Fat-tissue signal (TNF-α)
TNF-α↓
Also searched as TNF-α.
You want it low, and that is not a contradiction. It does raise fat release, but it is the pathological version: it works by tearing down the droplet coat rather than opening it on demand, and the release it causes is the uncontrolled kind that raises circulating fat and worsens insulin resistance. It needs sugar to work: neither this signal nor glucose alone did much to release in cells, but together they markedly stimulated it — and only sugars that the cells could turn into lactate would do. The fed, inflamed state is the one that makes this happen, which is precisely the state a person taking a fat-release product is usually trying to leave.
In depth
An inflammatory signal that drives fat release — the unwanted kind, in the wrong conditions.
What drives it
Insulin sensitivity reduces it.
'TNF-alpha and hyperglycemia both impair insulin sensitivity in vivo. This may be secondary to stimulation of adipose tissue lipolysis and consequent increased circulating free fatty acids' (PMID 14693700). Drawn as insulin opposing it, on the reciprocal relationship the source describes; not on an experiment measuring insulin lowering this cytokine. PMID 14693700
Calms body-wide inflammation increases it.
This cytokine is the canonical inflammatory signal named as a non-adrenergic lipolytic stimulus (Endotext NBK555602 (NIH Bookshelf)), and its lipolytic action is glucose-dependent (PMID 14693700). Drawn on the category relationship; the specific experiment measuring systemic inflammation raising adipose TNF-alpha was not retrieved and this edge is marked accordingly. PMID 14693700
What it drives
It increases Releases stored fat.
It raises release, and it is the wrong kind. Named as a non-adrenergic lipolytic stimulus: 'Non-adrenergic lipolytic stimuli include glucocorticoids, natriuretic peptides, growth hormone, and TNF-alpha' (Endotext nbk555602 (NIH Bookshelf)). And it is conditional on sugar: 'neither TNF-alpha nor glucose alone has a pronounced effect on lipolysis in 3T3-L1 adipocytes. However, the combination of TNF-alpha plus glucose markedly stimulates lipolysis... The effectiveness of the sugars corresponded with production of lactate' (PMID 14693700). ⇒ the fed, inflamed state is what makes this happen. PMID 14693700
It reduces Fat droplet coating.
The mechanism is demolition, not unlocking. 'The ability of TNF-alpha to phosphorylate extracellular signal-regulated kinase 1 (ERK1) and ERK2 and to downregulate perilipin (which has been implicated in the lipolytic effect of TNF-alpha)' (PMID 14693700). ⇒ It tears down the droplet coat rather than phosphorylating it open — which is why the release is uncontrolled rather than on-demand. PMID 14693700
It increases Heart muscle inflammation.
How the body first notices — and why noticing is not harmless. 'release of PAMPs and DAMPs activates innate immune pathways' (Heart Failure Reviews 2026, doi 10.1007/s10741-026-10654-y), which run through the same master inflammatory switch that drives the inflammatory response. ⇒ the alarm that detects the virus is the same alarm that, left on, damages the muscle. That is why the second phase is the dangerous one. doi 10.1007/s10741-026-10654-y
It increases Fat cell clearance.
TNF-alpha induces apoptosis of human adipose cells. PMID 9392477
It increases Adrenaline responsiveness.
Prolonged TNF-alpha exposure decreases the sensitivity of fat cells to beta-adrenergic stimulation. PMID 24368730
