You want it steady: supporting mood underlies motivation, relationships and quality of life.

In depth

Not happiness — stability, and the whole positive dimension of how life feels. The finding that changes how to read this pathway: feeling good is not simply the absence of feeling bad. When researchers statistically removed depression and anxiety from the analysis, the health benefits of positive wellbeing were still there — they are independent dimensions, not two ends of one line. That is why mood runs outward to sleep, inflammation, glucose, wound healing, infection resistance and survival itself, and not only inward from them.

What drives it

Supports BDNF signaling increases it.

Depression is strongly and consistently linked to low BDNF across preclinical and clinical studies, and BDNF signaling is a principal target of antidepressant action (PMC5742683). Context that matters more than the arrow: the neuroplasticity hypothesis exists because the monoamine one fell short. The same review states that although the monoamine hypothesis dominated the 1980s and early 1990s, subsequent studies showed depletion of serotonin and norepinephrine precursors did not decrease mood in healthy subjects — though it did in patients with mdd in remission. BDNF and neuroplasticity were advanced as the alternative account. serum/plasma BDNF is a peripheral proxy for a central process, and the same literature notes BDNF measurement is methodologically sensitive and influenced by sex hormones and by treatment itself. PMC5742683

Training load increases it.

Randomized trials pooled by systematic review/meta-analysis show physical exercise (aerobic, resistance and group formats) improves depressive symptoms in older adults (PMC11427357). Mechanistically consistent with the exercise→BDNF→mood route on the map. (1) BJSM 2026, PMID 41667154: 63 studies, 81 meta-analyses, 1,079 component studies, 79,551 participants — exercise reduced depression SMD -0.61 (95% CI -0.69 to -0.54) and anxiety SMD -0.47 (-0.59 to -0.36), aerobic exercise most substantial. (2) J Affect Disord 2024: 27 reviews, 190 experiments — SMD -0.67, number needed to treat 2.78. (3) JAACAP 2025: 21 reviews, 375 RCTs, >38,000 children and adolescents — SMD 0.45. Where the effect is largest, as stated: emerging adults 18-30 and postnatal women; greater reductions with group and supervised settings. For anxiety the pattern inverts — shorter duration and lower intensity worked best. Heterogeneity is substantial (I-squared 68-71 percent). PMID 41667154 · PMC11427357

The Two-Faced Peptide reduces it.

Bruchas 2010, Brain Res — dynorphin activation of kappa-opioid receptors inhibits mesolimbic dopamine, producing dysphoria and negative mood (the dysphoric component of stress). PMID 19716811

Endocannabinoid tone increases it.

Endocannabinoid/mood review 2012 — CB1-endocannabinoid signaling modulates emotional homeostasis; deficient signaling is associated with anxiety and depression, and enhancing it improves mood. PMID 23261685

Alertness chemical increases it.

Norepinephrine/mood-disorders review 1999 — locus coeruleus-norepinephrine dysregulation contributes to depression and is a target of antidepressants. PMID 10560027. A systematic umbrella review — the highest level of evidence synthesis — concluded there is NO consistent evidence of an association between serotonin and depression (Moncrieff, Cooper, Stockmann, Amendola, Hengartner & Horowitz 2022, Molecular Psychiatry, doi 10.1038/s41380-022-01661-0). The specifics: a meta-analysis of plasma serotonin showed NO relationship with depression (n=1,869) and found that lowered serotonin was associated with antidepressant use; 5-HT1A receptor-binding (largest n=561) and SERT-binding (largest n=1,845) meta-analyses showed weak and inconsistent evidence with prior antidepressant use not reliably excluded; and tryptophan depletion produced NO effect in most healthy volunteers (n=566), with weak evidence only in those with a family history (n=75). Separately, a meta-analysis of monoamine-depletion studies concluded monoamines alone ARE not sufficient to cause depression and depression has NO direct causal relation with monoamine depletion (Ruhe 2007, Mol Psychiatry 12:331). The dispute is live and both sides ARE cited here: Jauhar and colleagues replied under the title 'A leaky umbrella has little value: evidence clearly indicates the serotonin system is implicated in depression' (doi 10.1038/s41380-023-02095-y), and methodological critiques argue Moncrieff et al. mixed study types inappropriately and used unclear primary-study selection criteria (PMC9957868). One finding both sides accept: depletion did lower mood in patients with MDD in remission — the system matters in the vulnerable, not in everyone. doi 10.1038/s41380-022-01661-0 · PMID 10560027 · PMC9957868

Lowers cortisol reduces it.

Holsboer 2000, Neuropsychopharmacology 23:477 — impaired glucocorticoid-receptor feedback drives HPA hyperactivity and hypercortisolism in depression. DOI 10.1016/S0893-133X(00)00159-7. Convergent: HPA hyperactivity is described as a primary factor leading to depression under chronic stress, and cortisol is called one of the most significant biomarkers of depression. Mechanism: chronic HPA activation interacts with inflammatory pathways, generates oxidative stress, and produces structural and functional alteration of the hippocampus -- chronic cortisol exposure causes atrophy in hippocampus and amygdala. depression is explicitly multifactorial in these same reviews; HPA hyperactivity is one contributor. And the hippocampal change is reversible in many cases when stress exposure is reduced. doi 10.1016/S0893-133X(00

Brain inflammation reduces it.

Depression neuroinflammation review 2025 — cytokines drive the kynurenine pathway, depleting serotonin and dysregulating the HPA axis, lowering mood. PMID 40305200. cytokine-to-symptom correlations are moderate, not strong -- IL-6 and IL-1beta showed moderate correlation with HAM-D scores. The review states depression is increasingly linked to neuroinflammation, which is an association claim, not proof of causation. PMID 40305200

Mood (serotonin) increases it.

Delgado 2000 — the monoamine hypothesis: serotonin deficiency underlies depressed mood. PMID 10775018. Motivation increases it.

Delgado 2000 — the monoamine hypothesis: dopamine deficiency contributes to low mood and anhedonia. PMID 10775018. Quiets overexcitation reduces it.

It broke the monoamine monopoly. Elevated levels of glutamate have been found in the blood, cerebrospinal fluid and brains of patients with depression, alongside NMDA receptor subunit disturbances. The inhibition of NMDA receptor function has antidepressant effects. Ketamine is mechanistically novel, effective and rapidly acting — hours rather than the weeks every monoamine drug requires. That time course is itself the argument: whatever ketamine does, it is not waiting for monoamine receptor adaptation. The glutamate story and the neurotrophin story are one pathway, not two competing ones. The sign is subtle: drawn inhibitory because elevated tonic glutamate and NMDA overactivity associate with depression — yet the antidepressant works by causing a burst at AMPA. Tonic NMDA and phasic AMPA point opposite ways. Anything sold as raising or lowering glutamate is ignoring which receptor and which timescale. doi 10.1002/mco2.156 · PMC12124046

Gut barrier increases it.

Transmitter supply: gut microbiota influence 5-HT, dopamine, norepinephrine, glutamate and GABA and modulate tryptophan metabolism — roughly 90 percent of serotonin is made peripherally by enterochromaffin cells. Barrier and immune: changes in gut microbiota can damage the gut barrier and increase peripheral inflammatory cytokines. Neurotrophin: short-chain fatty acids such as butyrate increase BDNF, whereas dysbiosis decreases it. Germ-free animals show altered brain structure, behavior and emotional regulation. The reviewers state the limit plainly: more clinical trials ARE required to determine whether probiotics ARE effective in treating depression, and the underlying mechanisms need to be elucidated. A rich mechanism with an unproven intervention. doi 10.1002/mco2.156

Calm increases it.

Drawn as half of a balance rather than a level. A review of treatment-resistant depression lists among its core mechanisms an imbalance between glutamate and GABA that impairs synaptic plasticity — not low GABA, not high glutamate, but the ratio failing to support plasticity. The ketamine mechanism puts GABA at the center: NMDA blockade acts on inhibitory GABAergic neurons, and it is their disinhibition that releases the glutamate burst. The antidepressant target is a GABAergic cell. a review synthesis in treatment-resistant depression, a selected population (20-30 percent of MDD), which may not describe first-episode illness. No measurement here establishes a simple dose-response. doi 10.3390/jox6040134 · PMC4803609

Body clock increases it.

The causal work is as strong as anything on this pathway. Clinical: treatments directly targeting the circadian system — acute sleep deprivation, bright light therapy and sleep phase advance — can be successful in treating depression. You can treat this illness by moving the clock. Molecular and causal, two independent ways of breaking it: targeted deletion of BMAL1 in CaMK2a excitatory neurons of the medial prefrontal cortex abolished the behavioral and molecular response to sleep deprivation; and pharmacological activation of the clock repressor REV-ERB suppressed the antidepressant effects of sleep deprivation. Knock out the clock gene or drug the clock repressor — either way the antidepressant stops working. That is the clock being necessary for a treatment to act. A genuine paradox, recorded rather than smoothed: sleep deprivation and ketamine ARE both rapid-acting antidepressants, yet they exert opposing effects on mPFC circadian gene expression — SD enhances the negative clock-loop genes Per and Cry, mirroring stress, while ketamine downregulates them. Any account claiming a single clock-gene direction for depression is ignoring one of the two. PMC12916475

Sleep increases it.

Conditional, because chronic loss and one deliberate night do opposite things. Chronic: major depression involves increased sleep fragmentation, disinhibition of REM sleep and decreased slow-wave sleep, with reduced slow-wave amplitude possibly reflecting impaired plasticity, and a sleep phase delay with onset between 2AM and 6AM. Acute: one night of total sleep deprivation is a rapid-acting antidepressant, used clinically alongside light therapy and phase advance, the effect correlating with modulation of slow-wave activity. The practical reading matters: this is not licence to lose sleep. The deprivation effect is transient and is deployed under supervision alongside phase-shifting to hold the gain; chronic short sleep is on the other arm entirely. PMC12916475

What it drives

It increases Female libido.

Cyranowski 2004, Arch Gen Psychiatry (PMID 15003432) — prospective 1-year assessment of 68 women treated for recurrent major depression: controlling for partner availability and SSRI status, depressive symptoms were associated with decrements in sexual desire, sexual cognition/fantasy and arousal, while SSRI treatment tracked orgasmic difficulty only — separating the mood effect from the medication effect. Reinforced by Worsley 2017 (PMID 28499520), a community-based sample of midlife women in which moderate-to-severe depressive symptoms carried an adjusted OR of 2.69 (95% CI 1.99-3.64) for hypoactive sexual desire dysfunction. PMID 15003432. PMID 15003432

It increases Cardiac output.

Depression increases the risk of heart disease at RR 1.81, the same magnitude as its effect on overall mortality (RR 1.81). Most meta-analyses have been based on longitudinal studies using depressive symptom checklists which pick up many subthreshold depression cases - the exposure is broader than clinical diagnosis. PMC3661358

It reduces Type 2 diabetes.

Depression increases the risk of diabetes at RR 1.60 in initially disease-free subjects. Consistent with the metabolic dysregulation named as a mechanism, and with obesity at RR 1.58 in the same series. Depression and anxiety behave differently on the same endpoint, and the map shows it. PMC3661358

It increases Sexual function.

Named across six reviews of endothelial dysfunction as part of the shared mechanism set: reduced nitric oxide availability, oxidative stress, inflammatory responses, apoptosis of endothelial cells, and the emerging concept of endothelial-mesenchymal transition. doi 10.3390/jcm15041621 · PMID 33923709 · PMC11988752

It increases Men's hormone balance.

The finding that unifies all three nodes is that the same androgen does opposite things in different tissues: testosterone acts directly, and indirectly through intracellular conversion to dihydrotestosterone by 5-alpha reductase, as in prostate cells, and separately through estrogen effect when converted to estradiol by aromatase. Raise testosterone and you also raise DHT and estradiol - which is why the same intervention supports male hormonal health and can enlarge the prostate. And the clinical data shows both arms at once: in 183 men over 50 with BPH, testosterone was significantly lower in patients with severe LUTS (285.94 ng/dL) than mild (544.09 ng/dL) - so low testosterone tracks with worse urinary symptoms, while DHT drives the gland. That is not a contradiction to resolve, it is two different molecules. Bidirectionality is explicit: the relationship between hypogonadism and metabolic disorders is bidirectional, and a vicious circle between the two components has been documented. PMC6669361

It increases Stress resilience.

The cellular aging list is the thirteen hallmarks themselves, each mapped to its node and each labeled with its hallmark number - so the aging outcome now enumerates the framework rather than gesturing at it. (from the reviews of this outcome). doi 10.1002/med.21743 · PMC12680924

It reduces Calm pain signaling.

The defining feature is that the pain is real and the tissue is not damaged: central sensitization manifests as hyperalgesia, allodynia, expanded receptive fields, and impaired endogenous pain inhibition, and in osteoarthritis the same phenomenon does not correlate with radiographic findings - severe pain may persist despite stable joint structure. The mechanism is a balance failure in both directions at once: in the dorsal horn there is excessive release of glutamate, ATP, and substance P, leading to activation of NMDA receptors - while at the same time descending inhibitory mechanisms, notably those mediated by GABA, ARE weakened. Too much excitation and too little inhibition. And mast cells carry a specific hypothesis: thalamic mast cells releasing histamine, IL-1-beta, IL-6, TNF, CGRP and substance P, either stimulating thalamic nociceptive neurons directly or via microglia. Stated by the authors as a hypothesis, not an established mechanism. The reviewers ARE blunt about the state of knowledge: the pathophysiology and etiopathogenesis remain largely unknown, and there is NO definitive pathogenesis except for altered central pain pathways. doi 10.3390/biomedicines12071543 · PMC6687840

It increases Alcohol recovery.

It increases Bone health.

Depression induces bone loss through stimulation of the sympathetic nervous system; antidepressant therapy completely inhibits the decrease in bone formation and the beta-adrenergic antagonist propranolol blocks it. doi 10.1073/pnas.0604234103 · PMID 17075068 · PMC3120042

It reduces Calms body-wide inflammation.

patients with MDD required a greater intensity of the happy expression to recognize a face as happy. After three months, this negative perceptual bias was reduced in comparison to the control group. The reduction in negative perceptual bias correlated with the reduction of depressive symptoms. Verbatim from the source for this edge — PMC4454562. PMC4454562
