Protein quality control
You want it maintained: proteostasis is a hallmark of youth, and its collapse underlies the aggregation diseases of age.
In depth
The protein quality-control network — the running balance between folding, chaperoning and clearing that keeps the proteome healthy.
What drives it
Heat shock response increases it.
Hartl 2011, Nature 475:324 — molecular chaperones (heat-shock proteins) prevent aggregation and promote folding, maintaining protein homeostasis (proteostasis). doi 10.1038/nature10317
Heat shock master switch increases it.
HSF1 governs the heat-shock response and the chaperone network that maintains protein folding - 'HSF1 is the master regulator in vertebrates'. doi 10.1038/nrm2938
Cellular cleanup increases it.
Degradation is one of the three branches of the proteostasis network, alongside synthesis and folding, and autophagy is the branch that handles what a chaperone cannot refold: 'the autophagy system clears larger aggregates or membrane-associated proteins'. doi 10.1083/jcb.201709072
Heat exposure increases it.
Verbatim: 'The heat shock response (HSR) is a cellular stress response that senses protein misfolding and restores protein folding homeostasis, or proteostasis.' (10.1534/g3.116.038232). Tier: animal. Caenorhabditis elegans (whole organism, tissue-specific reporters) · heat shock + RNAi knockdown/overexpression · dose not stated in abstract · n not stated · genetic/reporter stu. doi 10.1534/g3.116.038232
What it drives
It increases Memory & focus.
Hartl 2011, Nature 475:324 — an age-related decline in proteostasis capacity permits the protein-aggregation diseases (Alzheimer's, Parkinson's), tying proteostasis to cognitive integrity. doi 10.1038/nature10317
It increases Muscle.
The organising frame of the whole node: the core mechanisms of sarcopenia lie in protein homeostasis imbalance - the molecular scissors gap between anabolic decline (mTORC1 signal resistance) and catabolic hyperactivity (ubiquitin-proteasome and autophagy overactivation). Muscle mass is a balance, not a quantity, and both sides move with age. doi 10.3390/ijms26146740 · PMC13074595
It reduces Cellular aging.
Named across five inflammaging and hallmarks-of-aging reviews as part of the shared mechanism set that links molecular damage to organ decline: nutritional perception dysregulation and inflammatory aging ARE the core key elements linking upstream molecular damage to downstream organ function decline. doi 10.1016/j.molmet.2023.101755 · PMC11233824
It increases Recovery.
10.3390/cells12020249 — Recovery is assembled from the sarcopenia and VO2max literatures; none of the papers cited studied recovery itself. doi 10.3390/cells12020249
It increases Vision.
Why the retina is the most oxidatively stressed tissue in the body, stated directly: the retina is highly susceptible to oxidative damage due to its elevated oxygen consumption, abundant polyunsaturated fatty acids, and continuous exposure to light. Three risk factors that cannot be separated from its function. And the aging failure is specific and double: in the elderly both the mitophagic process and Nrf2 activation ARE impaired, thus causing retinal cell death. The carotenoid case is unusually mechanical: lutein and zeaxanthin ARE selectively taken up by the RPE, preferentially accumulated in the human macula, and transferred to photoreceptors - a targeted delivery route, not a general antioxidant claim. The Eye Disease Case-Control Study found high plasma levels of lutein and zeaxanthin associated with reduced risk of neovascular AMD. By contrast the reviewers say the relationships between dietary and serum levels of the antioxidant vitamins and age-related macular disease ARE less clear - with alpha-tocopherol the exception, convincingly demonstrated. Lipofuscin is itself a photoreactive substance - the waste product becomes a new damage source. doi 10.3390/antiox14101251 · PMID 11033038 · PMC12026614
It reduces Eases ER stress.
The unfolded protein response governs proteostatic control: chaperone synthesis improves folding capacity and XBP1 drives the degradation that clears the misfolded backlog. doi 10.1016/j.jhep.2018.06.008 · PMC4100367
