Vessel growth signal
VEGF/HIF-1α

The balance here is between goals — blood supply is good until it serves the wrong thing. Raised, it drives hair growth, bone density, aerobic capacity, muscle recovery, hearing and erectile function: all of these are downstream of building new vessels. The costs shown here are ocular and angiogenic. And the cost that matters most after fifty is the one least visible here: tumor angiogenesis is vegf-driven, which is why anti-vegf drugs are cancer drugs. The count of benefits below looks decisive and is not.
In depth
The angiogenic signal — the instruction to grow new blood vessels. Directionally double-edged: essential for wound healing and muscle adaptation, and exploited by tumors for the same reason.
What drives it
Oxidative stress increases it.
Oxidative-stress/angiogenesis review 2014, Blood — reactive oxygen species act as signaling molecules that stimulate VEGF expression and angiogenesis. PMID 24300855. Mechanism runs by two separate routes: the VEGF-dependent route raises VEGF expression via MAPK and PI3K/Akt/mTOR-PTEN signaling through p70S6K1 and HIF-1-alpha; the VEGF-independent route drives angiogenesis via oxidized lipid ligands activating NF-kappa-B through Toll-like receptors. Loss-of-function: siRNA knockdown of NADPH oxidase NOX4 reduced insulin-stimulated ROS and inhibited VEGF transcriptional activity through HIF-1. this is physiological-level H2O2 acting as a signaling molecule, not oxidative damage. The concentration decides the outcome, including in cancer — stated here as the literature states it: at moderate concentration ROS activates cancer-cell survival signaling (MAPK/ERK1/2, p38, JNK, PI3K/Akt), which in turn activates NF-kappa-B, MMPs and VEGF — augmenting tumorigenesis and angiogenesis. At high concentration ROS causes cancer-cell apoptosis. The same molecule promotes or kills tumors depending on level (PMC6920770). This is recorded because it is what the evidence says. PMID 24300855 · PMC6920770
Growth factor increases it.
IGF-1 boosts VEGF expression, which supports microcirculation and nutrient delivery to hair follicles, and it promotes VEGF expression, enhancing angiogenesis around the hair follicle. IGF-1 to hair and VEGF to hair are both drawn; this link says the first is partly upstream of the second rather than parallel to it. PMID 41020895 · PMC12468416
Adenosine A2B receptor increases it.
Adenosine upregulates VEGF gene expression in cultured dermal papilla cells. Minoxidil-induced VEGF production in these cells is itself blocked by an adenosine-receptor antagonist, which is how the adenosine route was first identified. cultured human cells. Iino 2007, J Invest Dermatol 127(7):1825. PMID 17301835

It increases Bone health.
Named across six reviews of bone remodeling and osteoarthritis pathogenesis. Bone remodeling relies on the coordinated actions of bone-resorbing osteoclasts, bone-forming osteoblasts, and osteocytes serving as mechanosensors and local remodeling coordinators. PMC10648156
It increases Sexual function.
Named across five reviews of erectile and female arousal physiology. Key contributors include reduced nitric oxide bioavailability due to endothelial oxidative stress and inflammation, diminished cavernosal smooth muscle content leading to impaired veno-occlusion, and increased arterial stiffness compounded by metabolic disorders. doi 10.21037/tau-2025-419 · PMID 19138376 · PMC11988752

It increases Physical performance.
A negative that corrects the obvious assumption: at least the structural aspects of the capillary bed do not appear to be reduced in a manner that would compromise the capacity for muscle oxygen diffusion even in senescence. Capillary rarefaction is the intuitive explanation and this review says it is not the limiting one. doi 10.1139/H07-174 · PMC10102650
It increases Hearing.
Mitochondrial antioxidants, modulators of mitochondrial function, calcium channel blockers, and mtDNA mutations that affect mitochondrial activity in the auditory system have emerged as critical factors in hearing protection. The reviewers state their own gap: there is still a lack of information on the underlying mechanisms of redox homeostasis and molecular redox networks in hair cells - so these are protective-target candidates, not demonstrated interventions. The clinical fact that makes protection the only strategy: mammalian cochlear hair cells do not regenerate, which is why SNHL is described as the most common type of permanent hearing impairment. doi 10.1016/j.mito.2025.102063 · PMC10102650
It increases Recovery.
PMC1764718 — Recovery is assembled from the sarcopenia and VO2max literatures; none of the papers cited studied recovery itself. PMC1764718
It reduces Blood pressure.
The pharmacology is the evidence and it is unusually clean: VEGF-pathway inhibitors used in oncology cause hypertension as a class effect - which establishes that VEGF signaling is required to maintain normal vascular tone, and removing it raises pressure. The sign is counterintuitive and deserves care: VEGF supports normal vascular tone (so the edge is drawn as beneficial to bp), and it is the blockade of VEGF that raises pressure. Drawn on the drug-class effect, which is an inference from the inhibitor to the ligand - a negative-evidence argument, and weaker than a direct measurement. doi 10.1038/s41581-026-01066-0 · PMC11725408
It reduces Vision.
Why the retina is the most oxidatively stressed tissue in the body, stated directly: the retina is highly susceptible to oxidative damage due to its elevated oxygen consumption, abundant polyunsaturated fatty acids, and continuous exposure to light. Three risk factors that cannot be separated from its function. And the aging failure is specific and double: in the elderly both the mitophagic process and Nrf2 activation ARE impaired, thus causing retinal cell death. The carotenoid case is unusually mechanical: lutein and zeaxanthin ARE selectively taken up by the RPE, preferentially accumulated in the human macula, and transferred to photoreceptors - a targeted delivery route, not a general antioxidant claim. The Eye Disease Case-Control Study found high plasma levels of lutein and zeaxanthin associated with reduced risk of neovascular AMD. By contrast the reviewers say the relationships between dietary and serum levels of the antioxidant vitamins and age-related macular disease ARE less clear - with alpha-tocopherol the exception, convincingly demonstrated. Lipofuscin is itself a photoreactive substance - the waste product becomes a new damage source. doi 10.3390/antiox14101251 · PMID 11033038 · PMC12026614
It increases Wound healing.
The one measured finding among them is the periodontal-anemia link: a meta-analysis of sixteen studies on hemoglobin and iron metabolism in periodontitis, whose authors call the association still controversial and ask for larger cohorts. doi 10.1152/japplphysiol.00971.2016 · PMC7005133
It increases Female fertility.
The central finding is that reactive oxygen species ARE required, not merely harmful: ROS in the ovary regulate ovarian steroid hormone production, follicle synthesis, maturation, ovulation, and tubal function. ROS and antioxidant levels regulate the microenvironment of the follicular fluid, which determines which follicle becomes dominant. ROS can trigger ovulation in mature GRAAF follicles, and any change in this ROS level can disrupt ovulation. So an antioxidant claim here cuts both ways - the requirement is redox homeostasis, not suppression. Excess oxidative stress damages the oocyte through mitochondrial dysfunction, follicular atresia, meiotic errors, DNA damage and telomere shortening; too little disrupts the ovulatory trigger itself. Scale: infertility affects approximately 10-15 percent of couples, with nearly half the cases attributed to female reproductive dysfunction. doi 10.3390/antiox12081490 · PMID 40712896 · PMC10666387
