What drives it
Mitochondrial pore reduces it.
10.1038/s41418-023-01187-0 — Once the pore opens the membrane potential collapses — measured by confocal microscopy and high-resolution respirometry in a pulmonary-hypertension model, where the compromised energetics degraded heart-muscle function. Verbatim: “PAH significantly affects mitochondrial function in RV; PAH is prone to mitochondrial permeability transition pore (mPTP) opening, thus decreasing the mitochondrial membrane potential”. doi 10.1038/s41418-023-01187-0 · PMID 34239697 · PMC8238598
Ceramide load reduces it.
Gudz 1997, J Biol Chem — cell-permeable ceramide directly inhibits mitochondrial respiratory-chain complex III, collapsing oxidative phosphorylation and generating ROS. PMID 9305864. Mechanism is electron diversion, not enzyme inhibition: ceramides impair mitochondrial function by diverting electrons in the electron transport chain into reactive oxygen species instead of using them for oxidative phosphorylation, diminishing the organelle's ability to generate ATP — the same electrons become damage instead of energy. Depletion rescues it: blocking ceramide synthesis with myriocin in high-fat-fed mice improved mitochondrial performance — increased oxygen consumption, increased citrate synthase activity, and preserved PGC-1 expression. The rescue was not clean: effects on lipid oxidation were unclear — long-chain acylcarnitines rose (suggesting incomplete oxidation) while short-chain fell. Ceramides also alter membrane permeability directly rather than acting purely as signals. PMID 9305864
Builds mitochondria increases it.
Ventura-Clapier 2008, Cardiovasc Res 79:208 — PGC-1α is the central transcriptional regulator of mitochondrial biogenesis and respiratory function. PMID 18430751. It regulates nuclear and mitochondrially-encoded electron-transport-chain subunits. PGC-1-alpha is one of a family — PGC-1-beta and PRC share overlapping functions, so it is not a unique switch; and it is itself regulated upstream by SIRT1/SIRT3, TFAM and AMPK. PMID 18430751

Muscle-oxidative-capacity study 2024 — skeletal-muscle mitochondrial oxidative capacity is associated with whole-body aerobic performance (VO2max); higher mitochondrial respiratory function supports endurance and lowers fatigability. PMC10828458
It reduces Vessel formation.
The body does not remodel delivery and utilization systems independently: PGC-1-alpha promotes VEGF expression, linking angiogenesis and mitochondrial adaptation so that oxygen delivery and oxygen utilization scale together. The causal pair is clean: overexpression of PGC-1-alpha increases skeletal muscle capillarisation, while deletion prevents exercise-induced angiogenesis — in part through PGC-1-alpha-regulated secretion of VEGF. Add it and vessels grow; remove it and exercise cannot grow them. And the dependence is stated as a rule of muscle architecture: the primary determinants of muscle fiber capillary density ARE fiber size and oxidative capacity — that is, mitochondrial volume. Highly oxidative fibers have both more mitochondria and greater capillary density; the vasculature is built to match the demand. Human corroboration in the elderly: lifelong endurance-trained subjects aged 71 had 27 percent more capillaries per fiber than untrained age-matched controls, alongside higher oxidative enzyme activities, and cycled 35 percent longer before exhaustion. What this changes: mitochondria and new vessels look like parallel drivers of physical performance. They are coupled, and PGC-1-alpha - also a driver of this outcome - is the coupler. doi 10.1371/journal.pone.0028290 · PMC9949767
It increases Hair.
The claim: once redox balance is broken, the dermal papilla cells that regulate the formation, proliferation and cyclicity of hair follicles show obvious oxidative stress characteristics such as mitochondrial dysfunction, cell apoptosis, and perifollicular inflammation, which is associated with follicle miniaturization. And it is self-reinforcing: mitochondrial dysfunction in HFDPCs can exacerbate ROS production, creating a vicious cycle that impairs hair follicle regeneration. It names three links already on this pathway in one sentence — mitochondrial dysfunction, apoptosis, and perifollicular inflammation — and the route to the executioner is specified: activation of the TGF-BETA1/SMAD pathway by ROS leads to upregulation of TGF-BETA1 and TGF-BETA2, triggering senescence and damage of dermal papilla cells. Review-level synthesis with a mechanistic chain rather than a single decisive experiment. PMC12913381
It reduces Hair pigment loss.
Proteomic analysis shows that graying is marked by the upregulation of proteins related to energy metabolism, mitochondria, and antioxidant defenses. Upregulation, not loss. The white hair is not metabolically quiet — it is running its energy and antioxidant machinery harder. That is the same shape as the oxidative finding on the hair node, where balding papilla cells had higher catalase and glutathione and handled stress worse. Twice now, in two separate follicular outcomes, more defense protein has accompanied worse outcome. Which means the sign is an inference, and it is stated as one: the edge is drawn inhibitory on the reading that mitochondrial insufficiency drives graying and the protein rise is compensatory. The alternative reading — that the metabolic upregulation is itself part of the pathology — is not excluded by this data, and the proteomics is descriptive rather than interventional. doi 10.7554/eLife.67437 · PMC8219384

It reduces Attention regulation.
The split of labor is the finding, and it is drug-resolved: DA may be more essential to attaching attention; NE may contribute more to executive function. Methylphenidate blocks DA reuptake; amphetamines block reuptake and increase production of both DA and NE, with the noradrenergic effects contributing to increased energy and enhanced executive function; atomoxetine acts primarily on NE with secondary effects of increasing prefrontal cortical DA. Four drugs, four profiles, one dissociation. And the reviewers refuse the simple story: a hypothesis suggesting too much OR too little of a single neurotransmitter will NO longer suffice. Anything selling a dopamine-deficit account of attention is selling what the field explicitly abandoned. (PMC4033238; doi 10.1016/j.biopsych.2011.03.003; PMC2265273. doi 10.1016/j.biopsych.2011.03.003 · PMC4033238
