What is behind Libido & Drive
Serum testosterone — the anabolic signal behind muscle, drive and libido in men. The evidence shows a finding that overturns the common story: suppression under stress is centrally mediated — the brain down-regulating the axis — 'despite intact gonadal capacity'. The organ is rarely the problem. Evidence A — Human trial evidence.
Nitric-oxide blood-vessel relaxation and flow. Evidence A — Human trial evidence.
The chemistry of wanting, not of liking. Dopamine sets how much effort a goal feels worth — which is why its loss reads as apathy long before it reads as sadness. More is not better: the same system that drives pursuit drives compulsion. Evidence A — Human trial evidence.
The hypothalamic–pituitary–adrenal axis and its hormone cortisol — the body's central stress-and-recovery command. Evidence A — Human trial evidence.
Female desire and arousal â the scored clinical construct captured by the Female Sexual Function Index (FSFI): desire, arousal, lubrication, orgasm and satisfaction. Evidence B — Preclinical.
Desire, arousal and function — three separable things that the literature routinely conflates. They are kept apart because an intervention can move one and leave the others untouched. Evidence B — Preclinical.
The few seconds in which the tension built during arousal is released, through rhythmic contractions of the pelvic floor. Those contractions have been recorded instrumentally in both sexes since 1980, and the recordings show real diversity of pattern in women rather than one stereotyped response. It is a separate faculty from desire and from arousal, and the evidence keeps it separate because the evidence does: two independent botanicals moved desire and arousal in trials and left orgasm untouched. Evidence B — Preclinical.
The pituitary hormone that rises sharply after orgasm. Its relationship with dopamine is mechanical rather than mysterious: dopamine holds prolactin down, so when dopamine withdraws, prolactin rises. That makes the post-orgasm surge a readout of what dopamine has just done, not an event in its own right. Evidence A — Human trial evidence.
Desire, arousal and function — three separable parts of sexual health. Evidence A — Human trial evidence.
Evidence on these pathways
Dopamine → Melatonin. “Through α1B-D4 and β1-D4 receptor heteromers dopamine inhibits adrenergic receptor signaling and blocks the synthesis of melatonin induced by adrenergic receptor ligands.” — Circadian-Related Heteromerization of Adrenergic and Dopamine D4 Receptors Modulates Melatonin Synthesis and Release in the Pineal Gland. PLoS Biology, 2012, volume 10, e1001347. doi:10.1371/journal.pbio.1001347
Dopamine → Mood (Positive Affect). “Boosting dopamine also increased happiness resulting from some rewards.” — Dopaminergic Modulation of Decision Making and Subjective Well-Being. Journal of Neuroscience, 2015, 35(27), 9811-9822. doi:10.1523/JNEUROSCI.0702-15.2015 | PMC4495239
HPA Axis / Cortisol → Cognition. “Whereas treatments used in previous studies generally affect more than one memory phase and therefore cannot detect possible differential effects of glucocorticoids on the distinct phases of acquisition, consolidation and retrieval, here we show that treatment of healthy humans with cortisone at acute-stress levels specifically impaired retrieval of declarative long-term memory for a word list.” — Acute cortisone administration impairs retrieval of long-term declarative memory in humans. Nature Neuroscience, 2000, vol 3, pp 313-314. doi:10.1038/73873
HPA Axis / Cortisol → Digestive Function. “Reports have consistently established that central administration of CRF acts in the brain to inhibit gastric emptying while stimulating colonic motor function through modulation of the vagal and sacral parasympathetic outflow in rodents.” — Corticotropin-Releasing Factor and the Brain-Gut Motor Response to Stress. Canadian Journal of Gastroenterology, 1999, vol 13, pp 18A-25A. doi:10.1155/1999/375916

What this page does not claim
This page names the pathways involved and the published evidence on each. It does not claim that any formula treats, cures or prevents Libido & Drive. A pathway opens here only on graded, published evidence, under a standard fixed before the evidence was gathered, and every figure above carries its grade.
