Neuronal overexcitation

Excitotoxicity

When The Signal Becomes The Poison

1 blend on this site modulates it.

Also searched as Excitotoxicity.

Glutamate is how neurons talk. When it is not cleared fast enough, the conversation becomes a scream: nmda receptors stay open, calcium floods in, and the mitochondria that normally soak it up saturate and collapse. Sodium swells a neuron and it recovers — calcium kills it. Trace it upward: how much glutamate is outside the cell, how much magnesium is blocking the channel, and how much energy the mitochondria have left to buffer with. That last one is why riboflavin, magnesium and CoQ10 act on the threshold rather than the event.

In depth

Neurons killed by too much of the brain's own excitatory messenger — the failure mode shared by stroke, epilepsy and slow neurodegeneration.

What drives it

Quiets overexcitation increases it.

Glutamate is the brain's principal excitatory messenger, and it becomes a neurotoxin the moment clearance fails: the resulting rise in extracellular glutamate drives a massive influx of sodium and calcium through NMDA receptors. The transporter modeled here exports glutamate while importing cystine for glutathione, so raising it protects the cell against oxidation and raises the excitotoxic load at the synapse - the same molecule reads opposite by compartment. doi 10.1186/s40035-021-00278-7 · PMC8788129

Cortical spreading wave increases it.

Cortical spreading depression travels as a wave of mass glutamate release and NMDA-dependent depolarization, carrying the excitotoxic load across adjacent cortex. doi 10.1186/s40035-021-00278-7 · PMC8788129

Mitochondrial energy reduces it.

Mitochondria buffer the calcium that excitotoxicity admits, so their capacity sets the threshold at which injury begins. Calcium arrives in two peaks, and the second - delayed calcium deregulation - marks the moment the buffer saturates. doi 10.1186/s40035-021-00278-7 · PMC8788129

Magnesium status reduces it.

Magnesium sits in the NMDA channel as its physiological voltage-dependent block; the depolarization that relieves it is what opens the route for calcium. doi 10.1186/s40035-021-00278-7 · PMC8788129

Oxidant-producing enzyme increases it.

NADPH oxidase is the primary source of superoxide generated by NMDA receptor activation - not a general contributor but the named origin. doi 10.1038/nn.2334 · PMC8788129

Oxidative stress increases it.

Oxidative stress both follows excitotoxic injury and precipitates it; the reviews state plainly that excitotoxicity can be regarded as a consequence of oxidative stress as much as a cause of it. doi 10.1186/s40035-021-00278-7 · PMC8788129

Brain inflammation increases it.

Inflammatory activation lowers the threshold at which excitatory signaling becomes injurious, and excitotoxic injury recruits further inflammation in turn. doi 10.1186/s40035-021-00278-7 · PMC8788129

Calm reduces it.

Inhibitory tone opposes the depolarization that lifts the magnesium block and admits calcium; GABAergic signaling is the counterweight to excitation. doi 10.1186/s40035-021-00278-7 · PMC8788129

Supports BDNF signaling reduces it.

Neurotrophic signaling protects neurons against excitotoxic injury. The evidence here is weaker than the calcium and mitochondrial arms and is recorded at that strength. doi 10.1186/s40035-021-00278-7 · PMC8788129

Taurine status reduces it.

Taurine prevents calcium overload and inhibits specific endoplasmic reticulum stress pathways - a named, orally available intervention on this cascade rather than a laboratory tool. PMC4630664

Cellular fuel reduces it.

Altered energy metabolism is named among the mechanisms that lead to excitotoxic damage; cellular energy status governs how much calcium the mitochondria can absorb before the buffer fails. doi 10.1186/s40035-021-00278-7 · PMC8788129

Heart calcium handling increases it.

The massive influx of calcium through overactivated NMDA receptors is the central event, triggering mitochondrial collapse and activating a host of cell-destroying enzymes. Sodium influx results in swelling…. PMC8788129

What it drives

It increases Cell turnover.

Sustained calcium entry releases pro-apoptotic factors and commits the neuron to programmed death. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Memory & focus.

Excitotoxicity is the primary mechanism of neuronal loss in acute central nervous system injury and is implicated across Alzheimer's, Parkinson's, Huntington's and amyotrophic lateral sclerosis. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Mood.

Neuronal loss and the inflammatory response that accompanies excitotoxic injury are documented contributors to depressed mood in the neurodegenerative literature. doi 10.1186/s40035-021-00278-7 · PMC8788129

It increases Brain inflammation.

Excitotoxic injury recruits inflammatory activation, which in turn lowers the threshold for further injury. doi 10.1186/s40035-021-00278-7 · PMC8788129

It increases Oxidative stress.

Excessive NMDA receptor activation produces reactive oxygen species in quantity, with NADPH oxidase the primary source. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Mitochondrial energy.

Calcium overload collapses mitochondrial function - the same organelle whose buffering capacity determined the threshold in the first place. doi 10.1186/s40035-021-00278-7 · PMC8788129

It increases Neurovascular calm.

Cortical spreading depression and glutamate overload connect this cascade to the migraine attack, where the wave that produces aura carries the excitotoxic load into the trigeminal system. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Hearing.

Noise-induced hearing loss is named explicitly among the conditions produced by this mechanism; cochlear hair cells die by the same route as cortical neurons. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Nerve insulation.

Oligodendrocytes and the myelin they maintain are damaged by the same calcium-driven injury that kills neurons. doi 10.1186/s40035-021-00278-7 · PMC8788129

It reduces Sustained focus.

It increases Eases ER stress.

Excitotoxicity, endoplasmic reticulum stress and mitochondrial dysfunction are treated as one axis in the neuroprotection literature, and the two agents that protect against excitotoxic injury - taurine and DETC-MeSO - both also block specific ER stress pathways. It reduces Plasticity switch.

The excitotoxic glutamate flood reaches extrasynaptic receptors, and those receptors antagonise excitation-transcription coupling directly - inactivating ERK-MAPK signaling and shutting CREB down. The authors' own conclusion is that specific blockade of extrasynaptic receptors may prevent neuron loss. doi 10.1038/nn835 · PMID 11953750 · PMC5339681
