Acute vs chronic: tgf-β is how a wound closes and how collagen is laid down. Held on, the same signal becomes fibrosis — in the liver, the heart and the follicle. You want it to finish the job and stop. You want measured repair, not runaway scarring: enough to rebuild, restrained enough to keep tissue supple.

In depth

TGF-β / Smad signaling — the builder that lays down collagen and directs repair, and the same hand that, overused, drives fibrosis.

What drives it

Blocks the hormone that shrinks follicles increases it.

Androgen-induced TGF-β1 from dermal papilla cells is an established route of follicular growth inhibition in AGA co-culture models (AGA transcriptome review, PMC8484716); the DHT–androgen-receptor complex up-regulates TGF-β1, IL-6 and DKK-1 while down-regulating Wnt agonists. Demonstrated in COCULTURE, not inferred: with AR-overexpressing human dermal papilla cells from AGA scalp cocultured with normal human keratinocytes, the androgen R1881 suppressed keratinocyte growth through androgen-inducible TGF-beta-1. The founding paper is titled 'androgen-inducible TGF-BETA1 from balding dermal papilla cells inhibits epithelial cell growth: A CLUE to understand paradoxical effects of androgen on human hair growth' (Inui 2002). Human tissue confirmation: TGF-beta-1 protein was elevated in balding follicles versus non-balding occipital follicles from the same 15 men (Lu 2016). PMC8484716

Oxidative stress increases it.

Liu & Desai 2015, Redox Biol — redox imbalance (ROS) activates latent TGF-β1 (oxidation of the latency-associated peptide) and induces TGF-β1 expression. PMID 26496488 Verbatim: “ROS can activate latent TGF-β1 (LTGF-β1) through direct oxidation of latency-related peptide (lap) or indirect activation of matrix metalloproteinase-2 (MMP-2) and MMP-9”. doi 10.3389/fnut.2022.850689 · PMID 26496488 · PMC9197188

Follicle oxidative stress increases it.

Balding dermal papilla cells secreted higher levels of the negative hair growth regulators transforming growth factor beta 1 and 2 in response to H2O2. Replicated in independent work: HFDPCs from balding scalp ARE more vulnerable to oxidative stress, leading to enhanced secretion of TGF-beta, a well-established inhibitor of hair growth, and accelerated cellular senescence. The point: peroxide is not corroding the hair. It is making the papilla issue the shutdown order. That is why an antioxidant that reaches the papilla is a different proposition from one that does not and a specificity the first paper insists on: the TGF-beta rise occurred in response to H2O2 but not cell culture-associated oxidative stress. Not all oxidative stress is the same stimulus. doi 10.1038/JID.2015.28 · PMC12898851

Galectin-3 increases it.

The core mechanism, and it is not what people assume. Galectin-3 does not create the fibrosis instruction; it keeps the receiver switched on. Verbatim: 'Galectin-3 has been shown to prolong cell surface residence and thus enhance responsiveness of the TGF-beta receptor (Partridge et al., 2004), which in turn regulates alternative macrophage differentiation into M2 macrophages and myofibroblast activation.' Independently stated for the heart: it 'can regulate the effects of other molecules involved in fibrosis processes, such as cytokines, by retaining their receptors on the surface membrane of the atrial myofibroblast' (PMC8393977). And: 'Galectin-3 stimulates an M2-like phenotype in macrophages, stimulating synthesis of fibroblast-activating mediators, such as IL-10 and TGF-betas'. doi 10.1093/cvr/cvad156 · PMC8393977

What it drives

It reduces Hair.

Foitzik 2000, FASEB J 14:752 — TGF-β1 is an endogenous inducer of catagen (the follicle regression phase), inhibiting keratinocyte proliferation and triggering apoptosis; this is the catagen signal apigenin down-regulates. PMID 10744631. Mechanism is catagen entry — the growth phase is ended, not slowed. The in-vivo paper is titled 'control of murine hair follicle regression (catagen) by TGF-BETA1 in vivo' (Foitzik 2000). Paired human biopsies: in 15 AGA men, balding frontal follicles showed elevated TGF-beta-1 protein and elevated T-beta-RI and T-beta-RII versus their own non-balding occipital follicles — an internally controlled comparison rather than between-subject. PMID 10744631

It increases Tissue remodeling.

Wang 2013, PLoS One — TGF-β/Smad2/3 signaling drives type I/III collagen synthesis in human dermal fibroblasts. doi:10.1371/journal.pone.0055899. TGF-beta is described as a potent inducer of collagen synthesis; in normal human dermal fibroblasts TGF-beta signaling increased collagen production, and in human cardiac myofibroblasts TGF-beta1 induced collagen synthesis, gel contraction and structural ECM remodeling. the route runs through fibroblast-to-myofibroblast transition — myofibroblasts do not generally exist in normal tissue and appear in injured, fibrotic and cancer tissue. This is a REPAIR/PATHOLOGY program, not baseline collagen maintenance. Fn14 siRNA blocked ECM gene expression even when TGF-beta1 was active, so the arrow is not sufficient on its own. doi 10.1371/journal.pone.0055899.

It increases Cell migration switch.

Ramachandran 2018, eLife — tgf-β-induced epithelial-to-mesenchymal transition requires smad signaling (SMAD3 with SMAD1/5). Doi:10.7554/eLife.31756. TGF-beta is the most potent factor engaged to initiate and complete emt. emt and its reverse, met, both occur during physiological wound healing as well as pathological fibrosis — emt is not intrinsically pathological, and the same transition serves repair. doi 10.7554/eLife.31756.

It reduces Liver.

TGF-β hepatic fibrosis review 2006 — tgf-β1/Smad3 activates hepatic stellate cells to deposit collagen, driving liver fibrosis. PMID 16420505. Implicated across hepatitis, cirrhosis and hepatocellular carcinoma. Tgf-beta is not one signal — its effect depends on which smad phospho-isoform is engaged, and the paper is titled for it: 'reversible human tgf-beta signal shifting between tumor suppression and fibro-carcinogenesis'. Four forms exist: cytostatic pSmad3C, mitogenic pSmad3L (Ser-213), fibrogenic pSmad2L/C and migratory pSmad2/3L/C. After acute injury tgf-beta raises pSmad3C and terminates proliferation — the tumor-suppressor arm. Under chronic injury the linker-phosphorylated fibrogenic and mitogenic arms dominate. PMID 16420505

It increases Supports heart function.

TGF-β is the switch that turns load into scar: myocardial TGF-β is upregulated after infarction, in cardiac hypertrophy and in dilated or hypertrophic cardiomyopathy; it transduces hypertrophic signals in cardiomyocytes and drives myofibroblast transdifferentiation with matrix deposition, and overexpression in the mouse heart produces fibrosis and hypertrophy (PMC3072437). Note — the effect is rather than simply bad: blocking TGF-β receptor signaling after pressure overload markedly reduced collagen deposition but produced greater ventricular dilation and systolic dysfunction. A baseline level appears necessary to hold the structure together. Both canonical and non-canonical TGF-beta pathways are implicated in cardiac fibrosis; TGF-beta1 induced significant myofibroblast activation and ECM dysregulation in human ventricular and atrial cardiac myofibroblasts. Structural cardiac remodeling with myocardial fibrosis is a hallmark of progression to heart failure. LMW-FGF-2 significantly attenuated TGF-beta1-induced myofibroblast-mediated ECM remodeling — the pathway is modifiable downstream of TGF-beta itself. PMC3072437

It increases Cell turnover.

Intravital microscopy in live mice: elimination of epithelial cells during hair follicle regression involves basal cell apoptosis through synergistic action of TGF-beta and mesenchymal-epithelial interactions — and crucially the basal epithelial cells ARE not internally committed to death; the mesenchymal dermal papilla plays an essential role in death induction. The cell is not dying of its own accord. It is being told to. Corroborated from the catagen-trigger side: TGF-beta1 and TGF-beta2 are listed among the defined apoptotic triggers promoting catagen alongside Bcl2/Bax, p53 and p57. And it is the convergence point of this whole node: dht acts through TGF-beta, oxidative stress acts by making the papilla secrete TGF-beta, and the vascular loss on the Endothelial (eNOS) → Hair Growth link is AR-mediated TGF-beta killing endothelium. Three drivers, one messenger, one executioner. doi 10.1016/j.jid.2022.01.005 · PMC10588663

It reduces Gum health.

The mechanism is a self-feeding loop and the fuel is your own tissue: inflammation and dysbiosis positively reinforce each other because inflammatory tissue breakdown products ARE used as nutrients by the dysbiotic microbiota - specifically collagen peptides and heme-containing compounds. The bacteria eat the damage the immune response causes, which causes more damage. And it is not an infection in the ordinary sense: periodontitis is caused mainly by dysbiosis of the bacterial community, rather than an infection by individual bacteria. Keystone pathogens, whose colonization is facilitated by accessory pathogens, initially subvert the host response, after which PATHOBIONTS - commensals turned pathogenic - over-activate inflammation. A keystone pathogen works at low abundance by changing the community, not by dominating it. The tissue-destruction route is specific: pathogens induce epithelial-mesenchymal transition via TGF-beta/Smad, Wnt/beta-catenin and Notch, causing loss of E-cadherin, junctional epithelial instability, pocket formation and attachment loss. doi 10.3390/jmp7010011 · PMID 25534621 · PMC5646588
