Sleep pressure
Adenosine
Also searched as Adenosine.
The balance here is timing, and it is the sleep/wake trade itself. Adenosine accumulates through the waking day and is the pressure that makes sleep happen — raised, it improves sleep, circadian rhythm and myelin. The same signal costs you sustained attention and cholinergic tone, and can raise anxiety. This is what caffeine blocks: alertness now, borrowed against the pressure that was going to put you to sleep. You want it high at night and low in the morning — the balance is in the timing, not the amount.

Adenosine A1 signaling — the molecule that builds through waking hours to create sleep pressure and helps insulate nerve fibers.
What drives it
Pathogen tripwire increases it.

Quiets the hair-suppressing prostaglandin increases it.
The mechanistic step, drawn separately because PGD2 does not ACT on sleep neurons itself. PGD2 stimulates DP1 receptors and increases the local extracellular concentration of adenosine in the basal forebrain as a paracrine sleep-promoting molecule. And the handoff is proven by knockout: PGD2 increases the extracellular level of adenosine in wild-type but not DP-receptor knockout mice, and A2A-receptor deficiency attenuates the somnogenic effect of PGD2. Not complete HANDOFF: PGD2-induced sleep in mice is mediated by both adenosine A2A-dependent and A2A-independent systems — A2A knockouts lose the episode-duration effect but keep the episode-number effect. Which is why the Prostaglandin D2 → Sleep link is drawn as well as the Prostaglandin D2 → Adenosine A1 link, rather than routed entirely through it. doi 10.1038/aps.2016.140 · PMID 22647469
Social engagement increases it.
[18F]CPFPX PET in healthy men showed that 24 hours of sleep deprivation - an environmental/behavioural exposure - significantly increased A1 adenosine receptor binding potential across human brain regions compared with rested wakefulness. doi 10.1523/JNEUROSCI.5066-06.2007 · PMID 17329439
Curbs the main NAD⁺ consumer increases it.
The second output of CD38, and it is not a loss but a product. Extracellular NAD+ is converted to AMP by CD38 and CD203a, and AMP is then hydrolyzed by CD73 to adenosine. So the same enzyme that drains the NAD+ pool also manufactures an immunosuppressive signaling molecule from the wreckage — CD38 is a source, not only a sink. This is why CD38 sits in tumor immunology as well as in aging: adenosine acting at its receptors suppresses inflammatory and anti-tumor immune responses. this is the extracellular arm of CD38 activity and the chain is enzymatic and mechanistic — no human trial measures it, and this edge makes no claim that a botanical moves it. Found in the Chinese-language immuno-oncology literature, which foregrounds this route where the NAD+ aging reviews do not. PMC9346148 — Blocking Adenosine/A2AR Pathway for Cancer Therapy. PMC9346148

It increases Sleep.
Porkka-Heiskanen 1997, Science 276:1265 — adenosine accumulates during waking and inhibits arousal circuits, driving sleep pressure. PMID 9157887. Gain of function: pharmacologically raising adenosine in the basal forebrain, or giving A1 receptor agonists there, increases sleep. Loss of function: inactivating A1 receptors in the basal forebrain decreases EEG delta power and NREM sleep time. Mechanism: during prolonged waking, extracellular adenosine accumulates selectively in basal forebrain and cortex and restrains wake-promoting neurons via A1 — cholinergic cells in the basal forebrain, orexin cells in the lateral hypothalamus, histaminergic cells in the tuberomammillary nucleus. Caffeine is the proof running the other way: as an A1/A2A antagonist it removes adenosine's brake on orexin neurons and raises alertness. Two receptors, different circuits: A1 acts presynaptically and in the VLPO/median preoptic sleep circuits; A2A is Golf-coupled, enriched in striatum and nucleus accumbens, and works through the indirect basal-ganglia pathway. The basal forebrain contains A1 but not A2A receptors. And one arm is disputed at the receptor level: a 2025 study reports it saw neither inhibition of firing nor modulation of inhibitory potentials by adenosine in rat histaminergic neurons, and found NO A1 receptor in any histaminergic single-cell transcriptome database — concluding most histaminergic neurons ARE not inhibited by adenosine (PMC11711771). The cholinergic and orexinergic arms are secure; the histaminergic arm is contested. Species: rat and mouse. PMID 9157887 · PMC11711771
It reduces Sustained focus.
Adenosine is not a peripheral addition — it sits upstream of all four other drivers of focus. The proof is the most widely self-administered drug on earth: caffeine exerts its effects by blocking adenosine receptors — the only likely mechanism at physiological concentrations — and following low to moderate doses, alertness, vigilance, attention and reaction time improve. Every cup of coffee is an uncontrolled experiment confirming this link. Why it is a hub and not a detail: adenosine inhibits the release of many neurotransmitters in the central nervous system — A1 receptor agonists inhibit release of glutamate, serotonin, acetylcholine, norepinephrine and dopamine. Three of those five are the other drivers of focus. Adenosine does not sit beside them; it sits on them. The same literature states the limit on caffeine: caffeine increases energy metabolism throughout the brain but simultaneously decreases cerebral blood flow, inducing a relative brain HYPOPERFUSION; and effects are less consistent on memory and higher-order executive function such as judgment and decision making than on simple vigilance. It sharpens the watchman, not the strategist. doi 10.1016/j.neubiorev.2016.09.001

It reduces Motivation.
Hub edge,Adenosine inhibits dopamine release, and the same inverted-U caution applies as for norepinephrine: the Dopamine → Sustained Attention link is a balance edge whose peak moves with the task, so raising dopaminergic tone by blocking adenosine is only an improvement on one side of that curve. doi 10.1016/j.neubiorev.2016.09.001
It reduces Calm under stress.
10.1038/sj.npp.1301001 — Adenosine is the brake caffeine blocks; blocking enough of it does not simply produce alertness, it produces anxiety. The alertness effect on focus runs through exactly this mechanism, and the anxiogenic effect is the same pharmacology past its useful point. The effect is established enough to be used as an experimental tool: a controlled trial is titled acute exercise reduces caffeine-induced anxiogenesis — caffeine-induced anxiety is the independent variable, reliable enough that researchers induce it deliberately to test something else against it. this is about antagonism at dose, not about endogenous adenosine tone. There is no evidence here that low natural adenosine causes anxiety (Youngstedt, O'Connor & Crabbe 1998, Med Sci Sports Exerc 30:740). doi 10.1038/sj.npp.1301001
It increases Body clock.
The human trial is unusually well built: double-blind, placebo-controlled, within-subject, running about 49 days. A caffeine dose equivalent to a double espresso taken 3 hours before habitual bedtime induced a roughly 40-minute phase delay of the circadian melatonin rhythm. And the comparison is the line worth remembering: that delay was nearly half the magnitude of the phase delay produced by three hours of evening bright light at about 3000 LUX. One evening coffee does half of what a light box does — in the wrong direction. The receptor is named and isolated in vitro: chronic caffeine lengthens the circadian period of molecular oscillations in human U2OS cells carrying bmal1:luciferase, primarily through an adenosine receptor / cyclic-AMP-dependent mechanism, with period lengthening reproduced by the adenosine receptor antagonist CGS-15943 alone. Replicated in mice with a cleaner tool: a selective adenosine A1 receptor antagonist reproduced caffeine’s enhancement of photic phase-delays. Sleep regulation is classically taught as two independent processes — homeostatic and circadian. This edge shows they share a node. The molecule that tracks how long you have been awake also sets the clock that decides when you should be. central versus peripheral again: acute caffeine shifts clock genes in the liver and kidney but not in the SCN. doi 10.1126/scitranslmed.aac5125
