Blocks new fat cells

The Balance here is Between Goals, and it is close to even — three benefits, three costs. Raised, it improves lipid handling, raises adiponectin and improves insulin sensitivity: this is why the glitazone drugs work at all. The same activation raises CD36 fatty-acid uptake and, over time, fasting insulin and diabetes risk — which is why those drugs also cause weight gain. the honest reading: PPARγ redistributes fat rather than removing it. Whether that is worth it depends on what you are treating.
In depth
PPARγ — the master switch of fat-cell formation and lipid handling that also improves insulin sensitivity.
What drives it
Longevity gene reduces it.
Picard 2004, Nature 429:771 — SIRT1 binds and represses PPAR-γ (docking with NCoR/SMRT corepressors), promoting fat mobilization in white adipocytes. PMID 15175761
Alcohol metabolism increases it.
With SIRT1 repressed, acetylated PPAR-gamma rises with high transcriptional activity and drives its target MGAT1 — a second, transcriptional route to hepatic fat on top of the redox one. PMC4941543
Obesogen exposure increases it.
The mechanism at the receptor, which is why this pathway earns a link rather than only an association. PPAR-gamma is the central transcriptional regulator of adipogenesis and a key molecular target of various obesogens. Its activation promotes the genes for lipid metabolism and fat storage. Organotins ARE the cleanest case: they are described as promoting adipocyte differentiation as agonists of the peroxisome proliferator-activated receptor gamma / retinoid X receptor pathway, and as potent INDUCERS of adipogenesis in vertebrates. Tributyltin also triggers lipogenesis in macrophages via modifying the PPAR-gamma pathway. Phthalate MONOESTERS activate PPAR-alpha and PPAR-gamma directly. And bisphenol A reaches the same receptor epigenetically rather than as a ligand: low-dose BPA acts on adipogenesis by reducing PPAR-gamma promoter methylation in preadipocytes. Two different chemical classes, two different molecular routes, one receptor. And the same BPA paper is honest about what that did not produce: in BPA-exposed 3T3L1 cells, PPAR-gamma expression only transiently increased, without altering the adipogenic potential of the precursor cells — and the authors note contradictory evidence and unanswered questions ARE reported in the literature concerning the BPA effects on adipogenesis. The promoter changed; the cell fate did not necessarily follow. Cell culture and rodent throughout. doi 10.3389/fendo.2026.1782328 · PMC7696502
Fat-releasing enzyme (HSL) increases it.
This lipase does a second job nobody expects: it manufactures the fat cell's own growth signal. In humans carrying a homozygous lipe deletion that removes the protein entirely: 'Although a defect in an enzyme that is critical for lipolysis would be expected to result in excess lipid storage, we observed the opposite, providing evidence that HSL plays a key role in maintaining adipogenesis and adipocyte function... These findings are consistent with the hypothesis that HSL deficiency in humans results in decreased production of endogenous ligands for PPAR-gamma, RXR-alpha, OR both' (doi 10.1056/NEJMoa1315496 (nejm)). And the authors draw the therapeutic conclusion themselves: 'Our results suggest that activation of HSL with consequent generation of endogenous ligands' is a route worth taking — in the same paper, in the same breath as noting that direct PPAR-gamma agonists 'can have untoward side effects'. doi 10.1056/NEJMoa1315496

It reduces Fasting insulin.
insulin, fasting insulin, type 2 diabetes and blood lipids share one literature: heart, kidney, liver, and metabolic diseases share interlinked pathological mechanisms, including inflammation, oxidative stress, insulin resistance, and endothelial health issues. These four overlap by nature: fasting insulin is the level, insulin is the signaling, type 2 diabetes is the diagnosis and blood lipids are the substrate. The same driver legitimately reaches all four, and a reader should not mistake four arrows for four independent findings. doi 10.1038/s41581-026-01066-0 · PMC11725408

PPAR-gamma is the central transcriptional regulator of adipogenesis and the key molecular target of obesogens, acting as a PPAR-gamma-RXR heterodimer to drive preadipocyte commitment and lipid accumulation. 10.3389/fendo.2026.1782328. doi 10.3389/fendo.2026.1782328
It increases Fat-releasing enzyme (ATGL).
The two lipases are coupled, not independent. 'ATGL is an established PPAR-gamma target, and its decreased expression in lipe-null adipocytes probably contributes to the global lipolytic defect' (doi 10.1056/NEJMoa1315496 (nejm)). ⇒ lose the second lipase and you lose some of the first one too, because the first is a transcriptional target of the receptor the second one's products activate. That is why a human without this enzyme has a global lipolytic failure rather than a partial one. doi 10.1056/NEJMoa1315496
