The cell's go-signal
Cyclic AMP (cAMP)
Also searched as Cyclic AMP (cAMP).
This is the bottleneck, and that is what makes it useful. A nerve fires, a receptor is occupied, a channel opens — none of it reaches the mitochondria directly. All of it converges on one molecule made inside the cell by adenylate cyclase, which activates protein kinase a, which releases stored fat and switches on the heat protein. Cold does it. Noradrenaline does it. The beta-3 receptor does it. And several plants reach the same molecule without needing the receptor at all. Three different ways in, and a blend can hold all three. Forskolin activates adenylate cyclase directly, so it makes cyclic AMP without a hormone or a receptor involved. Black galanga works at the opposite end — its principal polymethoxyflavone inhibits the phosphodiesterase enzyme that destroys cyclic AMP, so whatever signal arrives lasts longer. Caffeine and the other methylxanthines do the same thing, and when the question was tested directly it was phosphodiesterase inhibition rather than adenosine blockade that explained the effect at ordinary dietary doses. Green tea catechins act one step earlier, slowing the enzyme that clears noradrenaline from the synapse. What has to be said against it. The catechin step fails in people: a study measuring the enzyme directly in human volunteers found its activity not impaired by high doses of EGCG in vivo, so that link is in-vitro only. The dose specification matters more than the ingredient name — black galanga's phosphodiesterase evidence rests on a 4.07% dimethoxyflavone standardization, and a commercial product at 2.5% showed no effect on resting expenditure once its caffeine was removed. And two separate genes, COMT and TRPV1, change how much of this a given person gets.
In depth
Cyclic AMP — the single message inside the cell that every fat-burning signal, whatever door it came through, has to become.
What drives it
Beta-3 receptor signaling increases it.
The betaAR stimulation elevates intracellular cAMP production through the guanine nucleotide-binding alpha stimulating protein (Gnas)... Elevated cAMP levels activate protein kinase A (PKA). PMID 1435297
Alertness chemical increases it.
Sympathetic inputs regulate adaptive thermogenesis in brown adipose tissue through cAMP... specific ablation of cAMP production in brown adipocytes led to reduced lipolysis, mitochondrial biogenesis,…. PMID 1435297
Cold exposure response increases it.
stimulation by cold or beta3AR agonists is associated with increased energy expenditure and enhanced disposal of circulating glucose and fatty acids. PMID 1435297
Messenger-clearing enzyme reduces it.
How insulin actually works — it deletes the message. 'Insulin signaling activates phosphodiesterase 3B (PDE3B) through phosphorylation at Ser273 via the PKB... Activated PDE3B hydrolyzes cAMP to 5-AMP thereby reducing the cAMP levels and, therefore, PKA activity' (doi 10.3389/fphys.2022.826314). The source does not overstate it: 'several studies have challenged this and suggested that while PDE3B is essential under certain conditions, its function alone cannot fully explain the antilipolytic action.'. doi 10.3389/fphys.2022.826314
Adenosine A1 receptor reduces it.
The receptor is Gi-coupled: 'Gi signaling mainly inhibits the cAMP dependent pathway by inhibiting adenylate cyclase activity, decreasing the production of cAMP from ATP' (doi 10.3389/fphys.2022.826314). And it reverses by depot: 'In human and murine brown fat, ADORA2A is the most abundantly expressed adenosine receptor, which is coupled with a Gs protein... adenosine exerted pro-lipolytic action and thermogenic effects.'. doi 10.3389/fphys.2022.826314
The Lactate Veto reduces it.
'the mechanism underlying the insulin-induced reduction in adipocyte cAMP levels is more complex and involves insulin-stimulated release of lactate from adipocytes to inhibit cAMP formation via GPR81. Gi-coupled receptor GPR81 is specially expressed in adipocytes and serves as a lactate receptor'. doi 10.3389/fphys.2022.826314
The Droplet Gatekeeper reduces it.
'loss of galectin12 increases the efficiency of cAMP signaling at the lipid droplet, perhaps by altering the recruitment OR function of a phosphodiesterase, and consequentially promoting PKA-dependent recruitment of lipases to lipid droplets' (doi 10.3389/fphys.2022.826314). The source states its own gap: 'the precise mechanism of lipolysis control by galectin12 is not clear.' Its partner VPS13C is required for its stability, and deleting VPS13C also raises release. doi 10.3389/fphys.2022.826314
Short-chain fatty acid receptor reduces it.
The fiber paradox, named. 'ligands of Gi-coupled receptors, such as succinate, short-chain fatty acids, medium- and long-chain fatty acids, ketone body beta-hydroxybutyrate, nicotinic acid and neuropeptide Y, can also conduct anti-lipolytic effects via binding to their respective Gi-coupled receptors GPR91, GPR43, GPR120, GPR109A and Y1R to inhibit cAMP formation'. doi 10.3389/fphys.2022.826314
Niacin receptor reduces it.
A supplement that works against this product. 'nicotinic acid and neuropeptide y can also conduct anti-lipolytic effects via binding to their respective Gi-coupled receptors... GPR109A and Y1R to inhibit cAMP formation' (doi 10.3389/fphys.2022.826314). Nicotinic acid is sold as a supplement and used clinically for blood lipids — a plausible companion to a fat-release product that opposes it here. doi 10.3389/fphys.2022.826314
Fatty acid receptor reduces it.
The outside half of the self-inhibition loop. 'MEDIUM- and long-chain fatty acids (LCFAs)... can also conduct anti-lipolytic effects via binding to their respective Gi-coupled receptors GPR91, GPR43, GPR120, GPR109A and Y1R to inhibit cAMP formation' (doi 10.3389/fphys.2022.826314). The fat leaving the cell signals from outside for the cell to stop. doi 10.3389/fphys.2022.826314
The Metabolite Veto reduces it.
'ligands of Gi-coupled receptors, such as succinate, short-chain fatty acids... GPR91' (doi 10.3389/fphys.2022.826314). Included for completeness of the veto family, not as a target — its specific role in this braking is less studied than lactate or the fatty-acid receptors, and nothing in this formula addresses it. doi 10.3389/fphys.2022.826314
Bone-building receptor increases it.
10.1186/s40510-025-00555-5 — one receptor, two opposing arms. 'Binding of parathyroid hormone (PTH) to parathyroid hormone receptor 1 (PTHR1) activates Gs- and Gq-coupled signaling. Whereas Gs activates the AC-cAMP-PKA pathway'. doi 10.1186/s40510-025-00555-5 · PMID 29514884
The Stress Veto reduces it.
The sixth and least-known veto on fat release, and the one stress opens. Alongside lactate, short-chain fatty acids, long-chain fatty acids, succinate and nicotinic acid, neuropeptide y acts through its own Gi-coupled receptor on the fat cell: 'many of lipolysis' regulators act on the pro-lipolytic PKA pathway, which is typically activated by catecholamines and can be inhibited by different factors upon binding to their respective Gi-coupled receptors' (doi 10.3389/fphys.2022.826314). Why it matters more than the other five for a real person: this ligand rises with stress, and it is released alongside the very epinephrine that is supposed to be driving release. The accelerator and this brake arrive together. doi 10.3389/fphys.2022.826314
cAMP-making enzyme increases it.
10.3389/fonc.2021.738300 — 10.1371/journal.pone.0100252 — from a published diagram titled 'control DE LA LIPÓLISIS EN EL adipocito HUMANO' — a human-specific control map. Its legend enumerates the whole system: 'Receptores β- y α2A-Adrenérgicos (RA); Adenilato Ciclasa (AC); proteínas G heterotriméricas (Gs y Gi); Proteína Cinasa A (PKA); Lipasa Sensible a Hormona (LSH); Receptor de Insulina (IRS-1/2); Proteína Cinasa B (PKB); Cinasa (PI3-K); Fosfodiesterasa tipo 3B (PDE-3B); GUANILATO Ciclasa (GC); Receptor de Péptidos Natriuréticos (NPR-A); Proteína Cinasa G (PKG); Lipasa de Monoglicéridos (LMG); NEUROPÉPTIDO Y (NPY) Y PÉPTIDO YY (PYY); Proteínas que enlazan lípidos en adipocito (ALBP); Ácidos Grasos No Esterificados (AGNE)' (Revista de Educación Bioquímica, UNAM — 'Control de la lipólisis en el adipocito HUMANO' (ESPAÑOL)). The factory itself, which this MAP had been skipping over. Every stimulating hormone and every inhibiting one converges here: the stimulators switch this enzyme on through one class of G-protein, the six vetoes switch it off through another. It is the single point where the whole argument is settled — and it is the exact enzyme one driver in this formula activates directly, bypassing every receptor above it. doi 10.3389/fonc.2021.738300

The canonical cascade, stated identically by four independent groups. Verbatim: 'When catecholamines bind to these receptors, stimulatory Gs proteins activate adenylate cyclase, causing a rise in cAMP levels and elevated activity of cAMP-dependent protein kinase-A (PKA)... PKA-mediated phosphorylation of target proteins, including lipolytic enzymes and lipid droplet-associated proteins, induces an increased release of FAs and glycerol from adipose tissue up to 100-fold.' the same pathway brakes itself: 'Through the same PKB-dependent pathway beta-adrenergic receptor signaling, via cAMP and PI3K-alpha, is anti-lipolytic and inhibits its own stimulation of lipolysis by 50%'.
It increases Lipolysis signal.
10.1017/S0029665109990279 — 'A rise in cAMP activates protein kinase (PK) A, which phosphorylates hormone-sensitive lipase (HSL) and LD-coating protein perilipin (PLIN) to stimulate lipolysis' (Proc Nutr Soc, Regulation of adipose tissue lipolysis revisited). The step is definitional rather than contested. doi 10.1017/S0029665109990279
It increases Fat-releasing enzyme (HSL).
10.1017/S0029665109990279 — 'A rise in cAMP activates protein kinase (PK) A, which phosphorylates hormone-sensitive lipase (HSL) and LD-coating protein perilipin (PLIN) to stimulate lipolysis' (Proc Nutr Soc, Regulation of adipose tissue lipolysis revisited). Drawn as the messenger-to-lipase route that runs through the kinase. doi 10.1017/S0029665109990279
It reduces Reduces clotting.
The platelet brake. Verbatim: CME-1 'inhibits human platelet activation by activating adenylate cyclase/cyclic AMP' (PMID 25294588), with a pharmacological counterfactual - SQ22536, an adenylate cyclase inhibitor, reversed both the cAMP rise and the aggregation inhibition. Five unrelated molecules on this blend converge here. cAMP is a general second messenger; the edge claims only that raising platelet cAMP lowers platelet activation. PMID 25294588
