In depth

PGC-1α — the master regulator of mitochondrial biogenesis, building new cellular powerplants and oxidative endurance.

What drives it

Training load increases it.

Pilegaard 2003, J Physiol 546:851 — acute exercise transiently activates transcription of PGC-1α in human skeletal muscle, initiating mitochondrial biogenesis. PMID 12563009. Intensity selects the isoform, and by different receptors: a single bout of low-intensity exercise raises PGC-1-alpha-b and -c mRNA via the beta-2 adrenergic receptor, whereas raising PGC-1-alpha-a requires high-intensity exercise and is independent of beta-2 AR. PGC-1-alpha-b is the predominant exercise-responsive isoform and shows the largest rise. 'Exercise raises PGC-1-alpha' is therefore three different arrows depending on how hard you go. Nitric oxide is not the mediator: neither pharmacological nos inhibition in rats nor genetic deletion of eNOS or nNOS in mice prevents exercise-induced PGC-1-alpha expression — NO supports PGC-1-alpha and fiber-type shift generally, but is dispensable for the exercise response itself. PMID 12563009

Cold exposure response increases it.

Puigserver 1998, Cell 92:829 — PGC-1α expression is dramatically elevated by cold exposure in brown fat and skeletal muscle, driving adaptive thermogenesis. PMID 9529258

Energy sensing increases it.

Jäger 2007, PNAS 104:12017 — AMPK directly phosphorylates PGC-1α (Thr177/Ser538), driving mitochondrial biogenesis. PMID 17609368

Longevity gene increases it.

Cantó 2009, Nature 458:1056 — SIRT1 deacetylates and activates PGC-1α. PMID 19262508. Mouse skeletal muscle. SIRT1 also deacetylates FOXO1 and FOXO3a in the same paper -- PGC-1a is one target among several, not the sole one. SIRT1 deacetylates PGC-1-alpha, and the effect is both post-translational and transcriptional: SIRT1 positively regulated the level of Pgc-1-alpha transcription, likely because a deacetylated PGC-1-alpha coactivates FOXO more strongly at its own promoter — a self-amplifying loop. In the SCN, neuronal SIRT1 and PGC-1-alpha bind cooperatively and in close proximity at the Bmal1 promoter. SIRT1 is NAD+-DEPENDENT, so this arrow only carries flux when NAD+ is available — see the NAD⁺ → SIRT1 link. PMID 19262508

Recycles mitochondria increases it.

Biogenesis–mitophagy crosstalk review 2023 — mitophagy is tightly coordinated with PGC-1α-driven biogenesis to renew the mitochondrial pool. doi 10.1186/s12929-023-00975-7

Calms inflammation reduces it.

Reverse ARROWTHE paper is titled for it: NF-kappa-B p65 attenuates cardiomyocyte PGC-1-alpha expression in hypoxia (Cells 2022;11:2193, doi 10.3390/cells11142193). Four independent lines in one paper: p65 associates with the PGC-1-alpha proximal promoter (EMSA and luciferase); TNF-alpha reduced PGC-1-alpha expression and p65 promoter binding; the effect was reversed by the NF-kappa-B inhibitor parthenolide; and p65-knockout cardiomyocytes showed altered chromatin-remodeling genes. Mechanism: p65 transcriptionally silences the PGC-1-alpha promoter by recruiting histone deacetylases — partially reversed by trichostatin A. And there is a second, independent mechanism: p65 physically binds PGC-1-alpha and inactivates it, an interaction stabilized by TNF-alpha-dependent NF-kappa-B activation (Antioxidants 2023;12:1075). Confirmed separately in a human cardiac cell model (PMID 19038972). This closes the vicious cycle the PGC-1α Biogenesis → NF-κB link describes: inflammation lowers PGC-1-alpha, low PGC-1-alpha lowers antioxidant genes, the resulting oxidative stress raises NF-kappa-B again. cardiomyocytes, hypoxia and TNF-alpha context. doi 10.3390/cells11142193 · PMID 19038972

Thyroid hormone conversion increases it.

T3-mediated expression of PGC-1alpha proceeds via a far upstream located thyroid hormone response element. Weitzel 2008, Mol Cell Endocrinol — T3-mediated expression of PGC-1alpha via a far upstream thyroid hormone response element. doi 10.1016/j.mce.2008.01.017

What it drives

It reduces Calms inflammation.

PGC-1α inhibits NF-κB, a master activator of proinflammatory gene expression, and additionally activates PPAR-α/γ which mediate anti-inflammatory effects (PMC8321869) — the mechanistic link from mitochondrial biogenesis to lower inflammatory tone. And the source states something stronger than this arrow: PGC-1-alpha and NF-kappa-B ARE reciprocally regulated, and the loop is vicious. During inflammation NF-kappa-B reduces PGC-1-alpha expression and activity; low PGC-1-alpha then downregulates antioxidant target genes, producing oxidative stress; and low PGC-1-alpha with that oxidative stress promotes NF-kappa-B activity, exacerbating the inflammatory response. PMC8321869

It increases Supports BDNF signaling.

PGC-1α-dependent route: exercise induces hippocampal Fndc5, a PGC-1α/ERRα-dependent myokine, which drives BDNF expression (Wrann 2013, documented in Sleiman 2016 eLife PMC4915811); BDNF then feeds back to inhibit Fndc5 via TrkB. Mouse evidence. PMC4915811

It increases Heat instead of storage.

Puigserver 1998, Cell 92:829 — PGC-1α coactivates the uncoupling-protein (UCP-1) promoter and induces the respiratory machinery of adaptive thermogenesis. PMID 9529258. (1) J Mol Med 2026, doi 10.1007/s00109-026-02694-6: in brown adipose tissue PGC-1-alpha promotes mitochondrial biogenesis and the expression of uncoupling protein 1, which uncouples the respiratory chain so protons re-enter the matrix without ATP synthesis, dissipating the gradient as heat. (2) PMC7601329: in brown fat PGC-1-alpha increases transcriptional activity at the UCP1 promoter. (3) The founding primary, Wu 1999, Cell 98:115-124, established PGC-1 as the thermogenic coactivator controlling mitochondrial biogenesis and respiration. Brown adipose and skeletal muscle specifically; PGC-1-alpha was originally identified as cold-induced, so this arm is a cold-adaptation program rather than a general metabolic lever. doi 10.1007/s00109-026-02694-6: · PMID 9529258 · PMC7601329

It increases Fatty-acid oxidation.

Vega 2000, Mol Cell Biol 20:1868 — the coactivator PGC-1α cooperates with (coactivates) PPARα to drive transcription of nuclear-encoded mitochondrial fatty-acid-oxidation genes.

It reduces Exports liver fat.

Chen 2010, J Biol Chem 285:25996 — PGC-1α stimulates hepatic VLDL assembly and secretion through activation of CideB.

It increases Physical performance.

PGC-1α-b overexpression study 2011, PLoS One — skeletal-muscle PGC-1α overexpression raises VO2max and exercise capacity via mitochondrial biogenesis and an oxidative fiber switch. doi:10.1371/journal.pone.0028290. Gain-of-function with a hard endpoint: muscle-specific overexpression of PGC-1-alpha-a improved exercise performance and produced about 20 percent higher peak oxygen uptake than wild-type; a separate transgenic line overexpressing the exercise-responsive PGC-1-alpha-b isoform is titled for increasing exercise capacity and peak oxygen uptake. The authors note their own confound: the MCK-PGC-1-alpha-a mice also had elevated PGC-1-alpha-a mRNA in the heart, and it is speculated that cardiac expression alone may have been sufficient to produce the VO2-peak effect — so the muscle-specific claim is not clean. And more is not always better: a companion PNAS paper is titled 'paradoxical effects of increased expression of PGC-1-alpha on muscle mitochondrial function and insulin-stimulated muscle glucose metabolism' (2008;105:19926). doi 10.1371/journal.pone.0028290.

It increases Mitochondrial energy.

Ventura-Clapier 2008, Cardiovasc Res 79:208 — PGC-1α is the central transcriptional regulator of mitochondrial biogenesis and respiratory function. PMID 18430751. It regulates nuclear and mitochondrially-encoded electron-transport-chain subunits. PGC-1-alpha is one of a family — PGC-1-beta and PRC share overlapping functions, so it is not a unique switch; and it is itself regulated upstream by SIRT1/SIRT3, TFAM and AMPK. PMID 18430751

It reduces Daily energy.

Lin 2002, Nature 418:797 — PGC-1α drives oxidative slow-twitch fibers and mitochondrial biogenesis, raising fatigue resistance. doi 10.1038/nature00904

It increases Body clock.

The source paper closes with the claim in its own words: we have thus identified PGC-1-alpha as a key component of the circadian oscillator that integrates the mammalian clock and energy metabolism. Not an input to the clock — part of it. The driver is itself rhythmic: PGC-1-alpha is rhythmically expressed in the liver and skeletal muscle of mice, with protein peaking in the early dark phase. The mechanism is named at the level of the transcription factor: PGC-1-alpha stimulates the expression of clock genes, notably Bmal1 and Rev-erb-alpha, through coactivation of the ROR family of orphan nuclear receptors. And loss of function was demonstrated at three independent levels: whole-animal — mice lacking PGC-1-alpha show abnormal diurnal rhythms of activity, body temperature and metabolic rate; tissue-specific — knockout in liver and skeletal muscle attenuates rhythmic expression of clock and PER1; and cell-autonomous — PGC-1-alpha-deficient fibroblasts and liver-specific knockdown indicate it is required for cell-autonomous clock function. The rhythm fails in a dish, not just in an animal. It sits alongside the SIRT1 → Circadian Rhythm link, already drawn: SIRT1 acts on the same machinery from the other side, binding and deacetylating BMAL1 and PER2. Two metabolic sensors, two routes, one oscillator. That should be searched on its own evidence rather than assumed. doi 10.1038/nature05767 · PMID 17476214
