Blood flow to fat tissue

Adipose Tissue Blood Flow

The Getaway

You want this open, and it is the step almost nobody counts. Neither stored fat nor the fatty acids it releases dissolve in water, so leaving the tissue requires a carrier and enough flow to move it. When perfusion is varied and the release rate is held constant, net fat output still changes — which means transport, not the enzymes, was limiting. The cruel part, measured in people: adrenaline raises fat release and reduces flow at the same time. And the tissue people most want to lose has the lowest flow to begin with — femoral is lower than abdominal, and superficial stores resist while deeper ones respond. That is what 'stubborn fat' actually is.

In depth

Blood supply to fat tissue — the only route by which released fat can actually leave.

What drives it

Fasting response increases it.

'Subcutaneous adipose tissue blood flow (ATBF) is relatively high (in the fasting state, similar to that of resting skeletal muscle, when expressed per 100 g tissue) and changes markedly in different physiological states. Those most studied are after ingestion of a meal, when there is normally a marked rise in ATBF, and exercise, when ATBF also increases' (Int J Obes, Regulation of human subcutaneous adipose tissue blood flow). Recorded honestly: the source says flow is high in fasting and rises after a meal and with exercise — it is not a simple fasting-only story, and the edge is drawn on the fasting baseline being adequate rather than on fasting being maximal. Alertness chemical reduces it.

The cruellest finding in this whole pathway, and it is measured in people. Human microdialysis of abdominal and femoral fat in young healthy subjects of both sexes: 'epinephrine and norepinephrine perfusion increased extracellular glycerol and reduced blood flow' (PMID 9655773). The very signal that releases the fat cuts the route out. Note the dissociation the same study found: 'Isoproterenol perfusion increased extracellular glycerol levels' and 'induced a greater increase in local blood flow in Fem adipose tissue' — a pure beta agonist raised flow where the natural catecholamines lowered it. PMID 9655773

Blood flow (nitric oxide) increases it.

10.1161/01.cir.0000105681.70455.73 — the answer to the riddle, part one — and it is independent of epinephrine. 'It seems that basal adipose blood flow is maintained by a basal nitric oxide production while vasodilatation in these vascular beds depends on beta-adrenergic signals.' The definitive human experiment tested all three arms at once: 'We investigated the endogenous preprandial and postprandial regulation of ATBF by applying local tissue blockade of beta-adrenergic (propranolol), alpha-adrenergic (PHENTOLAMINE and yohimbine), and nitric oxide (NG-monomethyl-L-arginine, L-NMMA) regulation of blood flow. Healthy subjects (body mass index, 18 to 31 kg/m2) were challenged with glucose' (Circulation 2004;109:47-52). The limit: oral nitric-oxide precursors do not automatically deliver this. Citrulline raised NO synthesis measurably yet the same study is titled 'does not increase blood flow' in its population. doi 10.1161/01.cir.0000105681.70455.73

Beta-3 receptor signaling increases it.

PMC1909425 — the answer to the riddle, part two — epinephrine has two arms and they pull opposite ways. 'Vasodilatation in these vascular beds depends on beta-adrenergic signals', while the alpha arm constricts. That is why the human microdialysis study found that natural catecholamines 'increased extracellular glycerol and reduced blood flow' but the pure beta agonist isoproterenol 'induced a greater increase in local blood flow' — and did so most in the femoral depot, the stubborn one. The apparent paradox was never a paradox; it was two receptors on one molecule. ⇒ keep beta, block alpha, support nitric oxide. Carries the same species caveat as the other b3ar edges: in human adipose tissue the lipolytic beta receptors are beta-1 and beta-2. PMC1909425

Blood protein status increases it.

'The supply of free fatty acid (ffa) carrier to the tissue was varied either by variations in the rate of blood flow OR by changes in the albumin concentration of the perfusing blood at constant flow rate. The net production of FFA from the tissue was found to depend on the supply of carrier' (PMID 7195565). ⇒ flow and carrier are two ways of delivering the same thing. PMID 7195565

What it drives

It increases Releases stored fat.

The causal proof that transport, not the enzymes, can be the limiting step. In perfused adipose tissue with lipolysis chemically stimulated, 'The supply of free fatty acid (ffa) carrier to the tissue was varied either by variations in the rate of blood flow or by changes in the albumin concentration of the perfusing blood at constant flow rate. The net production of ffa from the tissue was found to depend on the supply of carrier over a range from 0.1-12 micromoles of albumin x 100 g tissue-1 x min-1... The changes in ffa production appeared to be due to varying degrees of reesterification rather than changes in the rate of lipolysis' (PMID 7195565). The physiological framing: 'Neither TG nor NEFA are soluble to any extent in water, and their transport to and out of the tissue requires specialized transport mechanisms and adequate blood flow.' The perfusion experiment is canine tissue. PMID 7195565

It reduces Fat re-storage.

10.3389/fphys.2022.826314 — The same three-factor sentence identifies carriage as one of the determinants: 'the ability of plasma to carry away released ffa (i.e., blood flow and adequate albumin binding sites)'. And the perfusion experiment isolates it: with lipolysis held constant, raising carrier supply lowered re-esterification and raised net output. 'The findings suggest that the increase in adipose tissue blood flow demonstrated during various lipolytic conditions is of physiological importance by facilitating the removal of FFA from adipose tissue.'. doi 10.3389/fphys.2022.826314
