PEPTIDE · WHAT THE EVIDENCE SHOWS

BPC-157

Body Protection Compound 157, a gastric pentadecapeptide. Below is what the published evidence establishes, what pathways it is reached for, and what we carry on those same pathways — with the grade on each.

Where it stands

The evidence position

FDA: “The molecular targets for BPC-157 have not been identified, and the mechanisms of action of BPC-157 are unknown.” (slide 44)

FDA: “FDA found no studies that administered BPC-157 to humans via the proposed oral, SC, nasal, or transdermal ROA.” (slide 56) The single small ulcerative-colitis trial reaches FDA only as a meeting abstract, where “interpretation of the results is limited by the lack of details provided in the meeting abstract.” (slide 49)

FDA: “The nominator did not submit, and FDA did not identify, nonclinical PK and toxicological studies of BPC-157.” (slide 52)

WADA lists BPC-157 under S0, non-approved substances — “this class covers many different substances including but not limited to BPC-157”. Prohibited at all times.

In July 2026 the US Food and Drug Administration reviewed BPC-157 for the 503A Bulk Drug Substances list and proposed that it not be included. FDA briefing document, Pharmacy Compounding Advisory Committee, 23–24 July 2026.

The pathways it is reached for

1VEGF — The Vessel Signal

The angiogenic signal — the instruction to grow new blood vessels.

2Angiogenesis — The New Roads

The building of new blood vessels — restorative when it perfuses healthy tissue, pathological when it feeds disease.

3Endothelial (eNOS) — The Vasodilator

Endothelial nitric-oxide synthase — the vessel-lining enzyme that makes NO to relax and open blood vessels.

4Insulin-R / PI3K-AKT — The Growth Line

Insulin-receptor and PI3K–akt signaling — the anabolic line that clears glucose, supports the vessel wall and commands cell growth.

5JAK — The Cytokine Relay

A cytokine signaling relay. The evidence shows a finding that inverts the intuitive reading: blocking jak in macrophages increased inflammatory output, because it removed an IL-10 feedback brake. 'Calming' this pathway is not straightforwardly calming.

6Egr1 — The Step Between

The step between the growth sensor and the lipase gene.

7Intestinal Barrier — The Inner Gatekeeper

The intestinal barrier — the selective wall that admits nutrients, excludes toxins and anchors the gut–brain and immune dialogue.

8Type-I Procollagen — The Scaffold

Dermal type-I procollagen — the structural protein that gives skin its firmness, elasticity and support.

9Wound Healing — Repair of Broken Tissue

Repair as a scheduled sequence — hemostasis, inflammation, proliferation, remodeling. Skipping or prolonging the inflammatory phase is what produces both non-healing wounds and excessive scarring.

10Skin Structure & Barrier — The Outer Surface

The body's largest organ and its most visible record. Collagen, elastin and barrier lipids degrade on a measurable schedule — and the evidence shows the degradation mechanisms specifically, because that is what the evidence actually witnesses.

Type-I Procollagen → Contractility. “Cardiac fibrogenesis in the late stage of viral myocarditis causing contractile dysfunction and ventricular dilatation, is a major pathogenic factor for the progression of myocarditis to serious cardiovascular diseases including dilated cardiomyopathy (DCM) and congestive heart failure (HF).” — Adoptive Transfer of Regulatory T Cells Protects against Coxsackievirus B3-Induced Cardiac Fibrosis. PLoS ONE; 2013; volume 8; page/elocation not printed on either retrieved record. doi:10.1371/journal.pone.0074955

Type-I Procollagen → Osteoarthritis. “LMCP supplementation significantly reduced WOMAC pain compared to the placebo (−1.90 ± 4.14 vs. 0.61 ± 3.97; p = 0.006).” — Efficacy and safety of low-molecular-weight collagen peptides in knee osteoarthritis: a randomized, double-blind, placebo-controlled trial. Frontiers in Nutrition 2025;12:1644899. doi:10.3389/fnut.2025.1644899

Type-I Procollagen → Muscle Recovery. “12 weeks of 15 g SCP supplementation combined with CT intervention reduced acute markers of exercise-induced muscle damage and improved post-exercise regenerative capacity, as evidenced by the altered post-exercise time course.” — Reduction in systemic muscle stress markers after exercise-induced muscle damage following concurrent training and supplementation with specific collagen peptides – a randomized controlled trial. Frontiers in Nutrition, 2024, 11, 1384112. doi:10.3389/fnut.2024.1384112

Endothelial (eNOS) → Atherosclerosis. “Uncoupling of eNOS occurs in apoE−/− atherosclerosis but does not negate the enzyme's strong protective effects.” — eNOS Protects from Atherosclerosis Despite Relevant Superoxide Production by the Enzyme in apoE−/− Mice. PLoS ONE, 2012, volume 7, e30193. doi:10.1371/journal.pone.0030193

Endothelial (eNOS) → Renal Function. “Endothelial nitric oxide synthase (eNOS) deficiency exacerbates proteinuria and renal injury in several glomerular diseases, but the underlying mechanism is not fully understood.” — Endothelial Nitric Oxide Synthase Prevents Heparanase Induction and the Development of Proteinuria. PLoS ONE, 2016 (9 August), vol 11, e0160894. doi:10.1371/journal.pone.0160894

Intestinal Barrier → Ammonia. “This weakens the paracellular seal of the intestinal epithelium, enabling microbial products such as endotoxins, ammonia, and even whole microorganisms to translocate into the portal circulation.” — How the gut-liver axis shapes hepatic encephalopathy: mechanistic and therapeutic perspectives. Exploration of Digestive Diseases 2025;4:100597. doi:10.37349/edd.2025.100597 (no PMID exists — the journal is not MEDLINE-indexed; absence confirmed on file, not derived)

What a grade means here

Every ingredient row on our formula pages carries one of these grades, fixed by a standard set before the evidence was gathered. The grades below are reproduced from those pages.

AHuman trial evidence

Randomized human trials, or a meta-analysis of them, measuring this pathway's own endpoint — either a clinical outcome or a validated biomarker.

B+Corroborated

Human data, or animal evidence replicated by an independent laboratory. It opens a pathway only alongside an independent corroborating driver.

BPreclinical

Mechanistic, cell or animal evidence. No single preclinical driver opens a pathway on its own.

What this page does not claim

Sharing a pathway is not the same as sharing an effect. Nothing here says a botanical formula does what BPC-157 does, and nothing here recommends BPC-157. The pathways above are named because that is what the published work on BPC-157 points at; the formulas below are listed because our own map records graded, published evidence on those same pathways. Read both columns and judge them yourself — every figure on our side carries its grade and its source.
