Stress-resistance switch (FOXO)
Forkhead transcription factors

You want it free to work: foxo is the conserved longevity transcription factor (the daf-16 of the lifespan field), activated when insulin/igf-1 signaling drops.
In depth
The forkhead transcription factor held in check by insulin and growth signaling — released when nutrients fall to switch on stress-defense and autophagy genes.
What drives it
Growth & survival signaling reduces it.
Brunet 1999, Cell 96:857 — Akt phosphorylates and inhibits FOXO forkhead factors (14-3-3 nuclear export); when insulin/IGF signaling falls during fasting, FOXO is released. PMID 10102273. Mechanism is exclusion, not suppression: Akt phosphorylates and inactivates the FOXO transcription factors, causing their nuclear exclusion and cytoplasmic degradation. FOXOs normally drive transcription promoting cell-cycle arrest, cell death and oxidative-stress handling, so removing them from the nucleus triggers survival; decreased FOXO activity also blocks transcription of the CDK inhibitors p27 and p21. the tidy 'nuclear Akt phosphorylates nuclear substrates' model is challenged — proteomic studies show most Akt substrates annotated as nuclear actually shuttle between nucleus and cytoplasm. AKT is not one switch — its two activating phosphosites govern different substrates: selectively inhibiting Ser473 weakened FOXO1/2a and FOXO3 but did not affect GSK3-beta or TSC2, while it is THR308 phosphorylation that tracks p-eNOS Ser1177 — Ser473 exerts rare effects on eNOS (PMC8255840). Which Akt site is engaged decides which downstream arm moves. PMID 10102273 · PMC8255840
What it drives
It increases Cellular cleanup.
Mammucari 2007, Cell Metab 6:458 — FoxO3 is necessary and sufficient to induce autophagy in vivo; blocking FoxO3 abolishes fasting-induced autophagosome formation. Direct binding, not correlation: ChIP-seq shows FOXO3 directly binds a network of autophagy target genes in adult neural stem and progenitor cells, and the authors describe FOXO3 as a switch that induces autophagy. Functional consequence and rescue: in the absence of FOXO activity protein aggregates accumulate in those cells, and that accumulation is reversed by TOR inhibition — loss of function, phenotype, and pharmacological rescue in one experiment. The target genes ARE named in muscle too: FOXO3 upregulates LC3, BNIP3, GABARAPL1, VPS34, ULK2 and ATG12 during atrophy. The authors state their own limit, and it matters: a number of autophagy genes in the direct FOXO3 network were not transcriptionally regulated by FOXO3 — binding is not the same as driving, and they suggest additional co-factors are required for full activation. Scope, and it cuts both ways: the same FOXO program that clears aggregates also drives muscle atrophy — FoxO raises MAFbx/atrogin-1, MuRF1, ZNF216, Mul1 and BNIP3, and FoxO3 acts through both the ubiquitin-proteasome and autophagy-lysosome systems. In the heart, FoxO3 induces reversible cardiac atrophy. Autophagy here is a catabolic program, not a purely protective one.
It reduces Muscle.
The catabolic arm, and it is the same pathway as the anabolic one read backwards: IGF-1 binding activates PI3K/Akt, which both promotes protein synthesis and inhibits FOXO-mediated protein degradation. doi 10.3390/ijms26146740 · PMC13074595

Recovery is assembled from the sarcopenia and VO2max literatures; none of the papers cited studied recovery itself. Sources: PMC13074595; doi 10.3390/ijms26146740; doi 10.1139/H07-174. doi 10.3390/ijms26146740 · PMC13074595
