Active thyroid hormone (T3)

The Metabolic Thermostat

6 blends on this site modulate it.

Also searched as Active thyroid hormone (T3).

About 80% of it is made outside the thyroid, in your tissues. That is why the standard blood test can read normal while the hormone that does the work is low.

In depth

Active thyroid hormone (T3) — the signal that sets metabolic rate, temperature and energy availability.

What drives it

Thyroid hormone conversion increases it.

The thyroid does not make most of your active hormone — your tissues do. T4 is a PROHORMONE and must be activated to T3 by type-1 or type-2 selenodeiodinase, and about 80% of circulating T3 is generated outside the thyroid by those two enzymes (PMC3673746; American Thyroid Association-commissioned review, PMC10517335). A third deiodinase inactivates T3 and T4 by removing an inner-ring iodine, so the same enzyme family both switches the hormone on and switches it off. PMC3673746

Gut barrier increases it.

Causal by the one experiment that settles microbiome questions: fecal microbiota transplant from individuals with primary hypothyroidism into mice resulted in altered circulating thyroid hormone levels compared with mice receiving microbiota from healthy donors, supporting a causal role of gut dysbiosis in thyroid hormone regulation. Move the microbiome, move the hormone — in a recipient who never had the disease. Three mechanistic routes ARE named: intestinal uptake of thyroid-related micronutrients; microbiota-derived enzymes regulating IODOTHYRONINE conversion and storage; and interaction with host immune cells regulating thyroid immunity. The gut reaches depression both directly and through the thyroid. The reviewers ARE unusually blunt about maturity: the research on gut microbiota and thyroid has only presented the tip of the iceberg, and more robust clinical data and basic experiments ARE still required. One causal transplant experiment is not a clinical literature. PMC9433865

What it drives

It reduces Daily energy.

Low T3 is felt as tiredness even when the standard test looks normal: selenium is required by DIO1 and DIO2 to convert T4 into T3, and when selenium is deficient that conversion falls — producing hypothyroid symptoms including fatigue, weight gain and cold intolerance even though T4 levels may appear normal (PMC12672807, PMC8472829). Note: excess selenium is harmful, not better — the same review warns of gastrointestinal and neurological effects from over-supplementation. PMC12672807

It reduces Supports weight management.

The same conversion failure shows on the scale: reduced deiodinase activity lowers T3 and manifests as weight gain alongside fatigue and cold intolerance, again with T4 apparently normal (PMC12672807). This is why a thyroid complaint and a weight complaint can share one upstream lane rather than being two separate problems. PMC12672807

It increases Cardiac output.

NBK500006 — what the evidence here shows: the sources read here establish calcium handling and the beta-adrenergic pathway in detail but do not themselves demonstrate the thyroid arm. This link is drawn because the structure requires it; it is not yet evidenced to the standard of the others here, and no claim rests on it.

It increases Mood.

The intervention result is striking: a randomized trial found 53 percent of patients on T3 achieved at least 50 percent improvement in depression scores after three weeks, versus 19 percent on LEVOTHYROXINE. In STAR*D, T3 augmentation was comparable to lithium and better tolerated. And the part that makes it a real driver rather than a deficiency correction: adjunctive T3 may be effective for depressed patients WHO ARE euthyroid. Normal thyroid tests, and it still works. The conditional carries the distinction that matters most commercially: T3 as antidepressant augmentation works, while T3/T4 combination as thyroid replacement does not improve mood. Correcting a thyroid is not the same act as augmenting an antidepressant. The limits: most patients with depression do not have biochemical evidence of thyroid dysfunction, and the HPT abnormalities reported come mostly from cross-sectional studies, with longitudinal data scarce and findings inconsistent. Mechanism: T3 modulates transcription of the 5-HT1A and 5-HT1B receptors and may enhance neurogenesis — serotonin and BDNF are both drawn into mood here. doi 10.1176/appi.ajp.2020.19080848 · PMC12437707

clinical and subclinical hypothyroidism as well as hyperthyroidism are both associated with decreased cognitive functioning, especially memory, visuospatial organization, attention, and reaction time. PMC4531303

It increases Sympathetic drive.

The receptor finding is old, large and clean: cardiac membranes from hyperthyroid rats contained 196 fmol of beta-adrenergic binding sites per milligram of protein versus 89 fmol in controls, p < 0.005 — more than double — with the dissociation constant and the agonist affinity both unchanged. Number, not affinity. Replicated in humans: T3 for ten days raised receptor density about 60 percent in fat and 30 percent in skeletal muscle. And the mechanism is transcriptional — new receptor proteins ARE synthesized, and the effect is abolished by ACTINOMYCIN D OR CYCLOHEXIMIDE. And then the function does not follow. In the same human experiment: despite increments in beta-adrenergic receptor densities, metabolic and hemodynamic sensitivity to epinephrine in vivo was unaltered. The authors write the general lesson themselves, and it deserves to be read twice: the finding of altered adrenergic receptors, even in relevant target tissues, should not be extrapolated to altered sensitivity to catecholamines in vivo in the absence of direct testing of that hypothesis. Conscious baboons agree: the dobutamine response as percent change was similar before and after hyperthyroidism. And the levels move the other way entirely, which is the part most people get backwards: hyperthyroid patients look hyperadrenergic, yet direct measures of serum catecholamines and urinary metabolites demonstrate values equal to OR less than normal — while the hypothyroid patient, who looks under-stimulated, has increased epinephrine and norepinephrine. The edge is drawn activating on the receptor arm because that arm is real and reproduced, and the conditional carries the rest. Anyone reading this as thyroid raises sympathetic tone has not read the table: the tone is if anything lower and the machinery is more abundant. A structural note: sympathetic-to-heart and thyroid-to-heart are both drawn, and this link means part of what thyroid does to the heart it does by changing the adrenergic apparatus. doi 10.1161/01.CIR.96.2.592 · PMID 2537849

It reduces Hair pigment loss.

The mechanism is local and it matters: these findings may be explained by hair follicles’ ability to transcribe deiodinases — the enzymes that activate and inactivate thyroid hormone. The follicle is not merely exposed to circulating hormone; it runs its own local conversion, which is why scalp effects can appear at systemic levels read as borderline. Association: statistically higher TSH levels in the 71-case series, and several studies have identified an association between autoimmune thyroid disease and premature graying. Scope, same as the Micronutrient Status → Hair Greying link: this is the dysfunction fraction, not age-related graying in euthyroid people. Both edges describe reversible causes sitting inside a mostly irreversible process, which is exactly the distinction a reader needs and a marketing page will not make. doi 10.1111/ijd.17580 · PMID 28584374

It increases Hair.

The mechanism is direct and receptor-level, which is the part that matters: nuclear thyroid hormone receptors TR-alpha and TR-beta ARE found in various hair follicular cells, suggesting that thyroid hormone may directly influence hair growth rather than relying on intermediary mechanisms related to the body’s metabolic status. This is not thyroid slowing everything down; the follicle reads the hormone itself — the same story as the Thyroid Hormone (T3) → Hair Greying link, where the follicle transcribes its own deiodinases. And reversal carries it: restoration of normal thyroid hormone levels can reverse this effect, normalizing the hair growth cycle. In hypothyroid patients there is an increase in the proportion of telogen hairs compared to anagen. Scale: a 500-patient retrospective of women with telogen effluvium and thyroid function testing found 150 hypothyroid and 102 hyperthyroid against 248 euthyroid — both directions of dysfunction over-represented. And the authors of that study ARE explicit that it is contested: the link between thyroid disorder and telogen effluvium is still being debated, which is why their paper is framed as a question. Drawn at a moderate tier on the receptor mechanism plus reversal, not on the retrospective alone. PMC10766245

It reduces Fasting insulin.

insulin, fasting insulin, type 2 diabetes and blood lipids share one literature: heart, kidney, liver, and metabolic diseases share interlinked pathological mechanisms, including inflammation, oxidative stress, insulin resistance, and endothelial health issues. These four overlap by nature: fasting insulin is the level, insulin is the signaling, type 2 diabetes is the diagnosis and blood lipids are the substrate.
