Eases ER stress
ER stress↓
Also searched as ER stress.
Every protein a cell makes is folded in the endoplasmic reticulum. When misfolded ones pile up, the cell runs the unfolded protein response: slow translation, make more chaperones, degrade the wreckage. That Response is Protective when it is Brief and Destructive when it is Chronic — the same machinery that rescues a cell will eventually kill it. Trace it upward: oxidative load, calcium balance, excess fat, high glucose. It is also a calcium store, which is why the excitotoxic calcium flood reaches it too.
In depth
The cell's quality-control system for newly made proteins — and what happens when the backlog becomes permanent.
What drives it
Neuronal overexcitation increases it.
Excitotoxicity, endoplasmic reticulum stress and mitochondrial dysfunction are treated as one axis in the neuroprotection literature, and the two agents that protect against excitotoxic injury - taurine and DETC-MeSO - both also block specific ER stress pathways. The cascade does not stop at the mitochondrion. doi 10.1016/j.jhep.2018.06.008 · PMC4630664
Oxidative stress increases it.
Oxidative stress is named among the factors that perturb ER homeostasis, producing the accumulation of misfolded protein in the ER lumen that defines the condition. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
Taurine status reduces it.
Taurine prevents calcium overload and inhibits specific ER stress pathways, acting on two points of this cascade by name. PMC4630664
Protein quality control reduces it.
The unfolded protein response governs proteostatic control: chaperone synthesis improves folding capacity and XBP1 drives the degradation that clears the misfolded backlog. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
Supports weight management increases it.
Obesity sustains ER stress, and the resulting unfolded protein response drives the chronic low-grade inflammation and altered insulin signaling that follow. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
Blood-sugar support increases it.
High glucose raises ER stress in neurons; blocking it pharmacologically reduced the inflammation and cell death that followed, in vitro and in vivo. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
Heart calcium handling reduces it.
The ER is important for protein synthesis, folding and transport; lipid synthesis; and calcium HOMOEOSTASIS. Calcium imbalance may lead to perturbations in ER homeostasis. 10.1016/j.jhep.2018.06.008. doi 10.1016/j.jhep.2018.06.008
What it drives
It increases Fat-overload cell damage.
10.1371/journal.pone.0097868 — The unfolded protein response is the route by which lipid overload becomes cell death. Verbatim: alpha-linolenic acid 'prevents endoplasmic reticulum stress-mediated apoptosis of stearic acid lipotoxicity on primary rat hepatocytes' - it 'distinctly reduced cell death and levels of all three indicators of ER stress'. The pro-apoptotic arm is named in the same literature: palmitate drives 'nuclear translocation of pro-apoptotic C/EBP-homologous-protein-10 (CHOP)'. primary hepatocytes and hepatoma cells. Note this edge is a sequence, not a synonym - relieving ER strain is upstream of preventing the death, and a driver graded on one is not automatically graded on the other. doi 10.1371/journal.pone.0097868
It increases Cell turnover.
When the unfolded protein response cannot resolve the backlog, the same machinery that rescues a cell activates its death program through CHOP and IRE1. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It reduces Insulin sensitivity.
ER stress disrupts insulin action through every arm of the unfolded protein response - IRE1-alpha phosphorylates JNK and IkB kinase, and insulin signaling falls. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Inflammatory alarm.
A named molecular route rather than an association: IRE1-alpha degrades miR17, TXNIP rises unchecked, and the NLRP3 inflammasome activates. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Brain inflammation.
ER stress and unfolded protein response activation initiate and worsen chronic neuroinflammation alongside oxidative and nitrosative stress. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Calms body-wide inflammation.
Prolonged ER stress produces the chronic low-grade inflammation that accompanies obesity and cardiovascular disease. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It reduces Liver.
CHOP and GRP78 are elevated in liver biopsies from patients with steatohepatitis, and the CHOP/GRP78 ratio correlates with both disease activity score and liver injury. A brief unfolded protein response protects against steatosis; sustained ER stress aggravates it. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Arterial plaque.
ER stress and the cell death it causes play a non-redundant role in atherosclerosis and in diabetic macrovascular complications. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It reduces Heart & blood pressure.
Sustained ER stress compromises the cardiovascular system through unfolded protein response activation, chronic inflammation and altered insulin signaling. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It reduces Mitochondrial energy.
Unfolded protein response activation produces mitochondrial dysfunction across several dimensions, alongside oxidative and nitrosative stress. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Oxidative stress.
ER stress initiates and worsens oxidative and nitrosative stress - the reciprocal of the arm by which oxidative stress causes it. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It reduces Lowers cholesterol.
Lipid gene expression networks respond directly to sustained ER stress, and each arm of the unfolded protein response contributes to the disturbance. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Type 2 diabetes.
ER function becomes impaired in diabetes, and ER stress triggers insulin resistance through excessive fat accumulation and disrupted insulin signaling. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
It increases Liver cell swelling.
Verbatim (S2095177925001352, Atractylenolide II): 'AT-II effectively supresses accumulation of lipids and ER stress in palmitic acid-induced hepatocytes' and 'AT-II reduces ER stress, lipogenesis and inflammation... through stimulation of the hepatic FXR-SERCA2-eIF2alpha axis', confirmed by SERCA2 knockdown and FXR silencing. The COUNTERFACTUALS test the ER-stress step, not the ballooning readout. Ballooning was not scored in that paper. This edge asserts the mechanistic link that ER stress drives the swollen-cell phenotype; NO driver is graded on ballooning.
