Re-esterification — Fat Put Back

You want this low, and it is the reason a release product can do everything right and show nothing. Three things decide how much gets put back: how well the cell buffers the free acids internally, whether the blood can carry them away, and whether glucose is around to build the backbone they are re-attached to. The finding that governs the whole category: the rate of putting fat back rises in proportion to the rate of release. Push harder and more comes back. Which is why release alone is not a strategy — it has to be paired with carrying away and burning.

In depth

Released fat put straight back into the droplet — the futile cycle that quietly cancels the work.

What drives it

Releases stored fat increases it.

The sentence that caps this entire product category: 'the absolute rates of reesterification of FFAs are reported to increase proportionally with the rate of lipolysis' (pmc8907745, citing Reshef 1970 and Brooks 1982). Pushing release harder recruits more putting-back. That is why a release lane taken alone has a ceiling built into its own physiology, and why this blend carries a disposal lane and a perfusion lane rather than release alone.

Blood flow to fat tissue reduces it.

10.3389/fphys.2022.826314 — The same three-factor sentence identifies carriage as one of the determinants: 'the ability of plasma to carry away released ffa (i.e., blood flow and adequate albumin binding sites)'. And the perfusion experiment isolates it: with lipolysis held constant, raising carrier supply lowered re-esterification and raised net output. 'The findings suggest that the increase in adipose tissue blood flow demonstrated during various lipolytic conditions is of physiological importance by facilitating the removal of FFA from adipose tissue.'. doi 10.3389/fphys.2022.826314

Insulin sensitivity increases it.

Re-esterification needs a backbone to re-attach the fatty acids to, and that backbone comes from glucose: one of the three named determinants is 'the availability of glucose to produce glycerol 3-phosphate for reesterification' (PMC8907745). Insulin is what drives glucose into the adipocyte, so the fed state supplies both the brake on release and the substrate for undoing it. This edge is drawn on the substrate relationship, not on a single experiment measuring insulin raising re-esterification directly. PMC8907745

Blood protein status reduces it.

The reciprocal test, and it is the strongest evidence on this pathway. 'The inhibition of FFAs on lipolysis is associated with a reduced production of cAMP due to AC inhibition, which can be prevented by reducing intracellular ffa level via extracellular albumin scavenging' (doi 10.3389/fphys.2022.826314). Carry the fatty acids off and the feedback brake never engages. doi 10.3389/fphys.2022.826314

Fat transport protein increases it.

'It is believed that the primary factors determining the extent of reesterification within the adipocyte is the intracellular FFAs buffering via FABPs, the ability of plasma to carry away released FFA, and the availability of glucose' (doi 10.3389/fphys.2022.826314) — one of the three named determinants. doi 10.3389/fphys.2022.826314

Triglyceride assembly increases it.

10.1016/j.molmet.2022.101499 — 10.21748/lipidlibrary.39186 — The named enzyme that performs the return. Same source: dag-acyltransferase takes 1,3-dag straight back to triglyceride, and it prefers the lipase's own product, so the return arm is fed by the release arm (biokhimija.ru/biokhimiya-zhirovoj-tkani/lipoliz.html (русский)). This is why inhibiting that enzyme is a different and arguably better place to stand than pushing release harder: it removes the arm that is recapturing most of what was just released. doi 10.1016/j.molmet.2022.101499

Glycerol backbone increases it.

Fat assembly enzyme increases it.

The first step of putting fat back — named only in the arabic source. The full rebuilding sequence is set out there: 'مسار تخليق الدهون الجلسرين... الجلسرين-3-فوسفات أسيلترانسفيراز (GPAT)... حمض الليسوفوسفاتيديك أسيلترانسفيراز (LPAT)... فوسفاتيز حمض الفوسفاتيديك (PAP)... ديجليسريد أسيلترانسفيراز (DGAT)' = the glycerolipid synthesis pathway runs gpat → lpat → pap → dgat (chemistrysources.com / JoVE العربية — تحلل الدهن (arabic)). This matters because the MAP already had the last enzyme and not the first. This one takes the glycerol-3-phosphate backbone and puts the first fatty acid back onto it — it is the committed step, the point of no return for a fatty acid that was just released. A driver that inhibits the last enzyme stops the final bond. A driver that inhibited this one would stop the whole return. Nothing in this formula reaches it — stated as a limit, not hidden. Glycerol phosphate enzyme increases it.

The enzyme that builds the backbone fat is rebuilt on — and the reason one measurement in this field can mislead. 'GPDH is an important enzyme for lipid metabolism that is central to triglyceride synthesis' and has an 'important role... In converting preadipocytes to adipocytes' (doi 10.3390/molecules191216656 (pmc6270905) — Anti-Obesity Effects of Hispidin and Alpinia zerumbet Bioactives in 3T3-L1 Adipocytes). It makes the three-carbon scaffold onto which released fatty acids are re-attached. Here is why it matters for reading any fat-release number: the standard test measures free glycerol in the fluid around the cell. If this enzyme is blocked, glycerol is not consumed to rebuild fat — so it accumulates. A rise in the measurement can therefore mean more fat coming out, OR less being put back, and the two look identical on the readout. One compound in this formula inhibits it by nearly ninety-eight percent, which is genuinely useful — and means its headline release figure must be read with that in mind rather than at face value. doi 10.3390/molecules191216656

What it drives

It reduces Releases stored fat.

Net output is release minus what goes back, and what goes back is not small. 'It is believed that the primary factors determining the extent of reesterification within the adipocyte is the intracellular FFAs buffering via FABPs, the ability of plasma to carry away released ffa (i.e., blood flow and adequate albumin binding sites), and the availability of glucose to produce glycerol 3-phosphate for reesterification' (pmc8907745). This edge is drawn as inhibitory on net release, not on the hydrolysis reaction itself.
