Insulin/nutrient-driven mTORC1 signaling

The Second Off-Switch

The balance is state, not amount. Erasing the signal only silences the enzymes you already have; this route reduces how much of the rate-limiting lipase gets built in the first place. That is a slower, deeper suppression, and it is why a single fasted window does not undo a chronically fed state. It is not a villain: this same sensor governs growth and repair throughout the body, and it is also one of the two kinases that open the lysosomal route to fat breakdown. Blocking it wholesale is not a fat-loss strategy.

In depth

Insulin's other route — it stops the lipase being made, not just being switched on.

What drives it

Insulin sensitivity increases it.

'The binding of insulin to its receptor, irs-1, leads to the activation of both PI3K/Akt and mTORC1 pathways, which result in the inhibition of lipolysis, either through the decrease of cAMP levels by PDE-3B (PI3K pathway) OR by inhibiting the transcription of ATGL' (doi 10.3389/fphys.2022.826314, Figure 2 legend). Two arms from one hormone. doi 10.3389/fphys.2022.826314

What it drives

It reduces Fat-releasing enzyme (ATGL).

Insulin's second route — it stops the lipase being made. 'insulin may inhibit lipolysis by decreasing transcription of ATGL via the mTORC1-mediated pathway (mTORC1-Egr1-ATGL) or through FoxO1' (doi 10.3389/fphys.2022.826314). The source is candid about the whole area: 'the detailed mechanisms remain controversial and incomplete.' and it is not a villain: the same kinase is one of the two that induce the lysosomal route to fat breakdown. doi 10.3389/fphys.2022.826314

It increases Early growth response.

The route is named as a chain: 'the mTORC1-mediated pathway (mTORC1-Egr1-ATGL)' (doi 10.3389/fphys.2022.826314). This edge draws the middle step so the map does not jump it. doi 10.3389/fphys.2022.826314

It reduces Fat droplet autophagy.

The same kinase, the opposite sign, on a different route. 'Under conditions of nutrient deprivation, induced by the major metabolic kinases mTORC1 and AMPK, lipid droplets and autophagic machinery associate to form autophagic vacuoles' (doi 10.3389/fphys.2022.826314) — nutrient deprivation relieves mTORC1 and opens lipophagy, while nutrient abundance suppresses it. Drawn inhibitory because the active, fed-state kinase is what holds this route shut. doi 10.3389/fphys.2022.826314
