Also searched as DNA strand breaks and base oxidation.

This is where oxidative stress stops being abstract. The damage response decides between repair, permanent arrest and death — and a single oxidized base in the wrong place can start it inside thirty minutes.

In depth

Breaks and oxidized bases in DNA, and the signaling cascade that senses them and decides the cell's fate.

What drives it

UV radiation increases it.

Mechanistic edge. Gated on ONTRAC and the two primaries it cites for its mechanism. Verbatim: “Nicotinamide enhances repair of ultraviolet radiation-induced DNA damage in human keratinocytes and ex vivo skin.” (PMID 26488693; Carcinogenesis 2013 (keratinocytes)). PMID 26488693

Ionizing radiation increases it.

A mechanistic link, resting on double-strand-break and micronucleus evidence. Verbatim: “the betaD was reduced by 27 %, 36 %, and 47 %... a decrease by 39 %, 52 %, and 80 % of the amount of non-DSB cluster damage.”. PMID 39674631

Parasite burden increases it.

IARC Group 1 classification — three helminth species are definitive human carcinogens. Five independent sources agree. Verbatim: “The International Agency for Research on Cancer categorizes infection with the fish-borne trematodes Opisthorchis viverrini and Clonorchis sinensis and the blood fluke Schistosoma haematobium as Group 1 biological carcinogens.”. doi 10.1371/journal.ppat.1006393 · PMC4274992

Radiation exposure increases it.

Human occupational-exposure evidence. Verbatim: “The mean micronucleated binucleated cell rate was significantly higher in the industrial radiographers than in the controls (10.7 per thousand vs 6.6 per thousand, P=0.009); this difference was due to a significantly higher frequency of centromere-negative micronuclei (8.5 vs 2.2 per thousand, P<0.001).”. PMID 12438002

Oxidative stress increases it.

Oxidative stress writes itself into the genome. Hydrogen peroxide switches on both the ATR/CHK1 and ATM/CHK2 damage-signaling arms, indicating it produces single- and double-strand breaks, and the characteristic oxidative lesion is 8-OXO-guanine (Free Radic Biol Med 2021, DOI 10.1016/j.freeradbiomed.2021.06.032). The dose matters: p53 also supports survival at low stress by maintaining energy and redox balance — this pathway describes damage, not every encounter with an oxidant. (PMC8911600, Molecules 2022 doi 10.3390/molecules27051620; biorxiv 2021.05.05.442662). Hydroxyl radicals attack guanine at C8 to form 8-oxoguanine, the principal ROS-mediated DNA lesion and the standard biomarker of oxidative DNA damage. 8-oxoG MISPAIRS with adenine, driving G:C to T:A transversion mutations. Guanine-rich regions -- telomeres and promoters -- are preferentially attacked. Speed and potency: telomeric 8-oxoG production, in under 0.025 percent of the genome, was sufficient to activate ATM and ATR kinases and downstream p53 and p21 within 30 minutes, driving premature senescence by replication stress rather than by telomere shortening. doi 10.1016/j.freeradbiomed.2021.06.032 · PMC8911600

Reducing power supply reduces it.

Ribose-5-phosphate from this pathway supplies nucleotide synthesis for repair. Carried by the intermediary-metabolism and clinical reviews. doi 10.1016/j.molmet.2023.101755 · PMC9967593

Cellular fuel reduces it.

The aging-associated loss of NAD+ is related to increased expression of PARPs, caused by increased DNA damage and the need for DNA repair during aging. Covarrubias 2021, Nat Rev Mol Cell Biol — NAD+ metabolism and its roles in cellular processes during aging. PMID 33353981

Spares NAD⁺ during DNA repair reduces it.

PARP1 detects single-strand breaks and recruits XRCC1 to execute base-excision repair. doi 10.3390/ijms140918078

Antioxidant recycling reduces it.

Depleting the reduced glutathione pool in mouse embryonic fibroblasts raised reactive oxygen species and oxidative DNA damage, with nuclear glutathione showing the strongest inverse correlation with DNA strand breaks and 8-oxo-2'-deoxyguanosine lesions, so the reduced-glutathione pool that glutathione reductase regenerates suppresses DNA damage. doi 10.1093/mutage/gel043 · PMID 17012304

What it drives

It increases Damage checkpoint.

p53 is normally held at low levels by continuous MDM2-driven degradation; damage is what stabilizes it. On double-strand breaks ATM autophosphorylates and phosphorylates p53 at Ser15, and the ATM/CHK2 and ATR/CHK1 arms converge on p53 as their major downstream effector, producing G1 arrest through p21 (Free Radic Biol Med 2021; Blood 2018;131:488). Shown by blocking both ends: the ATM inhibitor caffeine rescued damage-induced arrest, and silencing p53 with siRNA attenuated it. PMC3660384

It increases Clears worn-out cells.

The cleanest causal demonstration in this part of the MAP. Using a precision chemoptogenetic tool to place 8-oxo-guanine only at telomeres — roughly 0.025% of the genome — a single induction activated ATM and ATR and the downstream effectors p53 and p21 within 30 minutes and triggered multiple hallmarks of senescence; knocking out p53 rescued the growth reduction, proving p53 enforces it. The mechanism was replication-stress-induced DDR and telomere fragility, not accelerated telomere shortening (Nat Struct Mol Biol 2022, DOI 10.1038/s41594-022-00790-y). The damage response is also required for a subset of SASP factors including IL-6 and IL-8. doi 10.1038/s41594-022-00790-y

It increases Cellular aging.

It reduces Stem cell reserve.

Genomic instability in the stem pool - hallmark 1 driving hallmark 8. doi 10.1016/j.molmet.2023.101755 · PMC12293478

It reduces Male fertility.

The finding that unifies all three nodes is that the same androgen does opposite things in different tissues: testosterone acts directly, and indirectly through intracellular conversion to dihydrotestosterone by 5-alpha reductase, as in prostate cells, and separately through estrogen effect when converted to estradiol by aromatase. Raise testosterone and you also raise DHT and estradiol - which is why the same intervention supports male hormonal health and can enlarge the prostate. And the clinical data shows both arms at once: in 183 men over 50 with BPH, testosterone was significantly lower in patients with severe LUTS (285.94 ng/dL) than mild (544.09 ng/dL) - so low testosterone tracks with worse urinary symptoms, while DHT drives the gland. That is not a contradiction to resolve, it is two different molecules. Bidirectionality is explicit: the relationship between hypogonadism and metabolic disorders is bidirectional, and a vicious circle between the two components has been documented. PMC6669361

It reduces Skin.

10.3390/cosmetics12040172 — The aging outcome lists the thirteen hallmarks themselves, each mapped to its pathway and labeled with its hallmark number. (from the reviews of this outcome). doi 10.3390/cosmetics12040172

It reduces Vision.

Why the retina is the most oxidatively stressed tissue in the body, stated directly: the retina is highly susceptible to oxidative damage due to its elevated oxygen consumption, abundant polyunsaturated fatty acids, and continuous exposure to light. Three risk factors that cannot be separated from its function. And the aging failure is specific and double: in the elderly both the mitophagic process and Nrf2 activation ARE impaired, thus causing retinal cell death. The carotenoid case is unusually mechanical: lutein and zeaxanthin ARE selectively taken up by the RPE, preferentially accumulated in the human macula, and transferred to photoreceptors - a targeted delivery route, not a general antioxidant claim. The Eye Disease Case-Control Study found high plasma levels of lutein and zeaxanthin associated with reduced risk of neovascular AMD. By contrast the reviewers say the relationships between dietary and serum levels of the antioxidant vitamins and age-related macular disease ARE less clear - with alpha-tocopherol the exception, convincingly demonstrated. Lipofuscin is itself a photoreactive substance - the waste product becomes a new damage source. doi 10.3390/antiox14101251 · PMID 11033038 · PMC12026614

It reduces Female fertility.

The central finding is that reactive oxygen species ARE required, not merely harmful: ROS in the ovary regulate ovarian steroid hormone production, follicle synthesis, maturation, ovulation, and tubal function. ROS and antioxidant levels regulate the microenvironment of the follicular fluid, which determines which follicle becomes dominant. ROS can trigger ovulation in mature GRAAF follicles, and any change in this ROS level can disrupt ovulation. So an antioxidant claim here cuts both ways - the requirement is redox homeostasis, not suppression. Excess oxidative stress damages the oocyte through mitochondrial dysfunction, follicular atresia, meiotic errors, DNA damage and telomere shortening; too little disrupts the ovulatory trigger itself. Scale: infertility affects approximately 10-15 percent of couples, with nearly half the cases attributed to female reproductive dysfunction. doi 10.3390/antiox12081490 · PMID 40712896 · PMC10666387

It increases Senescent cell signaling.

Genotoxic insults initiate the senescent phenotype and the persistent DNA-damage response sustains the secretory program. doi 10.1016/j.cellsig.2011.12.006 · PMID 21979375 · PMC7767554

It increases Spares NAD⁺ during DNA repair.

Accumulating DNA damage is what activates PARP1, and each repair event draws on the NAD+ pool. This edge is why DNA damage and energy metabolism are the same problem rather than two. mechanism, not a human outcome. Chini 2024, Aging Cell, doi 10.1111/acel.13920 / PMC10776128 — NAD metabolism: role in senescence regulation and aging. doi 10.1111/acel.13920 · PMC10776128
