Hypoxia response
Hypoxia/HIF-1α

You want it responsive, not stuck: adaptive in true hypoxia, quiet otherwise.
In depth
HIF-1α — the low-oxygen sensor that reprograms metabolism to adapt, and a driver of disease when chronically switched on.
What drives it
Oxidative stress increases it.
Chandel 2000 — mitochondrial (complex III) ROS are required and sufficient to stabilize HIF-1α; exogenous H2O2 stabilizes HIF-1α even under normoxia. PMID 10833514. ROS is dose-dependent: physiological ROS signals, excess ROS damages. 'Lower ROS' is therefore not universally desirable. Verified: oxidants activate HIF-1-alpha among the redox-responsive transcription factors, and HIF-1-alpha is further regulated by Ref-1 redox mechanisms. PMID 10833514
What it drives
It increases Sugar burning.
Semenza 1994, J Biol Chem 269:23757 — HIF-1 directly transactivates glycolytic enzyme genes (aldolase A, PGK1, PKM) via hypoxia-response elements. PMID 8089148
It increases Vessel formation.
Hypoxia is the signal that builds new vessels, and HIF-1 is the transducer: it is the most potent inducer of the genes encoding VEGF, the glycolytic enzymes and erythropoietin, and it is the central mediator of angiogenesis under low oxygen (PMC4656338, PMC11079266). Shown causally rather than by association — knockdown of either HIF-1α or HIF-2α strikingly attenuated hypoxia-induced VEGF production by endothelial cells and their colony and tube formation (PMC6366960). This lane runs alongside the VEGF one because HIF is upstream of it, not parallel to it. PMC4656338
It increases The Second Padlock.
HILPDA, the hypoxia-inducible lipid-droplet-associated protein, is a direct HIF-1alpha target gene. PMID 20624928
