The Decision To Build Another Warehouse

2 blends on this site modulate it.

Also searched as Adipogenesis.

Fat tissue does not only fill; it recruits. Precursor cells sit in reserve and, given the right signal, commit irreversibly to becoming storage. That commitment is governed by PPAR-gamma, the master transcription factor of fat-cell formation — and it is the same receptor that endocrine-disrupting obesogens hijack to drive adiposity. Restraining this step is different from emptying an existing cell: it changes how many warehouses get built rather than how full each one is. The honest boundary. Most evidence here is cell-culture and animal; measuring new fat-cell formation directly in a living person is not something a trial can readily do. And restraint is not the same as reversal — nothing here removes a cell that already exists.

In depth

Adipogenesis — the process by which a precursor cell commits to becoming a new fat cell.

What drives it

Blocks new fat cells increases it.

PPAR-gamma is the central transcriptional regulator of adipogenesis and the key molecular target of obesogens, acting as a PPAR-gamma-RXR heterodimer to drive preadipocyte commitment and lipid accumulation. 10.3389/fendo.2026.1782328. doi 10.3389/fendo.2026.1782328

Cortisol receptor signaling increases it.

It releases fat and builds the depot at the same time. 'GCs also promote adipocyte hypertrophy and differentiation of central fat depots that can lead to abdominal obesity and insulin resistance. In vitro adipogenesis studies include the wide use of the synthetic GC, dexamethasone' (Endotext nbk555602 (NIH Bookshelf)). The standard laboratory reagent for making fat cells is a glucocorticoid.

Fat-releasing enzyme (HSL) increases it.

This lipase does a second job nobody expects: it manufactures the fat cell's own growth signal. In humans carrying a homozygous lipe deletion that removes the protein entirely: 'Although a defect in an enzyme that is critical for lipolysis would be expected to result in excess lipid storage, we observed the opposite, providing evidence that HSL plays a key role in maintaining adipogenesis and adipocyte function... These findings are consistent with the hypothesis that HSL deficiency in humans results in decreased production of endogenous ligands for PPAR-gamma, RXR-alpha, OR both' (doi 10.1056/NEJMoa1315496 (nejm)). And the authors draw the therapeutic conclusion themselves: 'Our results suggest that activation of HSL with consequent generation of endogenous ligands' is a route worth taking — in the same paper, in the same breath as noting that direct PPAR-gamma agonists 'can have untoward side effects'. doi 10.1056/NEJMoa1315496

Glycerol phosphate enzyme increases it.

The enzyme that builds the backbone fat is rebuilt on — and the reason one measurement in this field can mislead. 'GPDH is an important enzyme for lipid metabolism that is central to triglyceride synthesis' and has an 'important role... In converting preadipocytes to adipocytes' (doi 10.3390/molecules191216656 (pmc6270905) — Anti-Obesity Effects of Hispidin and Alpinia zerumbet Bioactives in 3T3-L1 Adipocytes). It makes the three-carbon scaffold onto which released fatty acids are re-attached. Here is why it matters for reading any fat-release number: the standard test measures free glycerol in the fluid around the cell. If this enzyme is blocked, glycerol is not consumed to rebuild fat — so it accumulates. A rise in the measurement can therefore mean more fat coming out, OR less being put back, and the two look identical on the readout. One compound in this formula inhibits it by nearly ninety-eight percent, which is genuinely useful — and means its headline release figure must be read with that in mind rather than at face value. doi 10.3390/molecules191216656

Nitric oxide receptor enzyme increases it.

Two different enzymes make the same messenger, and in fat they do opposite things. This is the most counter-intuitive finding on this MAP and it overturns the obvious assumption. The messenger made at the heart-peptide receptor releases fat. The same messenger made by the nitric-oxide enzyme does the reverse. Measured in living people, not in cells: 'the NO synthase inhibitor, L-NMMA, but not the biologically inactive enantiomer, increased glycerol levels in the microdialysates without causing a change of local blood flow... Nitric oxide gas as well as the NO donor, NITROGLYCERINE, reduced glycerol release from isolated adipocytes' (PMID 10323597 — a nitric oxide-mediated mechanism regulates lipolysis in human adipose tissue in vivo (microdialysis)). ⇒ blocking nitric oxide raises fat release. Supplying it lowers fat release. And the blood-flow explanation is ruled out in the same experiment. It also favors fat cell formation: 'nitric oxide and downstream second messenger cGMP and cAMP enhance adipogenesis in primary human preadipocytes' (Cytotherapy 2010 — Nitric oxide and downstream second messenger cGMP and cAMP enhance adipogenesis in primary human preadipocytes). The practical rule this establishes: a compound that raises nitric oxide is not a fat-release agent, however good it is for blood flow. The two effects are separable and they point in opposite directions. The field itself is cautious: 'the effect of nitric oxide appears complex and is not fully understood'. PMID 10323597

NAD methylation leak increases it.

NNMT knockdown in adipose tissue protects against fat accretion, indicating a permissive role in adipogenesis. doi 10.1038/s41598-023-34916-z
