Heavy metal burden

Blood, urine and tissue metal concentration

The Load You Carry

You want it low: these metals have no biological job, and every one of them does damage while it stays.

In depth

Heavy-metal burden — how much lead, cadmium, mercury or arsenic is actually in the body right now, measured in blood, urine or tissue.

What drives it

Metal-binding protein increases it.

The body's own trap - and it is A TRAP. Confirmed across USA, Swedish and Japanese sources in two languages: MT lowers metal toxicity but raises body burden. Verbatim, doi 10.1093/toxsci/55.1.223 (Klaassen group, MT-null knockout): wild-type hepatic Cd reached '400 ug/g liver' while 'in MT I/II-null mice, hepatic Cd concentrations were about 10 ug/g liver' - 40-fold lower without MT - yet liver injury was worse in the null. Liu & Klaassen: 'the elimination of Cd was much faster in MT-null mice than in control mice'. Nordberg (PMC8945717, independent group): 'These observations support a role of MT in tissue retention and transport of cadmium to the kidney'. Japan Food Safety Commission: Cd binds hepatic MT and accumulates as Cd-MT; Cd-MT is filtered at the glomerulus, reabsorbed in the proximal tubule and accumulates in the renal cortex, with only a very small amount excreted in urine. Tokushima Bunri Univ: when MT is deficient the tissue concentration of zinc and cadmium falls, so MT functions to retain metals in tissue. Why detoxification studies do not answer this: two often-cited papers (PMC262672, PMC5634692) measure detoxification and 'concentration to a physiological or nontoxic level', a toxicity endpoint, not body burden. Different endpoints. Cd-dominant; not generalized to Pb/Hg/As. Counter-evidence: one Nrf2-KO chronic study reports lowered MT with subsequent Cd accumulation, but Nrf2 knockout is confounded across many genes; the direct MT-knockout designs govern. doi 10.1093/toxsci/55.1.223 · PMC8945717

Heavy metal excretion reduces it.

PMC7370001: 'Compared to baseline, post-edetate lead decreased by 36% (p = 0.0004)... Pre-edetate lead decreased by 60% (p = 0.003). Pre-edetate lead excretion became undetectable in nearly 40% of patients. This study suggests that edetate disodium-based infusions may decrease the total body burden of lead.' Converse direction, PMID 7590665: when biliary excretion was blocked to <2% of control, the whole-body methylmercury half-life rose to '46 to 54 days versus 18 to 22 days'. Metal uptake increases it.

PMID 12151624: 'The amount of Cd retained in the body was 10-fold higher in rats fed the FeD diet than in those fed the FeS diet. DMT1 mRNA was highly expressed in duodenum and was 15-fold higher in the FeD diet group' — raising intestinal uptake raised total body burden 10-fold. PMID 15105261: 'diets low in iron (Fe) result in increased absorption of Pb and Cd, suggesting common molecular mechanisms of Cd and Pb transport.'. What it drives

It increases Oxidative stress.

PMC3654245: cadmium, lead and mercury have no essential biochemical roles, but exert diverse, severe toxicities in multiple organ systems as they bind in tissues, create oxidative stress, affect endocrine function, block aquaporins, and interfere with functions of essential cations such as magnesium and zinc. Verbatim from the source for this edge — PMC3654245 Verbatim: “The authors conclude from this and their previous studies and findings reported by other groups that the cadmium-induced expression of mTOR in the CNS observed here is the result of cadmium-increased ROS production and oxidative stress”. doi 10.1155/2018/7612172 · PMC3654245

It increases Calms inflammation.

doi 10.1007/s12291-025-01354-9: heavy metals induce overproduction of ROS and RNS which overwhelm antioxidant defenses, and this disruption of cellular homeostasis 'activates inflammatory pathways, including the NLRP3 inflammasome and redox-sensitive transcription factors like NF-kB, leading to the release of pro-inflammatory cytokines such as TNF-alpha and IL-6.' Corroborated by doi 10.3390/metabo16050319: toxic metals 'disrupt redox homeostasis, impair cellular integrity, and activate inflammatory signaling pathways.'. Verbatim from the source for this edge — doi 10.1007/s12291-025-01354-9. doi 10.1007/s12291-025-01354-9:

It reduces Mitochondrial energy.

PMC12202593: 'we reviewed 20 studies on the mitotoxic effects from exposure to arsenic (n = 6), cadmium (n = 10), and lead (n = 3) as individual toxicants and as mixtures (n = 1)'; the results 'indicate mitochondrial dysfunction characterized by decreased cytochrome c oxidase activity after exposure.'. Verbatim from the source for this edge — PMC12202593. doi 10.1007/s11356-025-36538-6 · PMC12202593
