Brain adaptability
Synaptic Plasticity
Also searched as Synaptic Plasticity.
You want it robust: plasticity is the substrate of learning, recovery and cognitive longevity.
In depth
The brain's capacity to rewire in response to use. It is not unlimited and it is not uniform — some circuits stay plastic for life, others close after a developmental window that never reopens.
What drives it
Supports BDNF signaling increases it.
Minichiello 2002, Neuron — BDNF/TrkB drives hippocampal long-term potentiation (synaptic plasticity) via PLCγ-CaMKIV-CREB signaling. PMID 12367511
Longevity gene increases it.
Tested in both directions in the same work: SIRT1-knockout mice showed impaired immediate memory, classical conditioning and spatial learning, and the deficits were associated with defects in synaptic plasticity — with basal synaptic transmission and NMDA-receptor function unchanged, which localises the effect to plasticity itself; mice overexpressing SIRT1 in brain had normal plasticity and memory. The authors conclude SIRT1 is indispensable for normal learning, memory and synaptic plasticity (PMC2921958). Mechanism: a SIRT1/YY1 repressor complex regulates miR-134, which in turn regulates CREB and BDNF, and SIRT1-null mice show decreased long-term potentiation in hippocampal CA1 (PMC4040287). Hippocampal SIRT1 knockdown damages plasticity in healthy animals and re-expression rescues it (PMC10848102). Note this runs through BDNF rather than around it. PMC2921958
Sleep increases it.
chemogenetic enhancement of cAMP during the period of sleep deprivation prevents deficits in a persistent form of long-term potentiation that is induced at the Schaffer collateral synapses in the hippocampal CA1 region. doi 10.1152/jn.2002.88.2.1073 · PMC9829098
Lowers cortisol reduces it.
in all hippocampal areas, i.e., CA1, CA3, and dentate gyrus, induction of LTP is greatly impaired after a prolonged period of mild stress or chronic corticosteroid exposure; a brief 30 min restraint stress protocol reversed the impaired LTP in both Tg2576 and 5XFAD mice. doi 10.1038/s41598-019-47452-6 · PMC3859225
Brain inflammation reduces it.
IL-1 receptor antagonist had no effect on LTP at 21-24 degrees C, but displayed a concentration-dependent inhibition of LTP at 34-36 degrees C. doi 10.1007/s11064-018-2517-8 · PMID 14597099
Insulin sensitivity increases it.
Molecular evidence links alteration of hippocampal insulin sensitivity with changes of both adult neurogenesis and synaptic plasticity, with crosstalk between insulin and neurotrophin signaling - which is BDNF, already drawn into this pathway. doi 10.1007/s10522-025-10300-4 · PMC6685093
Stress & cortisol reduces it.
Chronic stress disrupts neurogenesis, synaptic plasticity, and neurotransmitter balance, ultimately accelerating cognitive decline and increasing susceptibility to neurodegenerative diseases. doi 10.1007/s10522-025-10300-4 · PMC6685093
Oxidative stress reduces it.
Named among the core mechanisms across six reviews of cognitive aging and synaptic plasticity: neurotransmitter systems, calcium signaling, neurotrophic support, cellular signaling pathways, and neuroinflammation, oxidative stress, and vascular integrity, all of which redirect the trajectory of synaptic failure associated with cognitive decline in aging. doi 10.1007/s10522-025-10300-4 · PMC6685093

What The Cold Turns On increases it.
RBM3 induction and synapse preservation are rodent findings; human equivalence is not established. Carried by the hormesis, inflammaging and thermogenesis reviews. doi 10.1002/med.21743
What it drives
It increases Memory & focus.
Synaptic plasticity is a well-established mechanism supporting learning and memory across the lifespan, and cognitive functions such as memory, processing speed and executive function commonly decline, driven largely by changes in neuroplasticity mechanisms. Anything sold on “growing new brain cells” is selling the rodent result. Synaptic plasticity is the human mechanism; neurogenesis is not established in adult humans. doi 10.1007/s10522-025-10300-4 · PMC6685093
It reduces Calm pain signaling.
The defining feature is that the pain is real and the tissue is not damaged: central sensitization manifests as hyperalgesia, allodynia, expanded receptive fields, and impaired endogenous pain inhibition, and in osteoarthritis the same phenomenon does not correlate with radiographic findings - severe pain may persist despite stable joint structure. The mechanism is a balance failure in both directions at once: in the dorsal horn there is excessive release of glutamate, ATP, and substance P, leading to activation of NMDA receptors - while at the same time descending inhibitory mechanisms, notably those mediated by GABA, ARE weakened. Too much excitation and too little inhibition. And mast cells carry a specific hypothesis: thalamic mast cells releasing histamine, IL-1-beta, IL-6, TNF, CGRP and substance P, either stimulating thalamic nociceptive neurons directly or via microglia. Stated by the authors as a hypothesis, not an established mechanism. The reviewers ARE blunt about the state of knowledge: the pathophysiology and etiopathogenesis remain largely unknown, and there is NO definitive pathogenesis except for altered central pain pathways. doi 10.3390/biomedicines12071543 · PMC6687840
