Fat-burning hormone
Adiponectin

You want it high: rising adiponectin tracks with a leaner, more insulin-sensitive metabolism.
In depth
An adipose-derived hormone that sharpens insulin sensitivity and steers fat toward burning rather than storage.
What drives it
Blocks new fat cells increases it.
PPARγ–adiponectin review 2005 — PPARγ agonists transcriptionally upregulate adiponectin. PMID 15711577
Supports weight management reduces it.
10.1186/s42506-020-00060-z — the one adipokine that runs backwards. Adiponectin is secreted only by fat tissue, yet its levels are paradoxically lower in obese than in lean people — the opposite of every other adipocytokine, which rise in proportion to fat mass (Diabetes Care 2003;26:2442). And the depot matters more than the weight: secretion from visceral fat falls as adiposity rises (r=−0.31, p=0.01 against BMI) while subcutaneous secretion is unchanged — which is why central obesity, not body mass as such, tracks with low adiponectin, and why lower-body fat associates with higher levels (PMC6010237). The limit: the relationship is not universal — a positive correlation appears in liver cirrhosis and in some obese individuals ('the adiponectin paradox'), so this is a strong tendency, not a law. doi 10.1186/s42506-020-00060-z · PMC6010237
Obesogen exposure reduces it.
EDCs modulate the adipokines, with reduced adiponectin together with increased leptin or leptin resistance. 10.3389/fendo.2026.1782328. doi 10.3389/fendo.2026.1782328
Beta-3 receptor signaling increases it.
Adiponectin, a WAT-derived [hormone], increased… chronic mirabegron therapy increased BAT metabolic activity. PMID 31961826
What it drives
It increases Ceramide-clearing enzyme.
PMC3134999 — Adiponectin signaling activates ceramidase activity, lowering ceramide and raising sphingosine-1-phosphate. This is the body's own clearance arm and part of why insulin sensitivity and ceramide levels move together. Shown predominantly in rodent and cell work. PMC3134999
It increases Metabolic health.
Adiponectin/insulin-sensitivity review 2003 — adiponectin increases muscle glucose uptake and fat oxidation and reduces hepatic glucose production (via AMPK), improving whole-body insulin sensitivity and glycemic control. PMID 12769732
It reduces Fasting insulin.
Adiponectin/insulin PNAS study — globular adiponectin improves insulin sensitivity (suppressing hepatic glucose output, raising muscle glucose uptake), lowering fasting insulin and glucose. doi 10.1073/pnas.1922169117
It increases Insulin sensitivity.
Yamauchi 2001, Nat Med 7:941 — adiponectin reverses insulin resistance by lowering muscle and liver triglyceride. doi 10.1038/90984
It reduces Type 2 diabetes.
insulin, fasting insulin, type 2 diabetes and blood lipids share one literature: heart, kidney, liver, and metabolic diseases share interlinked pathological mechanisms, including inflammation, oxidative stress, insulin resistance, and endothelial health issues. These four overlap by nature: fasting insulin is the level, insulin is the signaling, type 2 diabetes is the diagnosis and blood lipids are the substrate. The same driver legitimately reaches all four, and a reader should not mistake four arrows for four independent findings (doi 10.1038/s41581-026-01066-0; doi 10.3390/ijms26146962; PMC11725408; NBK279115; PMC12257864). REFUTED ON A SECOND READING. This link was shown by linked steps until a source was opened that does not support it: “These results do not provide any consistent evidence that interventions aimed at increasing adiponectin levels will improve insulin sensitivity or risk of type 2 diabetes.” — Mendelian Randomization Studies Do Not Support a Causal Role for Reduced Circulating Adiponectin Levels in Insulin Resistance and Type 2 Diabetes. Diabetes, 2013, 62(10), 3589-3598. doi 10.1038/s41581-026-01066-0 · PMID 23835345 · PMC11725408
It reduces Releases stored fat.
Adiponectin, an adipokine, is a brake on lipolysis. 'Adiponectin has been shown to inhibit lipolysis by suppressing PKA-mediated HSL activation. In addition, adiponectin reduces the amount of PKA RII-alpha, the regulatory subunit of PKA, by impairing its protein stability. Overexpression of RII-alpha abolishes the inhibitory effects of adiponectin on lipolysis. Consistently, adiponectin-knockout mice and their primary adipocytes exhibit an increased lipolysis.' The complication is real and stated: 'adipose tissue expression and circulating concentrations of adiponectin are decreased in both overweight and obesity, thereby implying a plausibly decreased anti-lipolytic effect' — so in overweight and obesity the brake is already partly off. doi 10.3389/fphys.2022.826314
