Adenosine A1 receptor
Autocrine antilipolytic signaling
Also searched as Autocrine antilipolytic signaling.
You want it not dominating. It is the predominant adenosine receptor in white fat and it suppresses release through the inhibitory arm of the same switch adrenaline uses, validated in both mice and people. It reverses by depot: in brown fat a different adenosine receptor predominates that is coupled the other way and promotes release and heat. Same molecule, opposite instruction, depending on which tissue hears it.
In depth
A receptor for a signal the fat cell makes itself — and which tells it to stop releasing.
What drives it
Sleep pressure increases it.
Adenosine is the endogenous ligand that occupies and activates the A1 adenosine receptor. PMID 19012505
What it drives
It reduces Releases stored fat.
'In white fat, adenosine receptor 1 (ADORA1) is predominantly expressed and attenuates lipolysis by Gi-protein coupled pathway, once activated by adenosine, which has been validated in both mice and humans' (doi 10.3389/fphys.2022.826314). The source states its own gap: 'lipolysis in the WAT of ADORA1-deficient mice did not increase, possibly owing to pleiotropic functions... therefore a tissue specific knockout of ADORA1 is required for in vivo validation.'. doi 10.3389/fphys.2022.826314
It reduces The cell's go-signal.
The receptor is Gi-coupled: 'Gi signaling mainly inhibits the cAMP dependent pathway by inhibiting adenylate cyclase activity, decreasing the production of cAMP from ATP' (doi 10.3389/fphys.2022.826314). And it reverses by depot: 'In human and murine brown fat, ADORA2A is the most abundantly expressed adenosine receptor, which is coupled with a Gs protein... adenosine exerted pro-lipolytic action and thermogenic effects.'. doi 10.3389/fphys.2022.826314
