You want it in healthy balance: sufficient dopamine tone sustains drive, focus and positive mood.

In depth

The chemistry of wanting, not of liking. Dopamine sets how much effort a goal feels worth — which is why its loss reads as apathy long before it reads as sadness. More is not better: the same system that drives pursuit drives compulsion.

What drives it

More Than Nature Made increases it.

The present data suggest that the stimulus properties of a novel receptive female may serve to increase NAC DA transmission in a sexually satiated male rat, and this, in turn, may be related to the reinitiation of sexual behavior. doi 10.1523/JNEUROSCI.17-12-04849.1997

Training load increases it.

Exercise raises brain dopamine: habitual physical activity of ~1-2 h/day increases brain dopamine levels (systematic review, DOI 10.1186/s12970-016-0155-6), and high-intensity interval training raises dopamine D2 levels and modulates dopamine signaling. Mechanistically, dopamine rises in the ventral striatum during exercise. doi 10.1186/s12970-016-0155-6

Gut barrier increases it.

The gut microbiome supplies and shapes dopamine: microbial decarboxylases (TDC/AADC) convert tyrosine and L-DOPA into dopamine in the intestinal lumen (PMC12837447), and a microbiome-dependent gut→brain circuit elevates striatal dopamine (PMC10264927). The limit: gut-made dopamine does not cross the blood-brain barrier — the central effect runs through neural/metabolite signaling, not direct transport. Convergent: dopamine is among the neuroactive molecules generated by both eukaryotes and prokaryotes, described as an interkingdom communication system; gut microbiota both produce and consume dopamine, norepinephrine, serotonin, GABA and glutamate, which reach cellular targets through the bloodstream. Produce and consume — the microbiota is a sink as well as a source, so the net direction is not guaranteed. Shared caveats on this whole row: (1) the route to the brain is not free — gut-derived neurotransmitters reach targets via the bloodstream and must cross the blood-brain barrier; elevated plasma 5-HT can itself raise BBB permeability, temporarily letting plasma 5-HT enter the CNS, so the barrier is a variable not a constant. (2) much of this literature is association — the reviews state plainly that the association studies linking microbiota to depression did not provide evidence on aetiology. (3) The enteric nervous system hosts more than 30 neurotransmitters; any single arrow is one strand of a dense web. PMC12837447

Endocannabinoid tone increases it.

Microbiome-dependent endocannabinoid metabolites (fatty-acid amides such as OEA) made in the gut stimulate TRPV1/CB1 sensory neurons, which elevates dopamine in the ventral striatum and drives exercise motivation; blocking peripheral endocannabinoid receptors, ablating spinal afferents or blocking dopamine each abolishes the effect (Nature 2022, DOI 10.1038/s41586-022-05525-z). Mouse study — mechanism, not yet a human outcome. doi 10.1038/s41586-022-05525-z

The Two-Faced Peptide reduces it.

Dynorphin/KOR-dopamine study 2017, eLife — kappa-opioid receptors (activated by dynorphin) decrease VTA dopamine-neuron firing via GIRK channels, inhibiting mesolimbic dopamine release. PMC5389861

Sleep pressure reduces it.

Hub edge,Adenosine inhibits dopamine release, and the same inverted-U caution applies as for norepinephrine: the Dopamine → Sustained Attention link is a balance edge whose peak moves with the task, so raising dopaminergic tone by blocking adenosine is only an improvement on one side of that curve. doi 10.1016/j.neubiorev.2016.09.001

Sleep hormone reduces it.

Dopamine has an inhibitory effect on melatonin release, while melatonin also inhibits dopamine release - that is a mutual inhibition. Both arrows are drawn, and the pair forms a switch. Mutual inhibition is a switch, not a dimmer: two stable states with a threshold between them, which is the correct shape for a nocturnal/diurnal transition. doi 10.37349/ent.2023.00062

Prolactin increases it.

In the presence of prolactin, spontaneously oscillating TIDA cells depolarized, switched from phasic to tonic discharge, and exhibited broadened action potentials. 10.1523/JNEUROSCI.0129-12.2012. doi 10.1523/JNEUROSCI.0129-12.2012

Monoamine breakdown reduces it.

10.1016/j.nbd.2021.105587 — Dopamine and tyramine are metabolized by both MAO-A and MAO-B — this edge is drawn alongside the monoamine oxidase B → dopamine link because neither isoform accounts for dopamine alone. StatPearls NBK539848 — Monoamine Oxidase Inhibitors. doi 10.1016/j.nbd.2021.105587

Dopamine breakdown reduces it.

10.1186/s12881-017-0469-5 — MAO-B is sensitive to selegiline and displays a higher affinity for phenethylamine and dopamine; it is found primarily in the brain, liver and platelets. dopamine is a substrate of both isoforms, so MAO-B is the higher-affinity route, not the only one. Sci Rep 2022, PMC8742005 — Phenethylamine is a substrate of monoamine oxidase B in the paraventricular thalamic nucleus ; StatPearls NBK539848 — Monoamine Oxidase Inhibitors. doi 10.1186/s12881-017-0469-5 · PMC8742005

What it drives

It increases Mood.

Delgado 2000 — the monoamine hypothesis: dopamine deficiency contributes to low mood and anhedonia. PMID 10775018. A systematic umbrella review — the highest level of evidence synthesis — concluded there is NO consistent evidence of an association between serotonin and depression (Moncrieff, Cooper, Stockmann, Amendola, Hengartner & Horowitz 2022, Molecular Psychiatry, doi 10.1038/s41380-022-01661-0). The specifics: a meta-analysis of plasma serotonin showed NO relationship with depression (n=1,869) and found that lowered serotonin was associated with antidepressant use; 5-HT1A receptor-binding (largest n=561) and SERT-binding (largest n=1,845) meta-analyses showed weak and inconsistent evidence with prior antidepressant use not reliably excluded; and tryptophan depletion produced NO effect in most healthy volunteers (n=566), with weak evidence only in those with a family history (n=75). Separately, a meta-analysis of monoamine-depletion studies concluded monoamines alone ARE not sufficient to cause depression and depression has NO direct causal relation with monoamine depletion (Ruhe 2007, Mol Psychiatry 12:331). The dispute is live and both sides ARE cited here: Jauhar and colleagues replied under the title 'A leaky umbrella has little value: evidence clearly indicates the serotonin system is implicated in depression' (doi 10.1038/s41380-023-02095-y), and methodological critiques argue Moncrieff et al. mixed study types inappropriately and used unclear primary-study selection criteria (PMC9957868). One finding both sides accept: depletion did lower mood in patients with MDD in remission — the system matters in the vulnerable, not in everyone. doi 10.1038/s41380-022-01661-0 · PMID 10775018 · PMC9957868

It balances Sustained focus.

D1 receptor stimulation in prefrontal cortex produces an INVERTED-U dose response, whereby either too little OR too much D1R stimulation impairs spatial working memory (PMID 17277774). A meta-analysis reviewed 646 papers and included 75 to quantify it (PMC9364670). The physiology says what each end costs you: low-dose D1 blockade produces noisy firing patterns; optimal D1 stimulation gates out noise but leaves signals intact; high-dose D1 stimulation suppresses all cell firing. Too little and you cannot separate signal from background; too much and there is no signal left to separate. And the peak moves with the task, which is why NO single direction is correct: low levels of D1 sculpting may be optimal for creative thinking when a broad range of inputs ARE needed, while moderate levels may be optimal for tasks requiring precise focus. Stress pushes you off the peak in the impairing direction: high levels of norepinephrine and dopamine released during stress suppress neuronal firing and impair prefrontal function, through alpha-1 and D1 receptors respectively. And region matters: manipulating dopamine has paradoxical consequences for distinct cognitive control processes, depending on the basal or optimal level in different brain regions — prefrontal cortex and striatum do not share an optimum, so a systemic change cannot be optimal for both at once. PMID 17277774 · PMC9364670

It reduces Attention regulation.

Focus is evidenced in healthy people; this link is evidenced in patients, and the two are kept apart. The finding is about clearance, not production: patients with ADHD have depleted levels of dopamine and norepinephrine thought to be largely the result of dysfunction of their respective transporter systems, with a higher density of the dopamine transporter reported in patients — more pumps removing it from the synapse, not less being made. Three drug classes, one converging endpoint, which is the strongest argument in this literature: DAT-selective (METHYLPHENIDATE), DAT/NET-SELECTIVE (amphetamine) and NET-selective (atomoxetine) medications have one pharmacological effect in common — to elevate extracellular dopamine and norepinephrine in the prefrontal cortex. Different molecules, different transporters, same destination. And the genetics disappoint, recorded honestly: heritability is high, but studies have failed to indicate a single gene responsible, implying complex polygenic mechanisms and gene-environment interactions. The DAT1 3-prime UTR VNTR is a candidate marker for executive-function performance, not a cause. doi 10.1007/s44192-022-00030-1 · PMID 18480676 · PMC2443797

It increases Default mode network.

in patients, drug-naive, with a behavioral correlate: in 42 stimulant-drug-naive boys newly diagnosed with ADHD, performing a continuous performance task before and 90 minutes after a methylphenidate dose, METHYLPHENIDATE ROBUSTLY reversed the transient functional connectivity increase seen pre-medication to a post-medication decrease — and methylphenidate-induced reductions in individual reaction time were correlated with corresponding reductions in functional connectivity. The brain change tracked the behavior change, subject by subject. And it replicates: on-versus-off stimulant imaging showed greater suppression of ventral anterior and posterior cingulate DMN nodes on medication, and a systematic review states that all studies demonstrated that METHYLPHENIDATE treatment results in a suppression of the DMN, suggesting a potential mechanism of action. Now the inversion, and it is the most important line on this edge: in 59 healthy individuals in a randomized double-blind placebo-controlled study, METHYLPHENIDATE increased time in the default mode network state (p<0.001), alongside increased time in the dorsal attention state. The same drug suppresses the DMN in diagnosed patients and increases time in it in healthy people. That is why adhd and focus ARE separate nodes here and why neither inherits the other's evidence: the population is not a detail of the study design, it is part of the finding. A mechanism established in patients does not transfer to a healthy user, and this is the cleanest demonstration of that on the whole map. PMC5167011

It reduces Sleep hormone.

10.1371/journal.pbio.1001347 — ,Enumeration 1 gave this pathway its catecholamine arm. Enumeration 3 gave it the circadian arm. This edge is where they meet. The mechanism is a direct receptor-receptor interaction: through alpha-1B-D4 and beta-1-D4 receptor heteromers, dopamine inhibits adrenergic receptor signaling and blocks the synthesis of melatonin induced by adrenergic ligands. The adrenergic receptors are the bridge between circadian norepinephrine release and pineal melatonin output, and dopamine reaches in and closes that bridge. And the receptor is D4 — the classic ADHD candidate gene. DRD4 is the dominant dopamine receptor gene expressed in the pineal gland, at levels greater than in other tissues, with circadian expression under photoneural control. A gene long studied for its ADHD association turns out to sit at the junction of the dopamine and melatonin systems, which is exactly where this pathway needed a bridge. At some hours the interaction does not exist at all. The authors call it the first example of a circadian-controlled receptor heteromer. Direction depends on receptor family, also in the conditional table: D1-like receptors stimulate adenylate cyclase and promote aanat, the key melatonin-synthesis enzyme, while D2-like (D2, D3, D4) inhibit it. And the axis is reciprocal: melatonin signaling decreases dopamine release while dopamine inhibits pineal melatonin output — antagonistic feedback that stabilizes circadian oscillation. doi 10.1371/journal.pbio.1001347

It reduces Sleep.

The dopaminergic ventral tegmental area is listed among the wake-active brain areas alongside the locus coeruleus, dorsal raphe, basal forebrain, posterior hypothalamus and orexin cells. The GABAergic VLPO neurons inhibit both the orexin system and the ascending arousal system; the arousal nuclei inhibit the VLPO back. A flip-flop has no comfortable middle — which is why sleep onset is abrupt rather than gradual, and why these arrows are drawn inhibitory as a class. Drawn from the consensus inventory, not from a lesion. PMC7851648

It increases Sexual function.

Named across five reviews of erectile and female arousal physiology. Key contributors include reduced nitric oxide bioavailability due to endothelial oxidative stress and inflammation, diminished cavernosal smooth muscle content leading to impaired veno-occlusion, and increased arterial stiffness compounded by metabolic disorders. doi 10.21037/tau-2025-419 · PMID 19138376 · PMC11988752

It increases Female libido.

It reduces Calm pain signaling.

The defining feature is that the pain is real and the tissue is not damaged: central sensitization manifests as hyperalgesia, allodynia, expanded receptive fields, and impaired endogenous pain inhibition, and in osteoarthritis the same phenomenon does not correlate with radiographic findings - severe pain may persist despite stable joint structure. The mechanism is a balance failure in both directions at once: in the dorsal horn there is excessive release of glutamate, ATP, and substance P, leading to activation of NMDA receptors - while at the same time descending inhibitory mechanisms, notably those mediated by GABA, ARE weakened. Too much excitation and too little inhibition. And mast cells carry a specific hypothesis: thalamic mast cells releasing histamine, IL-1-beta, IL-6, TNF, CGRP and substance P, either stimulating thalamic nociceptive neurons directly or via microglia. Stated by the authors as a hypothesis, not an established mechanism. The reviewers ARE blunt about the state of knowledge: the pathophysiology and etiopathogenesis remain largely unknown, and there is NO definitive pathogenesis except for altered central pain pathways. doi 10.3390/biomedicines12071543 · PMC6687840

It increases Alcohol recovery.

Verbatim: “Simultaneous assay by PET of pre- and postsynaptic markers of dopamine neurotransmission indicated that a striatal dopamine deficit correlated with alcohol craving, which was associated with a high relapse risk”. doi 10.1176/appi.ajp.162.8.1515 · PMID 16055774

It increases The Two-Faced Peptide.

Dopamine drives dynorphin expression in striatal neurons: 'This suggests that dopamine normally stimulates expression of dynorphin and substance P in striatal neurons' (Gerfen, McGinty & Young, Dopamine differentially regulates dynorphin, substance P, and enkephalin expression in striatal neurons: in situ hybridization histochemical analysis, J Neurosci 1991;11(4):1016-1031). Scope stated before anyone asks: that paper manipulated dopamine by lesion and by chronic agonist and does not identify the D1 receptor as the mediator, so no receptor subtype is claimed here. The return arrow - dynorphin restraining dopamine release - is carried separately on the Dynorphin → Dopamine arrow.

It increases Fear extinction.

Dopaminergic prediction-error signaling drives the update that the conditioned cue no longer predicts the aversive outcome. doi 10.1111/j.1601-183X.2012.00801.x · PMC2787650
