Innate immune sensor
TLR4 signaling↓

Acute vs chronic: TLR4 is your endotoxin alarm and you would not survive without it. Sharp acute defense protects you; chronic activation smolders. You want it vigilant and brief.
In depth
A frontline pattern-recognition receptor that detects microbial and danger signals and calls the inflammatory response.
What drives it
Pathogen tripwire increases it.
How the body first notices — and why noticing is not harmless. 'release of PAMPs and DAMPs activates innate immune pathways' (Heart Failure Reviews 2026, doi 10.1007/s10741-026-10654-y), which run through the same master inflammatory switch that drives the inflammatory response. ⇒ the alarm that detects the virus is the same alarm that, left on, damages the muscle. That is why the second phase is the dangerous one. doi 10.1007/s10741-026-10654-y
Allergic signaling reduces it.
IL-13 dampens airway epithelial innate immunity by inducing IRAK-M, a negative regulator of TLR signaling (J Allergy Clin Immunol S0091-6749(11)01740-4), and IL-4/IL-13 downregulate TLR4 expression and function in human intestinal epithelial cells (J Immunol 2006;176:5805-5814). This closes a negative-feedback loop: TLR4 drives Th2 cytokines including IL-13, and IL-13 in turn restrains TLR4. Human epithelial cells. Verified at source before addition -- gates 2 and 3 cleared.
Energy sensing reduces it.
AMPK suppressed production of nuclear factor (NF)-κB-dependent cytokines in TLR4-stimulated cells. Treatment of mice with 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside (AICAR) or metformin, two…. 10.2119/molmed.2011.00401. doi 10.2119/molmed.2011.00401
Social bonding hormone reduces it.
OT binds to the OT receptor (OTR) and reduces PTP1B levels... Furthermore, OTR signaling inhibits the p-eIF-2α–ATF4 pathway, thereby suppressing the production of TNF-α, IL-6, and IL-1β. 10.3390/cells8060527. doi 10.3390/cells8060527
Galectin-3 increases it.
Galectin-3 acts on innate immunity directly, not only through fibroblasts: it 'was reported as an endogenous paracrine ligand and activator of TLR-4 inducing an inflammatory response (Burguillos et al., 2015)', and the galectin-3/TLR-4/nf-kB/TNF-alpha axis was the mechanism through which modified citrus pectin ameliorated methotrexate-induced liver and lung toxicity (doi 10.3389/fphar.2025.1528978). Rodent toxicity model. A direct contradiction is recorded here rather than omitted: the same protein restrains the same receptor in other settings. Verbatim: 'galectin-3 has been recently shown to restrain the inflammatory response that involves the Toll-like receptor- (TLR-) 2 and 4 and the proinflammatory cytokines interleukin-1-beta and TNF-alpha in macrophages, an effect that contributes to delay the course of Wallerian degeneration in peripheral nerves, and to reduce the susceptibility to endotoxin shock.' These are different endpoints and different models (chemical liver/lung toxicity versus nerve degeneration and endotoxin challenge), so neither refutes the other — but the edge as drawn is the injury-model direction, and the opposite finding exists. doi 10.3389/fphar.2025.1528978
What it drives
It increases Ceramide load.
PMID 21490391 — Holland/Summers 2011, j Clin Invest (jci.org/articles/view/57144) - saturated fatty acids and LPS, acting through TLR4, switch on the whole ceramide production line at once. Verbatim: LPS and saturated fatty acids, via TLR4, increase levels of transcripts encoding virtually all of the enzymes involved in de novo ceramide synthesis, including spt subunits, CerS isoforms, and Des1. This is the master upstream control: one receptor governs transcription of the entire enzyme set. Shown in rodent and cell transcriptional work. PMID 21490391
It increases Cytokine signaling.
10.1186/s10020-024-00845-4 — TLR4/type-I-IFN study 2003, Nat Immunol — TLR4/LPS induces IFN-β, which acts through the JAK-family kinase Tyk2 and STAT signaling; Tyk2/IFN-β are essential effectors of LPS responses. doi 10.1186/s10020-024-00845-4
It increases Calms histamine response.
Varadaradjalou 2003, Eur J Immunol — human mast cells express TLR4; the TLR4 agonist LPS induces mast-cell release of TNF-α and cytokines (IL-5, IL-10, IL-13). PMID 12672055 Verbatim: “We also established that the stimulation of CBMC with LPS or with PGN is mediated through interactions with TLR4 or with TLR2, respectively”. doi 10.1002/eji.200323830 · PMID 12672055

It reduces Immune balance.
TLR4 is where the innate system decides what the adaptive system will do. Its signal transduction and NF-κB transactivation induce IL-12p40 and the costimulatory molecules CD80 and CD86 — the molecules that link innate to adaptive immunity by licensing antigen-specific responses in naive T cells (PMC3189122; TLR network review PMC1810480). Calibrated immunity therefore begins at the receptor, not at the cytokine. The innate-to-adaptive licensing claim is supported. demonstrated in the airway compartment in mice; the IL-12p40/CD80/CD86 arm is BM-derived, not stromal. PMC3189122
It increases Eases allergy load.
Endotoxin in inhaled air acts through TLR4: signaling in pulmonary stromal cells is required for early chemokine production, neutrophil and dendritic-cell recruitment to the lung and direct bronchoconstriction, and TLR4 signaling on epithelial cells induces a Th2-polarising response through Th2-inducing dendritic cells (PMC3189122). Inhaled endotoxin is thus a driver of airway inflammation in asthma, not merely a marker of it. MOUSE/stromal-cell work. PMC3189122
It increases Allergic signaling.
The innate receptor sets the Th2 tone: TLR4 signaling on pulmonary epithelial cells induces a Th2-polarising response through Th2-inducing dendritic cells, and the airway epithelium drives allergic disease by producing pro-Th2 cytokines including IL-4 and IL-13 (PMC3189122). An UNMAPPED negative feedback runs the other way: IL-13 dampens airway-epithelial innate immunity by inducing IRAK-M, and IL-4/IL-13 downregulate TLR4 expression in human epithelium (JACI S0091-6749(11)01740-4; J Immunol 176:5805). It increases Gut-brain signal.
TLR4 receptors ARE expressed on vagal afferent fibers themselves (Goehler 1999), so the nerve carries its own bacterial sensors rather than relying on relayed signals — and LPS activates vagal afferents directly at the level of the NODOSE ganglia (Hosoi 2005). The vagus is not the only route, and the source says so: subdiaphragmatic VAGOTOMY does not completely block the brain-mediated behavioral and neural effects of peripheral LPS or IL-1-beta (Schwartz 1997) — humoral routes persist. doi 10.3389/fnins.2018.00049 · PMC12293201
It increases Arterial plaque.
Reviews describe upstream signals converging on core inflammatory hubs, such as the toll-like receptor 4 / nuclear factor-kappa-B pathway and the NLRP3 inflammasome. The proximate trigger is specified: OX-LDL binds to LOX-1 on endothelial cells, activating the NF-kB signaling pathway and subsequently inducing the expression of adhesion molecules. PMC5666716
