causes oxidation of PTEN leading to the activation of the PI3K pathway”. doi 10.1016/j.cellsig.2012.01.008 · PMID 22286106 · PMC10886030

ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: oxidants activate p53 among the redox-responsive transcription factors; p53 is also subject to Ref-1 redox regulation. doi 10.1158/0008-5472.CAN-04-0494.

It increases Vessel growth signal.
Oxidative-stress/angiogenesis review 2014, Blood — reactive oxygen species act as signaling molecules that stimulate VEGF expression and angiogenesis. PMID 24300855. Mechanism runs by two separate routes: the VEGF-dependent route raises VEGF expression via MAPK and PI3K/Akt/mTOR-PTEN signaling through p70S6K1 and HIF-1-alpha; the VEGF-independent route drives angiogenesis via oxidized lipid ligands activating NF-kappa-B through Toll-like receptors. Loss-of-function: siRNA knockdown of NADPH oxidase NOX4 reduced insulin-stimulated ROS and inhibited VEGF transcriptional activity through HIF-1. this is physiological-level H2O2 acting as a signaling molecule, not oxidative damage. The concentration decides the outcome, including in cancer — stated here as the literature states it: at moderate concentration ROS activates cancer-cell survival signaling (MAPK/ERK1/2, p38, JNK, PI3K/Akt), which in turn activates NF-kappa-B, MMPs and VEGF — augmenting tumorigenesis and angiogenesis. At high concentration ROS causes cancer-cell apoptosis. The same molecule promotes or kills tumors depending on level (PMC6920770). This is recorded because it is what the evidence says. PMID 24300855 · PMC6920770
It reduces Vision.
AMD oxidative-stress review 2018 — excess reactive oxygen species damage the retinal pigment epithelium, driving macular degeneration and loss of central vision. PMC6230566. The mechanism: the retinal pigment epithelium is highly susceptible to oxidative damage because every one of its jobs -- photoreceptor outer-segment phagocytosis, retinoid cycling, blood-retinal barrier transport, light absorption by melanin and lipofuscin -- requires active oxidative metabolism. Excess ROS oxidize DNA, proteins and lipids; lipid oxidation degrades membrane and lipoprotein function and triggers inflammation; mitochondria and lysosomes break down, killing RPE and then photoreceptor cells. The dose-dependence is confirmed independently here: reactive oxygen intermediates are described as mainly byproducts of physiological processes and part of a normal feedback loop in redox signaling -- damage arises from OVEREXPOSURE plus impaired antioxidant defense with aging, not from ROS as such. AMD is multifactorial -- oxidative stress, chronic inflammation, immune dysregulation and genetic predisposition. ROS is one of four named contributors, not the account. And the 2025 review states plainly that effective therapeutic options remain limited and there is NO definitive cure for AMD. Verbatim: “This imbalance contributes to cellular damage and neurodegeneration, ultimately leading to vision loss”. doi 10.3390/ijms26083463 · PMC6230566
It reduces NRF2 restraint.
Dinkova-Kostova 2002, PNAS 99:11908 — oxidants/electrophiles react with Keap1’s cysteine thiols, disrupting Keap1-Nrf2 and releasing Nrf2. PMID 12193649. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified, and this is the same event read from the other side: oxidants and electrophiles react with Keap1's reactive cysteines, disabling its ubiquitination activity. Keap1 also acts as a direct H2O2 sensor by forming an intramolecular disulfide bond. PMID 12193649
It increases Calms inflammation.
Schreck, Rieber & Baeuerle 1991, EMBO J 10:2247 — reactive oxygen intermediates are common messengers activating NF-κB. PMID 2065663. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: oxidants activate the transcription factors Nrf2, HIF-1-alpha, AP-1, NF-kappa-B, FoxO and p53. NF-kB, p53 and HIF-1-alpha are additionally regulated by the redox factor Ref-1 under thioredoxin control. Verbatim: “NOX2, predominantly expressed in immune cells, produces ·O2− that activate the NF-κB signaling pathway, inducing pro-inflammatory cytokines like IL-6 and TNF-α, exacerbating acute inflammation, and damaging the mucosal barrier”. doi 10.1080/13510002.2025.2458942 · PMID 2065663 · PMC11792148
It increases Cell turnover.
Simon 2000, Apoptosis 5:415 — ROS mediate cytochrome-c release and caspase activation, triggering apoptosis. PMID 11256882. (PMC3920001; Exp Mol Med 2022 doi 10.1038/s12276-022-00822-z). Separately, PCBP1 binding to heavily oxidized o8G-RNA initiates a damage-signaling pathway leading to apoptotic death, with reduced caspase-3 activation and PARP cleavage in its absence. the closely related PCBP2 binds the same oxidized RNA but suppresses ROS-mediated cell death -- the same lesion routed through two paralogues gives opposite outcomes. Verbatim: “Chronic hyperglycemia induces an increase in reactive oxygen species (ROS), leading to cell apoptosis”. doi 10.1038/s12276-022-00822-z · PMID 11256882 · PMC3920001
It reduces Skin.
Photoageing is an oxidative process, and it acts on the same endpoint the procollagen lane above measures. UV exposure raises ROS, which up-regulates the matrix metalloproteinases (MMP-1, -3, -9) that degrade collagen, elastin and fibronectin — producing deep wrinkles and sagging (PMC8597149, PMC9988502). So the two lanes into this pathway are two ends of one balance: synthesis versus degradation. Verbatim: “Increased ROS levels due to PM exposure critically damage the respiratory and cardiovascular systems, other organs, and skin”. doi 10.7717/peerj.16589 · PMC8597149
