Fatty-acid oxidation

The Fat-Burning Switch

2 blends on this site modulate it.

You want it engaged: activating PPARα is how the fasted, exercised or botanically-primed liver turns fat into fuel.

In depth

The nuclear receptor that commands the liver's fatty-acid-oxidation program — the master switch for burning fat rather than storing it.

What drives it

Fasting response increases it.

Kersten 1999, J Clin Invest 103:1489 — PPARα mediates the adaptive response to fasting; fasting activates PPARα to switch the liver to fat oxidation and ketogenesis. PMID 10359558. Verified, and the mechanism is de-repression rather than direct activation: in the fed state the insulin-dependent PI3K pathway activates mTORC1, which drives nuclear localisation of NCoR1 -- the PPAR-alpha COREPRESSOR -- inhibiting PPAR-alpha activity and ketogenesis. Fasting lowers mTORC1 and S6 kinase 2, pushing NCoR1 to the cytoplasm and releasing PPAR-alpha (Cells 2020;9:1708). Fasting does not switch PPAR-alpha on; it removes the brake. PMID 10359558

Builds mitochondria increases it.

Vega 2000, Mol Cell Biol 20:1868 — the coactivator PGC-1α cooperates with (coactivates) PPARα to drive transcription of nuclear-encoded mitochondrial fatty-acid-oxidation genes.

Lipid phosphatase increases it.

Lipin activates the PPARalpha/PGC-1alpha regulatory pathway via multiple mechanisms including cooperation with PPARalpha and PGC-1alpha as well as transcriptional activation of the PPARalpha gene... the expression of lipin 1 is induced by PGC-1alpha. doi 10.1016/j.cmet.2006.08.005 · PMID 16950137

Acetaldehyde load reduces it.

Verbatim (pmc7530314): 'Ethanol inhibits DNA-binding and transcriptional activity of peroxisome proliferator-activated receptor alpha (PPAR-alpha), with resulting downregulation of fatty acid oxidation. this inhibition occurs directly through the effects of acetaldehyde or indirectly from CYP2E1-generated oxidative stress, adenosine, downregulation of adiponectin, and zinc deficiency.' PPARalpha was carried by zero blends before this build.

What it drives

It increases Fasting metabolic hormone.

Inagaki 2007, Cell Metab 5:415 — FGF21 is induced directly by PPARα in the liver during fasting, the endocrine mediator of the adaptive starvation response. Knockout-anchored in both directions. The founding paper is titled 'endocrine regulation of the fasting response by PPAR-alpha-mediated induction of fibroblast growth factor 21' (Inagaki 2007), and PPAR-alpha is required for FGF21 induction in response to fasting and in the early post-prandial phase. PPAR-alpha-null mice show the mirror phenotype — impaired ketogenesis and hepatic steatosis on fasting — and giving FGF21 to those mice raised beta-hydroxybutyrate about threefold, restoring roughly 30 percent of wild-type levels, and significantly reduced hepatic triglyceride accumulation. FGF21 partially rescues the PPAR-alpha-null fasting phenotype. But induced is not required: a paper is titled 'fibroblast growth factor 21-null mice do not exhibit an impaired response to fasting' — despite strong fasting induction of FGF21 in wild-types, FGF21-null mice showed only limited impairments (mild blunting of gluconeogenic transcription, hepatic triglyceride accumulation, reduced fatty-acid-oxidation gene expression). The hormone is a modulator of the fasting response, not its linchpin.

It increases Fat burning.

Kersten 1999, J Clin Invest 103:1489 — PPARα is the master regulator of the hepatic fatty-acid-oxidation response to fasting; PPARα-null mice fail to oxidize fat and accumulate hepatic lipid. PMID 10359558. Verified: PPAR-alpha activity governs ketogenesis and fatty-acid oxidation, and its de-repression during fasting is what licenses the oxidative program. this is the hepatic PPAR-alpha arm; the original primary (Hashimoto 2000, J Biol Chem 275:28918) is a mouse knockout demonstration that a defect in PPAR-alpha-inducible fatty-acid oxidation determines the phenotype -- a loss-of-function argument, not a gain-of-function one. The target list is explicit and spans the whole route: direct PPAR-alpha targets include the fatty-acid transport proteins CD36, SLC27A1 and FABP4, the mitochondrial entry transferase CPT1A, and the fatty-acyl-CoA dehydrogenases — uptake, entry and oxidation are induced together rather than sequentially. PPAR-alpha also induces lipophagy in the fasted state, in coordination with FXR which suppresses autophagy when fed. Knockout: the PPAR-alpha-null mouse is described as a model of fatty-acid oxidation disorders (Leone 1999). PPAR-alpha has the highest hepatic expression of the three PPARs and is activated by endogenous ligands — non-esterified and oxidized fatty acids, eicosanoids, bilirubin — so the fat itself switches on its own disposal. PMID 10359558

It increases Fat uptake enzyme.

10.1194/jlr.M200327-JLR200 — PPAR-alpha induces LPL transcription through a peroxisome-proliferator response element mapped in the human LPL promoter at -169 TGCCCTTTCCCCC -157, bound by PPARalpha-RXR heterodimers (PMID 8895578). Human monocyte-derived macrophages exposed to PPARalpha ligands increased LPL mass and activity with enhanced binding to that same human PPRE (PMID 12208475). Tissue-split recorded: PPARalpha drives LPL in liver, PPARgamma in adipose - the source paper proves the two are not interchangeable. doi 10.1194/jlr.M200327-JLR200 · PMID 8895578

It reduces Clears liver fat.

Named across five reviews of the cardiovascular-renal-hepatic-metabolic syndrome as part of the shared mechanism set: ectopic fat accumulation triggered by metabolic reprogramming, oxidative stress and inflammation induced by energy overload, modified lipids, UREMIC toxins, and senescence, as well as insulin resistance pathways activated by pro-inflammatory factors and lipotoxic products, collectively exacerbate simultaneous hepatic and renal injury. doi 10.1038/s41581-026-01066-0 · PMC11725408

It increases Insulin sensitivity.

insulin, fasting insulin, type 2 diabetes and blood lipids share one literature: heart, kidney, liver, and metabolic diseases share interlinked pathological mechanisms, including inflammation, oxidative stress, insulin resistance, and endothelial health issues. These four overlap by nature: fasting insulin is the level, insulin is the signaling, type 2 diabetes is the diagnosis and blood lipids are the substrate. The same driver legitimately reaches all four, and a reader should not mistake four arrows for four independent findings. doi 10.1038/s41581-026-01066-0 · PMC11725408
