Growth restraint
mTOR↓
Also searched as mTOR.
The Balance here is Between Goals, and it is the sharpest trade in the whole system. For muscle you want it up: mTOR is the growth switch, and leucine, protein and resistance training raise it on purpose — the two things below that benefit are Muscle Recovery and Skeletal Muscle Mass. For longevity you want it down: eight of the ten things it drives go the wrong way when it is raised — autophagy falls, cellular aging accelerates, insulin sensitivity drops, fasting insulin and srebp-1c rise, and the senescent secretome grows. Rapamycin, an mTOR inhibitor, is the most replicated lifespan-extending drug there is. What you do not want is it up all the time. The point is the pulse, not the plateau: raised after training and protein, low the rest of the day. Chronic elevation buys you the costs without the muscle.
In depth
The nutrient-and-anabolic sensor that commits the cell to building — protein synthesis, muscle and repair — while quieting cellular clean-up.
What drives it
Fasting response reduces it.
Amino-acid/mTORC1 study (PMC3908400) — withdrawal of amino acids reversibly abolishes mTORC1 signaling; nutrient deprivation (fasting) lifts the mTORC1 growth signal, a route independent of AMPK and insulin. PMC3908400. Mechanism named precisely: reduced levels of glucose and amino acids during fasting reduce mTOR activity, which inhibits protein synthesis and stimulates autophagy. It is substrate withdrawal, not an inhibitor — which is why it reverses the moment you eat. The reviewers themselves state the limit: most data come from animal studies OR short-term human trials, raising questions about generalisability to long-term clinical outcomes, and the degree to which autophagy induction produces observable improvement in human metabolic health is still not fully understood. PMC3908400
Growth factor increases it.
IGF-1 binding to IGF-1R recruits IRS-1 and PI3K, activating Akt and then mTORC1 — the canonical anabolic route that raises muscle protein synthesis (PMC7564605). With the existing mTOR→muscle edge this wires IGF-1’s hypertrophy effect mechanistically rather than as a shortcut. mTOR activity is also tightly regulated by amino acid availability independently of this arm, and mTORC1 vs mTORC2 differ -- Raptor-deficient muscle becomes dystrophic while Rictor knockout muscle is indistinguishable from wild type. Mouse + C2C12 myotubes. PMC7564605
Growth & survival signaling increases it.
Manning & Cantley 2007, Cell 129:1261 — PI3K/Akt activates mTORC1 (via TSC2), driving growth signaling. Hub caveat: the same review notes well over 100 reported AKT substrates and states that not all have been rigorously validated. Any single AKT arrow is one of a very large fan-out.
Testosterone increases it.
Activation of Akt leads to phosphorylation and activation of downstream molecules including mTOR and p70S6K, resulting in an increase in protein synthesis. And the same kinase does the other half: furthermore, Akt activation leads to phosphorylation and inhibition of FoxO transcription factors, which ARE required for upregulation of the ubiquitin ligases MuRF-1 and MAFbx, resulting in a decrease in protein degradation. Muscle mass is a balance of two rates and testosterone moves both of them the same way through one node. That is why the effect on lean mass is reliable even where the effect on function is not — see the physical-performance findings, where testosterone moves body composition and fails on functional performance. Causal in vivo: wire-induced immobilization raised Atrogin-1, MuRF1, F4/80 and IL-6 (p < 0.001) while muscular androgen receptor expression significantly decreased and myostatin rose. Castration made the AR loss and the muscle damage worse, and both were completely ameliorated by testosterone supplementation and AR upregulation. Remove the hormone, worse; restore it, fixed. doi 10.1038/s41598-025-95115-6 · PMID 22101547 · PMC4987228
Calms inflammation reduces it.
NF-kB turns them on: TNF-alpha and IL-1 activate the IKK complex, which phosphorylates IkB-alpha, marking it for poly-ubiquitination and proteasomal degradation, thereby releasing NF-kB. Activated NF-kB then activates MuRF1 and atrogin-1, both of which ARE ligases that promote protein degradation. And transcriptional activation of the proteasome ATPase subunits Mss1 and p42 is likewise NF-kB-dependent. mTOR/Akt turns the same two off: Akt phosphorylates and inactivates FoxO proteins by promoting their export from the nucleus, and FoxOs ARE the transcriptional regulators that upregulate MuRF-1 and atrogin-1. One pair of ligases, two opposing controls, and the muscle mass is whatever the argument settles on. The direct arm — why this edge is drawn inhibitory rather than left as a shared target: IL-6/STAT3/AMPK signaling deactivates mTORC1. Inflammatory signaling does not merely compete with growth signaling downstream; it switches it off upstream. And the intervention evidence is bidirectional: genetic OR pharmacological inhibition of IKK-beta/NF-kB reduced MuRF1 expression and attenuated DENERVATION- and tumor-induced muscle atrophy in mice, while transient overexpression of a constitutively active IKK-beta OR IKK-alpha consistently led to skeletal muscle atrophy. And a commercial reality the field states plainly: the NF-kB pathway has become a prime therapeutic target — and none of the NF-kB targeting drugs ARE currently being used to treat these diseases, which the authors attribute to limited knowledge and specificity for muscle biology. A perfect target with no usable drug is the same shape as the anti-TNF story on the inflammation-to-heart link. PMID 36831310 · PMC4987228
Energy sensing reduces it.
Gwinn 2008, Mol Cell 30:214 — AMPK phosphorylates Raptor (S722/S792), inhibiting mTORC1. PMID 18439900. Verified: AMPK inhibits mTORC1 by phosphorylating TSC2 and Raptor, alleviating mTORC1-mediated suppression of downstream anabolism. mTORC1 in turn drives translation via S6K1 and 4E-BP1, so AMPK inhibition of mTORC1 conserves ATP by reducing protein synthesis. AMPK inhibits mTORC1 by two routes: phosphorylating and activating TSC2 (which switches off Rheb), and directly inhibiting Raptor, mTORC1's binding partner. This is the energy-sensor-to-growth-switch link — no fuel, no anabolism. the same review marks several AMPK substrates as requiring independent validation; TSC2 and Raptor are not among the flagged ones. PMID 18439900
Branched-chain amino acids increases it.
Leucine is a direct mTOR signal, not merely a building block - which is the whole basis of the sports-nutrition claim. Carried by the intermediary-metabolism, micronutrient and redox reviews (PMC9967593; doi 10.3390/nu18132141; PMC12673670; doi 10.1016/j.molmet.2023.101755). Verbatim: “Leucine is a proteogenic amino acid that also regulates many aspects of mammalian physiology, in large part by activating the mTOR complex 1 (mTORC1) protein kinase, a master growth controller”. doi 10.3390/nu18132141 · PMID 26449471 · PMC9967593
What it drives
It increases Recovery.
Resistance-exercise mTOR study 2016, Sci Rep — mechanical load activates mTORC1 to drive muscle protein synthesis, ribosome biogenesis and hypertrophy; blocking mTOR impairs the growth response. PMC4977552. Mechanism: mTORC1 phosphorylates S6K1 (which activates eIF4B and ribosomal protein S6) and 4E-BP1 (which releases eIF4E to bind the mRNA 5' cap), together raising cap-dependent translation. Human: both high-load and low-load-to-fatigue resistance training raised S6K1 phosphorylation and mTORC1 signaling during the recovery period. PMC4977552
It increases Muscle.
Bodine 2001, Nat Cell Biol 3:1014 — Akt/mTOR drives skeletal-muscle hypertrophy; rapamycin blocks it and genetic activation is sufficient. PMID 11715023. It was hypothesized that constitutively raising mTORC1 activity — by muscle-specific knockout of its inhibitor TSC1 — would increase protein synthesis and produce hypertrophy. A sarcopenic phenotype was observed instead: accelerated age-related loss of muscle mass and strength. Conversely, genetic suppression of mTORC1's downstream protein-synthesis effectors delays sarcopenia in naturally aging mice (PMC9891956). And synthesis is not growth: muscle-specific Raptor knockout had NO effect on load-induced increases in protein synthesis despite ablating the hypertrophic response — the authors state the protein-synthetic response was completely dissociated from the hypertrophic response, implying a concurrent rise in protein degradation. More mTOR is not more muscle. PMID 11715023 · PMC9891956
It reduces Cellular cleanup.
Kim, Kundu, Viollet & Guan 2011, Nat Cell Biol 13:132 — mTORC1 phosphorylates ULK1 Ser757, blocking autophagy initiation. DOI 10.1038/ncb2152. Two separate blocks at two stages. Initiation: mTORC1 phosphorylates ULK1 at SER757, which disrupts ULK1's ability to interact with AMPK — the brake is applied by breaking a protein-protein contact, not by destroying the enzyme (Egan 2011; Kim 2011). Maturation: mTORC1 separately phosphorylates PACER at Ser157 and WIPI2 at Ser395 to suppress autophagosome maturation. Pharmacological proof: rapamycin activates autophagy, and when mTORC1 is inhibited by nutrient lack autophagy is initiated quickly as a compensatory recovery of nutrients from the cell's own components. There is a return arrow: ULK1 phosphorylates Raptor at Ser859/Ser792 and hinders substrate docking, inhibiting mTORC1 back — a negative feedback loop that keeps mTORC1 off once autophagy has started. doi 10.1038/ncb2152.
