Lipase brake
G0S2 lipase inhibition
Also searched as G0S2 lipase inhibition.
You want it low, and this is the honest hole in almost every fat-release product. It inhibits the lipase in a dose-dependent, non-competitive way even when the co-activator is there — meaning the usual route of freeing the activator does not get past it. Its levels swing across feeding, fasting and refeeding, so it is a real physiological switch and not a curiosity. It also builds fat: the same protein carries an enzyme activity that helps make triglyceride, so it locks the door and stocks the room. Nothing botanical is known to address it.
In depth
A protein that binds the rate-limiting lipase directly and shuts it — even when the activator is present.
What drives it
Blocks new fat cells increases it.
G0S2 is a PPAR target gene strongly induced during adipogenesis and in the fed state. doi 10.1016/j.molmet.2019.08.013
What it drives
It reduces Fat-releasing enzyme (ATGL).
The padlock nothing in this formula reaches. 'g0s2 inhibits ATGL activity and decreases lipolysis in rodents and humans with a dose-dependent, non-competitive mANNER even in the presence of the coactivator CGI58' (doi 10.3389/fphys.2022.826314). Mapped structurally: 'the central hydrophobic domain of G0S2 is required for interaction and inhibition of ATGL, as well as the N-terminal patatin-like region (up to Leu254) of ATGL.' ⇒ freeing the activator does not get past this. doi 10.3389/fphys.2022.826314
It reduces Releases stored fat.
'Changes in G0S2 levels are observed throughout the feeding-fasting-refeeding cycle, suggesting that G0S2-mediated modification of lipolysis is required for rapid metabolic adaptations' (doi 10.3389/fphys.2022.826314) — a real physiological switch. And it works both ends: 'Beyond its well-recognized antilipolytic function, G0S2 also harbors lysophosphatidyl-acyltransferase activity and plays a critical role in promoting hepatic triglyceride synthesis. This dual function, acting as both a lipid-synthesizing enzyme and an ATGL inhibitor, enables G0S2 as a master regulator of lipid storage and mobilization balance.'. doi 10.3389/fphys.2022.826314
