The body's danger-detection signals — molecular fingerprints of pathogens and damaged cells that trip the innate-immune alarm.
What it drives
It increases Sleep pressure.
Adenosine-in-inflammation review 2015 — adenosine accumulates in inflamed/injured tissue as CD39/CD73 dephosphorylate extracellular ATP (a damp) to adenosine, an anti-inflammatory brake. PMC4496696
It increases Innate immune sensor.
How the body first notices — and why noticing is not harmless. 'release of PAMPs and DAMPs activates innate immune pathways' (Heart Failure Reviews 2026, doi 10.1007/s10741-026-10654-y), which run through the same master inflammatory switch that drives the inflammatory response. ⇒ the alarm that detects the virus is the same alarm that, left on, damages the muscle. That is why the second phase is the dangerous one. doi 10.1007/s10741-026-10654-y
It increases Gum pockets and attachment.
The gum lesion begins as a bacterial-pattern signal: lipopolysaccharide from Porphyromonas gingivalis — the principal periodontal pathogen — strongly induces IL-1β, IL-8 and TNF-α in human gingival epithelial cells, and siRNA knockdown of the mediating step reduces all three (PMC5608282). Periodontitis is a chronic inflammatory disease in which the inflammatory response, not the bacterium alone, does the structural damage. PMC5608282
It increases Interferon response.
PAMPs are recognized by host pattern-recognition receptors, initiating the interferon system as a core innate defense against viral infection (PMC11172372). Provenance: the identifier previously cited here (PMC7255347) could not be confirmed to resolve and has been replaced with sources actually read. PMC11172372
It increases Gut-brain signal.
And the random test turned up a necessity demonstration that bears directly on probiotic claims: after VAGOTOMY, the beneficial effects of Lactobacillus and Bifidobacterium on cognitive function ARE abolished, and the positive effects of probiotic ingestion on cognition ARE only evident when the vagus nerve is intact. The nerve is not one route among several for this effect — it is required. shown by lesion in animals; the vagus is stated to be the primary sensory pathway for visceral information reaching the CNS despite there being NO direct contact between gut microbiota and vagal afferents — sensing is via diffusing metabolites and enteroendocrine intermediaries. SCFA specificity: butyrate acts directly on afferent terminals while oleate acts through a CCK-mediated mechanism — different metabolites, different routes. PMC12293201
It reduces Liver.
The anatomy is the mechanism: the portal vein gathers blood from the small and large intestine, spleen, and pancreas and directs it into the liver, so as intestinal venous blood drains into the portal vein, the liver becomes the first organ to receive gut microbiota-derived products and metabolites. And the intervention evidence is unusually direct: exogenous conjugated bile acids increase bile acid secretion and reduce bacterial overgrowth, endotoxemia and translocation; FXR agonists RECONSTITUTE microbiota composition, restore epithelial and vascular intestinal barrier function and decrease bacterial translocation and endotoxemia in experimental cirrhosis. Species: experimental cirrhosis is the authors’ phrase - animal models. doi 10.1038/s41522-022-00352-6 · PMC11265038
It reduces Digestion.
Named across six gut-liver axis reviews. The main alteration is dysbiosis - A decrease in beneficial bacteria, an increase in potentially harmful microbes, and a loss of overall microbial diversity - with intestinal barrier integrity determining the levels of microbial metabolites, ENDOTOXINS and ammonia that reach the liver and systemic circulation. doi 10.1038/s41522-022-00352-6 · PMC11265038
It increases Clears blocked nose & sinuses.
This is the same axis as eczema, and it splits the same way: type 2 endotypes are IL-4/IL-5/IL-13-driven and eosinophilic, while non-type-2 endotypes ARE driven by increased Th1 and Th17 responses, resulting in elevated neutrophil levels. Two endotypes, two cell types, two nodes. And the intervention evidence is unusually direct: dupilumab (IL-4Ralpha inhibitor) reduces polyp burden and restores smell, while MEPOLIZUMAB (anti-IL-5) and OMALIZUMAB (anti-IgE) address specific endotypes - three biologics, three targets, three endotype-matched responses. PMC11059040

It increases Urate clearance.
The architecture is unusually clean and worth stating: gout has two separable stages. Stage one is metabolic - urate underexcretion through renal and gut mechanisms is the major mechanism for hyperuricemia in most people, via URAT1, GLUT9 and ABCG2. Stage two is immunological - MSU crystals function as damage-associated molecular patterns which, upon phagocytosis by macrophages, activate the NLRP3 inflammasome through a two-step process: priming via NF-kB signaling and activation via lysosomal rupture, potassium efflux, and mitochondrial ROS. And the two stages come apart in people: not all individuals with hyperuricemia show deposition of MSU crystals OR develop gout. Local factors decide - reduced joint temperature, low pH, high sodium concentration, and preexisting joint damage. A urate number is not a diagnosis. The treatment target is explicit: long-term lowering of serum urate to less than 360 MICROMOL/L leads to crystal dissolution and ultimately to suppression of flares. PMID 39712248 · PMC8268734
It reduces Urinary health.
The genitalia and lower urinary tract share common estrogen receptor function. The receptor change is the estrogen receptors alpha and beta ARE present in the vagina, vestibule, urethra and bladder TRIGONE - while both receptors can be found in premenopausal women, only estrogen-beta receptors can be detected in postmenopausal women. The tissue does not merely see less hormone; it loses a receptor subtype. And a pH loop with a clinical endpoint: estrogen maintains vaginal glycogen, lactobacilli convert it to lactic acid holding pH at 3.5-5.0; loss of estrogen is responsible for the reduction of Lactobacillus, changing the vaginal fluid to an alkaline pH of 5.0 OR above, which promotes overgrowth of gram-negative rod fecal flora, inducing vaginal infection and UTI. Scale and underdiagnosis: GSM affects 40-54 percent of postmenopausal women and 15 percent of premenopausal women, and around 60 percent of these women have never had a diagnosis of GSM before. doi 10.3389/frph.2021.779398 · PMC10692865
It reduces Resists infection.
The headline on this pathway is a disappointment and it must stay there: the lack of effect of prophylactic vitamin C supplementation on the incidence of common cold in normal populations throws doubt on the utility of this wide practice. Regular supplementation decreases cold duration by 8 percent (3-12) and severity by 15 percent (CI 9-21) - but does not reduce how often you get one. With one striking exception that defines WHO it works for: a subgroup of six studies of marathon runners, skiers, and soldiers exposed to significant cold AND/OR physical stress experienced, on average, 50 percent reduction in common cold incidence. Extreme physical stress is the condition under which it prevents. (PMC10712193; PMC7356429; PMC1160577). REFUTED ON A SECOND READING. This link was shown by linked steps until a source was opened that does not support it: “Innate immunity is the first defense mechanism employed against invading bacterial pathogens through the recognition of conserved molecular patterns on bacteria by pattern recognition receptors (PRRs), especially the toll-like receptors (TLRs).” — SOCS Proteins as Regulators of Inflammatory Responses Induced by Bacterial Infections: A Review. Frontiers in Microbiology, 2017, 8. doi 10.3389/fmicb.2017.02431 · PMC10712193
It reduces Gum health.
The mechanism is a self-feeding loop and the fuel is your own tissue: inflammation and dysbiosis positively reinforce each other because inflammatory tissue breakdown products ARE used as nutrients by the dysbiotic microbiota - specifically collagen peptides and heme-containing compounds. The bacteria eat the damage the immune response causes, which causes more damage. And it is not an infection in the ordinary sense: periodontitis is caused mainly by dysbiosis of the bacterial community, rather than an infection by individual bacteria. Keystone pathogens, whose colonization is facilitated by accessory pathogens, initially subvert the host response, after which PATHOBIONTS - commensals turned pathogenic - over-activate inflammation. A keystone pathogen works at low abundance by changing the community, not by dominating it. The tissue-destruction route is specific: pathogens induce epithelial-mesenchymal transition via TGF-beta/Smad, Wnt/beta-catenin and Notch, causing loss of E-cadherin, junctional epithelial instability, pocket formation and attachment loss. doi 10.3390/jmp7010011 · PMID 25534621 · PMC5646588
It reduces Wound healing.
The critical point is a switch, not a level: the transition from an initial inflammatory phase to a proliferative phase is a critical regulatory point that determines wound healing outcomes, and macrophages drive it by switching from pro-inflammatory M1 to anti-inflammatory M2. When proinflammatory M1 macrophages persist without transitioning to anti-inflammatory phenotypes, this contributes to impaired wound healing. Which means anti-inflammatory is not uniformly good here: an ordered and well-controlled inflammatory phase is essential for the normal progression of healing. Suppressing the M1 phase does not accelerate repair - it stalls it. The requirement is timely transition, not less inflammation. Four phases, sequence-critical: hemostasis, inflammation, proliferation, remodeling - for a wound to heal successfully, these four phases must occur in an appropriate sequence and time frame. And an unexpected mechanism worth keeping: disruption of the epithelium generates a directional electrical field, which helps orient keratinocytes for directed migration. doi 10.3390/cells14231850 · PMC8432991
