Adenosine A2B receptor

Dermal papilla A2B signaling

The Other Adenosine Receptor

The same molecule, two entirely different stories, and keeping them apart is the whole point of this pathway. Caffeine works by blocking A1 — that has nothing to do with hair. On the dermal papilla, adenosine binding A2B raises FGF-7 more than two-fold, and the proof is unusually clean: the rise disappears when an A2B-specific blocker is added and survives blockers for every other subtype. Downstream it inhibits GSK3-beta, which sustains Wnt/beta-catenin signaling — the pathway that keeps a follicle in its growth phase. Japan grades topical adenosine as recommended for pattern hair loss, replicated in Japanese and in Caucasian men. One thing must travel with all of it: every trial applied it to the scalp.

In depth

Adenosine acts through four different receptors, and they do not do the same job. A1 is the one that makes you sleepy and blunts alertness. A2B is the one on the dermal papilla cells at the base of a hair follicle, and it is the most abundant subtype there.

What drives it

Curbs the main NAD⁺ consumer increases it.

Supplies the ligand to the receptor, by the established route: extracellular NAD+ is converted to AMP by CD38 and CD203a, and AMP is hydrolyzed by CD73 to adenosine — which is the agonist at the A2B receptor. Drawn so the A2B node is not an orphan and so the CD38 route to adenosine signaling is traceable end to end. enzymatic chain, extracellular arm; no human trial measures it and NO botanical claim attaches. What it drives

It increases Hair.

Receptor-specific, not generic adenosine: adenosine raises FGF-7 in human dermal papilla cells more than two-fold at mRNA and protein level, and the rise was abolished by the A2B-specific antagonist alloxazine but not by antagonists for the other subtypes — so the hair effect is A2B-mediated and does not belong to the A1 node. A2B is also the most abundant adenosine receptor in human dermal papilla cells. Clinical: the Japanese Dermatological Association grades topical adenosine recommendation B for androgenetic alopecia, replicated in japanese men (Watanabe, thick-hair ratio) and independently in Caucasian men (Iwabuchi, proportion of thick hair), plus a head-to-head against 5% minoxidil. Route limit, decisive for any blend: all of this evidence is topical (scalp application). No oral adenosine trial supports it, so this edge is mechanism and topical-clinical only and must not be used to grade an ingestible driver. It increases Tissue renewal signaling.

A2B activation inhibits GSK3-beta phosphorylation and thereby stimulates Wnt/beta-catenin signaling in cultured human dermal papilla cells, which is the pathway that sustains anagen. cultured human dermal papilla cells, not an in-vivo human outcome. Iino 2007, J Invest Dermatol 127(7):1825. PMID 17301835

It increases Growth factor.

Adenosine upregulates IGF-1 gene expression in cultured dermal papilla cells alongside FGF-7 and FGF-2. gene-expression endpoint in cultured human cells. Iino 2007, J Invest Dermatol 127(7):1825. PMID 17301835

It increases Vessel growth signal.

Adenosine upregulates VEGF gene expression in cultured dermal papilla cells. Minoxidil-induced VEGF production in these cells is itself blocked by an adenosine-receptor antagonist, which is how the adenosine route was first identified. cultured human cells.
